I reviewed the manuscript# seeds-1963573 entitled In vitro propagation of Garcinia livingstonei T. Anderson (African mangosteen) a woody tree species through the culture of matured seed segments”. The experiments described in the paper are sound and suitable for publication as a short communication in this journal after revision. There are some major queries/corrections which need to be addressed/corrected in the revised version.

1.     Line 18: After how many days/weeks the shoots were achieved….please mention.

2.     Line 20: periodic subculturing of the shoot clumps….. 

3.     The introduction is too short….please add shor para about in vitro culture and their role in conservation.

4.     Line 56-67: How many washes with sterile water?

5.     Line 58: What was the pH of the media? This is also a critical factor for shoot induction in vitro.

6.     Line 64: from the regeneration media…..

7.     Line 108-109: What happen with full MS containing auxin? Have you tested?

8.     Line 140, table 2 caption: auxin supplemented to 1/2 WP medium…..

9.     Line 167: supplemented….

10.  Lin 168: IBA proved better in inducing maximum number of roots with highest average root length.

11.  Line 171: effective for root induction…..

With reference to the above, the authors are thankful to the anonymous reviewer for their valuable suggestions on the manuscript. We have incorporated all the suggestions in the revised manuscript and the following are specific changes incorporated in the revised manuscript.

  1. Line 18: After how many days/weeks the shoots were achieved … please mention.

Answer: Shoot buds were initiated after 4-weeks of culture, shoot elongation was achieved in another 4 weeks of subculture, and complete shoot elongation was achieved in another 4-weeks after subculture. Thus, complete shoot development was achieved after 12 weeks of seed culture, this information is included in the revised manuscript.

  1. Line 20: periodic subculturing of shoot clumps

Answer: The suggestion is incorporated in the revised manuscript.

  1. The introduction is too short… please add short para about in vitro culture and their role in conservation.

Answer: The suggestion is incorporated in the revised manuscript.

  1. Line 56-67: How many washes with sterile water?

Answer: Three washes and this suggestion is incorporated in the revised manuscript.

  1. Line 58: What was the pH of the media? This is also a critical factor for shoot induction in vitro.

Answer: The pH was set at 5.8 and this suggestion is incorporated in the revised manuscript.

  1. Line 108-109: What happens with full MS containing auxin? Have you tested?

Answer: On full-strength medium containing auxin does not trigger the root induction from the shoot and this might be due to the high salt concentration of the medium.

Table 1 and 2 - please provide the number of explants analysed in each experiment.

Answer: The fruits which are ripened (turned yellowish-red) will be having matured seeds. This information is incorporated in the revised manuscript.

  1. Line 53 – what does the word ‘decorated’ means in this text?

Answer: It should be ‘de-coated’ and this information is incorporated in the revised manuscript.

  1. Line 54 – which liquid detergent was used?

Answer: Triton X 100 was used as a detergent and this information is incorporated in the revised manuscript.

  1. Lien 58 – please present the scheme, describing, how exactly the seed was cut into four pieces.

Answer: The seeds were cut into four pieces by cutting once vertically and then horizontally (Scheme 1). Scheme is provided in the revised manuscript.

  1. Line 88 – probably, it is better to use the word ‘demonstrated’ instead of ‘involved’
