Round 1

Reviewer 1 Report

In this manuscript, a study was conducted to learn more about the HSP70 genes (HSP70 genes family), or more specifically, how candidate genes of ClHSP70 were selected and analyzed its expression under high temperature in C. lavandulifolium. In my opinion, these data should be published, though some very minor editing of the submitted version would be necessary before the manuscript should be granted full acceptance by Horticulture, MDPI Journal.

Line 178: add a reference and correct to make “delta” superscript/subscript according to reference’s format.

Line 187: what is “pls? write full name first and abbreviation next.

Line 342: Are Figures 5A and 6 are interrelated? However, 5A is not related, make a correction.

Line 347: 5B has no relation to this, make the necessary correction.

Line 365: P probability values should be in italic.

Line 434: Is there a list of ten candidates genes that were selected? Supplemental able was not available….Because earlier in methodology you mention it was 15 candidate gene for tissue expression of C. lavandulifolium. Need explanation.

The authors of this research paper presented a well structured report regarding the HSP70 gene family in which the introduction satisfactorily states the intention of the study and gives a good overview of the previous research on the subject and presents the most interesting results that they obtain. The results section is well articulated, it follows a logic organization; the figures are complete and well designed which make the reading very straightforward.

However, the section that I found completely unsatisfactory is the discussion section. On that section the authors basically repeat the results that they obtained but the interpretation of them is not appropriate enough. The authors have a big opportunity to propose some candidate genes that can be used in genetic engineering programs to improve Chrysanthemum lavandulifolium heat stress tolerance. Additionally, the authors need to perform a full evaluation of other cases in which HSP70 gene family members have been reported that expression patterns changes in response to heat stress. Also, there are several reports of HSP70 gene family characterization in different species, it would be a good exercise to compare those data with the data that the authors obtained.

Below I enumerate some imprecisions that I found on the text:

Line20- Change “the genes responded significantly“ for “gene expression change significantly“ or similar.

Line 62 - The first time that you use an abbreviation (HSC70) it should be explained.

Line 75 - Which is the effect of the mutation on the resulting protein? It is important to understand the impact on the phenotype.

Line 89-92 - This part needs to be rephrased since it is not clear.

Line 184 - Where is Table S1? I believe it is not available.

Line 342 - Change Fig 5A for Fig 6A.

Line 372 - Change the phrase “Anniversary 372 production of flowers is an urgent requirement for chrysanthemum production.“ because it is not understandable.

Line 381-405 - This is a repetition of the results, should be abbreviated and relate it more with the findings of similar studies.

The authors have satisfactorily answered my comments and the discussion in its actual form it's good enough to be published.
