Reviewer 2 Report

This manuscript by Liu et al., describes the genetic diversity of tea plant germplasm resources in Wuyi mountain based on SNP marker. This  manuscript is well  written, however more information are lacking specially in the materials and method section. Author should describe about how they prepared samples for DNA extraction, how much leaf amount they used, chemicals with amounts should provide along with the DNA extraction kit name.  Also, author should provide the primers informations, length, PCR conditions and DNA sequence analysis procedure. I did not found any informations; how performed AMOVA analysis, how they calculate Fst value, how they construct hierarchical clustering tree?

In the results section, authors should provide the major polymorphic sites showing how they are different from each others in one table in manuscript and should provide complete informations  of all polymorphic sites along with genbank accession numbers in supplementary file.

Minor suggestions:

Line 82-85: Write the full name with abbreviations while they mention first time and use abbreviations later.

Line 84-85: Remove

Line 91-92: ....Which requires a DNA concentration of ......ratio of 1.7-1.9, remove it.

Table 1. List of 137 tea samples or tea germplasms (WY1-137)? because the meaning of samples and germplasm is totally different.

Please, reformat the table, so it will be easy to readers to differentiate the samples.

Mention the full form of abbreviations from table in the footnote.

Line 267-269: How this results suggest that genetic distance and genetic differentiations among the samples increase with the increasing the geographical distance? Please describe this results very carefully.

Figure 5, Please indicate the  samples locations in the figure by using different sign, so it will be more easy to see the genetic and geographic variation.

Round 2

Reviewer 2 Report

I found substantial improvement in the revised manuscript.

Please follow my minor suggestions as follows:

1. Mention the version of all the analysis softwares. like Mega 6, 7, 10  , or what?
