The research is not enough to being published, and the content lacks innovation.

1. Aspergillus sp. D-23 is a mutation from D-18but we don't know what happens to increase the activity of α-galactosidase, whether it induces a new α-galactosidase or a mutation of the original α-galactosidaseThe enzyme activity of D-23 was higher than D-18, but the differences of thermal and pH stability between them were not exhibited.

2. FIG. 2 The bands of purified proteins are blurred.

3. The content of the experiment was repetitive, the data of HPLC is sufficient to determine hydrolysis capacitythe results of Fig 5 and Fig 6 are meaningless. Besides, the method and calculation of the reducing sugar content in Table2 and Fig 6 was not mentioned in the part of Method.

Review of MS titled: “High hydrolysis activity of α-galactosidase from 2 the novel strain Aspergillus sp. D-23

Authors show a complete scientific approach to the study of α-galactosidase purified from Aspergillus sp. by ultraviolet-diethyl sulfate (UV-DES) compound mutagenesis. It exhibited maximal activity at 65 and pH 5.0 that showed broad high optimal temperature and pH stability range. Scientific approach is good and so the paper can be published in Processess journal after minor revisions.

The following substantive errors and editorial defects, mainly of the descriptive type, should be noted:

1.      Title:

Improve the title of the manuscript.

2.      Abstract:

Too short for an article. I would recommend adding salient depiction of results to the abstract as well, that have been described well in main text.

3.      Introduction:

4.      Introduction is appropriate, but there are some basic English grammatic errors. I would recommend having it proofread by a native English speaker.

5.      Figure 2. is well placed, serves the purpose. But enhance its quality please by replacing it.

6.      Check in whole text, the organism name should be written in italic.

7.      In section 3.2 purification of galactosidase, what do you mean by 4.5 fold. Please explain.

8.      Too many abbreviations have been used in whole manuscript. Write full forms at least once when using the first time in text.

9.      Replace Figure 5 with better resolution.

10.  References are not carefully prepared, in accordance with the citation rules in force in this Journal.

 

 

Overall, the article is well written, and in my opinion fit to be published after revisions.

It was difficult to understand this end of the conclusion, it seems that these two sentences do not match:

Thin layer chromatography and high-performance liquid chromatography, the enzyme had good hydrolysis ability and could efficiently hydrolyze melibiose and raffinose at 50 °C within 2 hours. They were practically totally hydrolyzed after 4 hours of treatment.”
