Round 1

Reviewer 1 Report

In this manuscript the authors reported that H. cordata extracts (HCA and HCE) obtained from aqueous and ethanol extraction, respectively provided anti-C. acnes, antioxidant, and anti-aging activity against collagenase, elastase, and hyaluronidase along with cytotoxicity and LC-MS/MS study. I would like to appreciate their effort. however there are some observations:

1. All abbreviations used should be elaborated if come for the first time in the text.

2. In section 2.3, 5% NaNO2 should be NaNO2 as same for the others as available.

3. Though some antioxidant assays but for more exploration of antioxidant and scavenging activity need to do some other assays please follow

Antioxidants 2020, 9(4), 282; https://doi.org/10.3390/antiox9040282 For section 2.12 please include microscopic images of cells after treatment as a proof of the findings For section 2.13. please exclude hame of expert for LC-MS/MS, rather acknowledge him in the acknowledgement sestion

Reviewer 2 Report

This manuscript entitled " Anti-Aging, Anti-Acne and Cytotoxic Activities of Houttuynia cordata Extracts and Phytochemicals Analysis by LC-MS/MS " studied the effect of Houttuynia cordata plant as an antiageing and anti-acne by two extraction fractions ethanolic (HCE) and Aquos (HCA).

The authors used relevant references in the research point, which added a distinctive value to the content of the manuscript and the state of the art of the scientific discipline in this field. The manuscript included 57 references, including modern 21 research published during the last five years with only one reference during this year 2022.

The methods used in this study covered the most relevant assays for antioxidants (Total Phenolic Content, Total Flavonoid Content, DPPH, ABTS, and Ferric Reducing Antioxidant Power). Furthermore, for enzymes as antiaging and antiacne properties, they measure the collagenase inhibitory, elastase inhibitory, and hyaluronidase inhibitory activities. Also, for the antimicrobial activity assay,  Cutibacterium acnes DMST 14916 was used as the standard organism. Cytotoxic activities on mouse macrophage (RAW 264.7 cell line) and human skin cells (HaCaT and NHDF cell lines) were also performed and phytochemical compounds in both HCE and HCA extracts were determined by LC-MS/MS.

The introduction reviewed the background and importance of Houttuynia cordata plant. It also listed the damage of algae to the environment of coral reefs and their effects on growth.

Materials and methods mentioned the sampling and detailed steps of the experiment protocols.

The results were monitored in a table and presented in four tables and two figures in a sound scientific manner, explaining the differences between the two different extracts.

My final decision is to accept this manuscript for publishing as it is in the present form without any corrections, edits or modifications
