The mutation identified has lead toa changed amino acid, if comment would have explained any tertiary or quarternary structure, it would have been very appropriate

Thanks for your comments.

Unfortunately, it is not possible for us to study the effect of the amino acid change on the   structure of the protein. Moreover, We do not access the case for resampling.

I have some comments that I think can improve the manuscript:

1)In table 1 add hemoglobin capillary electrophoresis indices (Hb A2%, Hb F%, Hb Mazandaran%).

2) Hb Mazandaran is a new Hb. Three different variants of Hb have been identified at codon 52, but amino acid change is different. Utilizing a program (for example PolyPhen-2 http://genetics.bwh.harvard.edu/pph2/), the authors can predict the potential effects of amino acid substitution on protein function.
