This manuscript investigates IL-12 family receptor expression in CLL intraclonal subfractions (PF and RF) and their modulation by antigen-independent stimulation, providing valuable insights into IL-23/IL-23R axis relevance in CLL pathogenesis. The study design integrates flow cytometry, RT-qPCR, and in vitro stimulation, with clear phenotypic characterization of cell fractions.

Line 61-64, Intermediate fractions (IF) reflect transitional stages in this trafficking cycle, which is also identifizated in flow cytometry (Section 4.5). However, I can’t find any IF results in the main text. Please confirm.

Besides, all the figures were not clear, please change them to a clearer version.

The document is well-structured; however, it does not sufficiently emphasize the importance of these receptors. The current focus is primarily on describing results rather than highlighting the significance of inhibiting or expressing these receptors and the biological mechanisms to which they are related. Additionally, the results section should clearly specify the source of the samples rather than deferring this information exclusively to the methodology section. Additional recommendations:

-Improve the quality and resolution of all figures

-Provide a comprehensive conclusion section

The scientific article ‘Intraclonal Enrichment of IL-23 Receptor Complex Expression in the Proliferative Fraction of Chronic Lymphocytic Leukaemia’ addresses the issue of expression and inducibility of IL-12 family receptor subunits on CLL cells. It is written in correct language. The introduction to the article is well written, in understandable language, and provides a good introduction to the topic of CCL, cytokines and their receptors.  The results of the study are presented in a way that is understandable to the reader. The methodology of the study is discussed correctly. However, I have a few comments on the content and structure of the article:

  1. Structure of the article:

1.1. I think it would be better to place the materials and methods section before the results section, as this would make the entire article easier to understand.

1.2  There is no section clearly defining the aim of the study.

1.3 There is no conclusions section at the end of the article clearly summarising the conclusions of the study.

  1. Figures:

2.1 I do not think it is a good idea to have a separate chapter for figures – it would be better to place them in the text directly below where they are referred to.

2.2 The captions for the figures do not explain the abbreviations used. A list of abbreviations should be provided under each figure.

2.3 Some of the data contained in the figures is illegible – I would recommend attaching them in a higher resolution/larger magnification.

  1. It would be a good idea to include an illustration in the Materials and Methods chapter summarising the successive stages of the study.
  2. There is no list of abbreviations with explanations at the end of the article.
  3. Line 28 – I am not sure if the letters ï are used correctly.

Once the above suggestions have been implemented, the article will be ready for publication.
