This manuscript entitled "Inherited genetic mutations and polymorphisms in malignant mesothelioma: a comprehensive review" is indeed a thorough review on the subject. The authors are well-known experts in the mesothelioma field and I commend them for their hard work in summarising a huge amount of literature into a very educational review. It is a well-written manuscript, with a good layout and easy to follow.

The review is well organized and easy to read. Moreover it is a complete revision of the literature if the shown inclusion criteria are considered.

However I suggest the authors to reinforce in conclusion section the message given in the abstract regarding the importance of a personalized investigation in high-risk individuals for a better target therapy.
