Complete chloroplast genome sequences are widely used in the analyses of phylogenetic relationships among angiosperms. As a species-rich genus, species diversity centers of Saxifraga L. include mountainous regions of Eurasia, such as the Alps and the Qinghai–Tibetan Plateau (QTP) sensu lato. However, to date, datasets of chloroplast genomes of Saxifraga have been concentrated on the QTP species; those from European Alps are largely unavailable, which hinders comprehensively comparative and evolutionary analyses of chloroplast genomes in this genus. Here, complete chloroplast genomes of 19 Saxifraga species were de novo sequenced, assembled and annotated, and of these 15 species from Alps were reported for the first time. Subsequent comparative analysis and phylogenetic reconstruction were also conducted. Chloroplast genome length of the 19 Saxifraga species range from 149,217 bp to 152,282 bp with a typical quadripartite structure. All individual chloroplast genome included in this study contains 113 unique genes, including 79 protein-coding genes, four rRNAs and 30 tRNAs. The IR boundaries keep relatively conserved with minor expansion in S. consanguinea. mVISTA analysis and identification of polymorphic loci for molecular markers shows that six intergenic regions (ndhC-trnV, psbE-petL, rpl32-trnL, rps16-trnQ, trnF-ndhJ, trnS-trnG) can be selected as the potential DNA barcodes. A total of 1204 SSRs, 433 tandem repeats and 534 Large sequence repeats were identified in the 19 Saxifraga chloroplast genomes. The codon usage analysis revealed that Saxifraga chloroplast genome codon prefers to end in A/T. Phylogenetic reconstruction of 33 species (31 Saxifraga species included) based on 75 common protein coding genes received high bootstrap support values for nearly all identified nodes, and revealed a tree topology similar to previous studies. Lyme borreliosis (LB), caused by Borrelia burgdorferi sensu lato (s.l.) and babesiosis, caused by Babesia divergens and Babesia venatorum, are both transmitted by the ixodid tick Ixodes ricinus. Although these diseases coexist in Spain and other European regions, no studies have been carried out to assess the impact on the health of patients exposed to both causative pathogens. This retrospective study, conducted in Asturias in northwestern Spain between 2015 and 2017, analyzed the possible complications arising from exposure to these pathogens. To this end, the epidemiological, clinical and analytical features of two groups of a cohort of 120 LB patients exposed to one or two of the pathogens were compared. The cohort comprised 73 patients who had only been infected with Bo. burgdorferi s.l. (Bb group) and 47 patients who were seropositive for Ba. divergens/venatorum in addition to being infected with Bo. burgdorferi s.l. (BbBdv group). The results showed that LB patients who had also been exposed to infection with Babesia spp. (BbBdv group) had significantly more cardiorespiratory symptoms, in particular dyspnea and first- and second-degree atrioventricular (AV) blocks, compared to those who had been infected with Bo. burgdorferi s.l. alone (Bb group). No relevant differences in other symptoms, epidemiological factors or analytical tests were observed between the two groups. Full article

Ticks carry numerous pathogens that, if transmitted, can cause disease in humans and animals. Research on pathogens transmitted from ticks to humans is essential for improving public health strategies against tick-borne diseases (TBDs). In this study, Ixodes ricinus ticks found on humans were collected and examined between 2019 and 2024. This study is about the molecular characteristics of tick-borne pathogens (TBPs) in the region of northern Serbia, including Borrelia burgdorferi sensu lato (s.l.), Anaplasma phagocytophilum, and Coxiella burnetii. We identified B. burgdorferi s.l. as the most prevalent in ticks (20.45%). Molecular analysis identified two genospecies, B. afzelii and B. burgdorferi s.s., indicating genetic diversity among Borrelia spirochetes. A. phagocytophilum was detected in ticks with a prevalence of (1.62%), while C. burnetii was not found in any of the ticks. Our findings highlight the necessity of monitoring tick pathogens in ticks removed from humans. Serbia is an endemic region for some tick-borne diseases, such as Lyme disease. Regular surveillance of tick populations, with molecular identification of pathogens, offers insight into transmission dynamics, allowing for monitoring and public health interventions to be created if needed due to increased risk. This study sought to investigate the presence of anti-Rickettsia spp. antibodies in georeferenced serum samples from equids across all regions of the state of Goiás, while also presenting variables that indicate risk factors for the circulation of rickettsiae, and evaluating the presence of rickettsial DNA in ticks collected from equids and the surrounding environment in the municipalities of Uruaçu and Porangatu, located in the northern region of the state. A total of 1156 equid serum samples provided by the Goiás Agricultural Defense Agency (Agrodefesa) were analyzed for antibodies against 4 Rickettsia antigens. Additionally, 39 blood samples were collected from equids during a 3-day field expedition in January 2024, alongside 938 ticks collected from both animals and the environment. In total, 297 adult ticks were randomly selected for molecular analysis and tested by qPCR targeting the rickettsial gltA gene. Positive samples were further analyzed using cPCR to target the ompA and gltA genes. Results revealed that 9.6% (111/1156) of the serum samples were seroreactive to at least one Rickettsia antigen. Among these, 36% had antibodies against Rickettsia rickettsii, 18.9% against Rickettsia parkeri, 69.4% against Rickettsia amblyommatis, and 14.4% against Rickettsia bellii. Furthermore, the possible antigen responsible for a homologous reaction was found in 3.6% of equids for R. rickettsii, and in the same proportion for R. bellii, while 23.4% of animals showed antibodies for R. amblyommatis, and none exhibited a homologous reaction to R. parkeri. Meanwhile, 28.2% of the serum samples collected during the field expedition had antibodies against R. amblyommatis, with 72.7% identifying R. amblyommatis as the antigen involved in the homologous reaction. In the logistic regression analysis, the variables of education level, altitude below 500 m, and presence of female mules demonstrated a positive effect on seropositivity. Ticks from animals were identified as Amblyomma cajennense sensu lato, Amblyomma sculptum, Dermacentor nitens, and Rhipicephalus microplus, while environmental ticks were identified as A. cajennense s.l., A. sculptum, and Amblyomma nodosum. In the qPCR assays, two specimens of A. cajennense s.l., one of A. sculptum, and one of D. nitens amplified a fragment of the gltA gene. Of these, one A. cajennense s.l. specimen amplified a fragment of both the ompA and gltA genes, and one A. sculptum specimen amplified a fragment of the gltA gene through conventional PCR. Sequencing confirmed the detection of R. amblyommatis. These findings highlight the presence of anti-Rickettsia spp. antibodies in equid serum samples from all regions of the state of Goiás, emphasizing the important role of equids as sentinels for Rickettsia spp. To our knowledge, this study represents the largest effort to detect anti-Rickettsia spp. antibodies in equid serum samples in Brazil. Additionally, it is the first nationwide investigation of its kind conducted in collaboration with the Agricultural Defense Agency (Agrodefesa), serving as a significant example of the One Health approach. This study presents a thorough revision of the Malvaceae s.l. (sensu lato) family within the Flora of the Toor Al-Baha District, Lahij Governorate, South Yemen. The documented taxa were subjected to morphological revision alongside an analysis of life forms and chorological affinities. Thirty taxa are categorized into 12 genera within four subfamilies. Each species is presented with its current classification, accepted names, synonyms (when applicable), vernacular names (when applicable), photographs, and distribution to aid in identifying and recognizing these species. The documented mallow species comprises 80% perennials and 20% annuals. Chamaephytes constituted the predominant life forms, comprising 50% of the total taxa surveyed. A phytogeographical analysis revealed that chorotypes with Sudano-Zambezian affinities are predominant, comprising 83.34% of the recorded mallows. The two subfamilies, Grewioideae and Sterculioideae, are distinguished by the lack of an epicalyx and the presence of an androgynophore, unlike the subfamilies Dombeyoideae and Malvoideae. The highest richness genera included Grewia, with seven taxa, and Hibiscus, Abutilon, and Corchorus, each comprising four species. Ixodes ricinus ticks play a crucial role as carriers of diseases, transmitting pathogens to vertebrate hosts, including horses. This study aimed to investigate the seroprevalence of IgG antibodies against Borrelia burgdorferi sensu lato (s. l.), Anaplasma phagocytophilum, and tick-borne encephalitis virus (TBE-virus) in equine sera collected in southern Norway. In total, sera from 331 horses stabled in four counties (Agder, Vestfold and Telemark, Vestland, and Viken) were analyzed by immunoblot. In total, 66% of the horses were IgG-seropositive for antibodies against one or multiple tick-borne pathogens. The highest seroprevalence was detected against B. burgdorferi s. l. (47%), followed by A. phagocytophilum (34%) and TBE-virus (10%). A significant difference between the counties regarding IgG antibodies against A. phagocytophilum was found, with the highest seroprevalence in horses stabled in the coastal areas of Agder and Vestland. In conclusion, the study demonstrates that horses in southern Norway are at high risk of contracting tick-borne infections. Full article

The spectrum of ixodid ticks that bite humans in Western Siberia has significantly changed over the past two decades. In this study, we determined tick species attacking people in the vicinity of Novosibirsk and the range of bacterial agents they were infected with. This study included 301 ticks taken from people and 46% were Ixodes pavlovskyi, followed by Ixodes persulcatus (19.6%), I. persulcatus/I. pavlovskyi interspecies hybrids (19.6%), Dermacentor reticulatus (12.8%), and single Dermacentor marginatus and Dermacentor nuttalli/Dermacentor silvarum. Human DNA was determined in ticks, first demonstrating that all Ixodes spp., including hybrids, can effectively feed on humans. The DNA of Borrelia spp., Rickettsia spp., and Anaplasmataceae bacteria was detected in different tick species. Borrelia garinii prevailed in Ixodes species, being found in 8.8% of ticks, whereas B. afzelii and B. bavariensis were found in single ticks. Borrelia miyamotoi was revealed in 3.7% of ticks. “Candidatus Rickettsia tarasevichiae” and R. raoultii were identified mainly in I. persulcatus and D. reticulatus (44.8% and 26.3%, respectively), while Rickettsia helvetica was found only in 2.2% I. pavlovskyi. The prevalence of Anaplasma phagocytophilum, Ehrlichia muris, and Neoehrlichia mikurensis did not exceed 2%. The obtained results indicate a high risk for humans to be infected with agents of Lyme borreliosis, primarily B. garinii. Full article

Bacillus cereus sensu lato (B. cereus s.l.) are significant spoilage and pathogenic microorganisms found in various foodstuffs. They are responsible for defects like sweet curdling in milk, which impacts dairy product storage and distribution. Nevertheless, the genetic mechanisms underlying B. cereus-induced sweet curdling remain poorly characterized. In this study, we investigated the genetic and functional basis underlying this phenomenon through whole genome sequencing of the newly isolated B. cereus strain BC46 and transcriptome sequencing at two phases of its growth in milk. Hybrid assembly of Illumina and Nanopore reads resulted in a 5.6 Mb genome with 35.1% GC content, classifying BC46 as B. cereus sensu stricto (B. cereus s.s.) within the panC group IV. Several virulence factors, antimicrobial resistance genes, and cold shock proteins were identified in the genome. A distinct functional profile of BC46 was observed before and after the development of sweet curdling in milk. Genes associated with sporulation, toxin production, hydrolysis, and proteolysis were upregulated in sweet-curdled samples. Our findings highlight potential gene targets that may play an important role in the BC46-induced sweet curdling in milk, enhancing our understanding of its molecular basis and supporting the development of new genetic approaches for early spoilage detection. The olive tree is one of the most important fruit crops grown in Morocco, yet extensive decline associated with the root rot of this crop has been observed in many regions. This study aimed to identify and characterize the oomycetes associated with root rot disease in olive trees. During the 2021 and 2022 growing seasons, symptomatic root tissues and soil samples were collected for isolation. Based on morphological traits and the sequencing of the internal transcribed spacer (ITS) region of rDNA, 10 oomycete species were identified, belonging to the Phytophthora and Pythium sensu lato (s.l.) genera. Seven species were assigned to Phytophthora, namely, P. palmivora, P. plurivora, P. acerina, P. oleae, P. cactorum, P. gonapodyides, and P. megasperma. The Pythium s.l. genus was represented by three species, including P. schmitthenneri, P. aphanidermatum, and P. irregulare. A pathogenicity assay was conducted by soil infestation to evaluate the effect of these pathogens on one-year-old olive saplings (var. Picholine Marocaine). Results revealed that all 10 species were pathogenic to olive saplings. Inoculated saplings exhibited symptoms, such as root rot, vascular discoloration, and wilting. The pathogens were successfully re-isolated from necrotic roots, thereby fulfilling Koch’s postulates. These findings highlight the complex etiology of root rot disease in olive trees, as multiple species can induce similar symptoms. This study represents the first detailed report of Phytophthora and Pythium s.l. species associated with olive root rot disease in Morocco. Full article

Throughout Europe, including Poland, Ixodes ricinus ticks are the main vector of numerous pathogenic agents that pose a serious threat to public health. Southern Poland attracts many tourists with its scenic landscapes and abundant recreational opportunities. These areas are ideal habitats for wild fauna, which serve as the main reservoirs and hosts for these pathogens and ticks. The large population and biodiversity of these hosts facilitate the proliferation of ticks. The aim of this study was to determine the potential exposure of humans to ticks and tick-borne pathogens such as Borrelia burgdorferi sensu lato, Anaplasma phagocytophilum, and Babesia spp., along the nature-educational and tourist trails of the Poprad Landscape Park. From 2020 to 2021, ticks were collected using the flagging method on three tourist trails and nature-educational paths within the Poprad Landscape Park. DNA was isolated from 213 I. ricinus ticks using the ammonia method. To detect pathogens in ticks, PCR and nested PCR methods were used. To detect B. burgdorferi s.l. and A. phagocytophilum, two pairs of primers specific to the flaB gene fragment and 16S rRNA gene fragment were used, respectively. For Babesia spp. detection, primers specific to the 18S rRNA gene were used. The amplification products were separated electrophoretically and visualized under ultraviolet light. In total, among the 213 examined ticks, B. burgdorferi s.l. was detected in 31% of the samples. Neither A. phagocytophilum nor Babesia spp. were detected in the studied material. These results indicate a potentially high risk of ticks and tick-borne B. burgdorferi s.l. infections for residents and tourists in the recreational areas of the Poprad Landscape Park. Full article

The epidemiological situation related to infectious diseases is influenced by many factors. To monitor actual trends in selected zoonoses, a total of 473 serum samples from farmers, forestry workers, and veterinarians were collected for serological examination. Anti-Borrelia burgdorferi sensu lato (s.l.) antibodies were tested with ELISA and Western blot (WB) tests; the detection of anti-Toxoplasma gondii antibodies was performed using an enzyme linked fluorescence assay (ELFA). Antibodies to bartonellosis, anaplasmosis, and chlamydiosis were determined by indirect immunofluorescent test (IFA), whereas antibodies to yersiniosis and mycoplasmosis were confirmed in the ELISA test. Positive or borderline results of antibodies against B. burgdorferi s.l. in the ELISA test were detected in 33.8% of the study population. The borderline or positive ELISA test results for at least one antibody class were confirmed by WB in 58.7% of cases. The IgG antibodies against Anaplasma phagocytophilum, Toxoplasma gondii, and Mycoplasma pneumoniae were detected in 9.6%, 51.7%, and 63.6% of samples, respectively. Antibodies against Yersinia spp., Bartonella henselae, and Chlamydia pneumoniae were found to vary between 43 and 47%. Full article

This study investigates the diversity and distribution of intertidal Cystoseira sensu lato (s.l.) species across different protection zones within the “Capo Gallo-Isola delle Femmine” Marine Protected Area (MPA) in the central Mediterranean Sea. Five Cystoseira s.l. species (Cystoseira compressa, C. foeniculacea, Ericaria amentacea, E. brachycarpa and E. crinita) were observed on the intertidal rocky shores, with varied abundances across the MPA’s protection zones. Ericaria amentacea was the only species found in all zones, with a much higher cover percentage in the most protected area (zone A). However, its morpho-functional traits showed an inverse trend, with the largest thalli found in the moderately protected zone B and C. The remaining Cystoseira s.l. species were only found in zones B and C probably due to their wider area and greater habitat diversity compared to zone A. The presence of two non-indigenous species, Asparagopsis taxiformis and Caulerpa cylindracea, was observed exclusively in the less protected zones B and C. Our findings highlight the complex interactions between MPA protection levels and Cystoseira s.l. species conservation, with protection having, in some respects, a positive influence on selected Cystoseira s.l. species, indicating the importance of careful consideration in MPA design and management. Furthermore, this study provides a baseline for future monitoring of Cystoseira s.l. populations in light of ongoing environmental changes in the Mediterranean Sea. Full article

Bacillus cereus sensu lato (B. cereus s.l.) comprises mesophilic and psychrotolerant bacteria commonly found in natural environments as well as in organic and conventional milk. Due to their potential toxigenicity and antibiotic resistance, these bacteria pose a significant threat to consumer health. Organic milk production, which prohibits the use of antibiotics and artificial additives, may influence the composition of microbiota between milk types. This study aimed to compare the antibiotic resistance profiles and enterotoxic potential of B. cereus s.l. isolates from organic and conventional milk. The results indicate that, although conventional milk contains on average 3 times fewer B. cereus s.l. isolates, it has 10–15% more resistant isolates to selected beta-lactams, macrolides, and aminoglycosides. Regarding drug resistance, 21% of B. cereus s.l. isolates were multidrug-resistant, and as many as 42% were non-susceptible to two classes of antibiotics. Even among the sensitive isolates, bacteria from conventional milk exhibited on average 2.05 times higher MICs (minimal inhibitory concentrations) for beta-lactams, 1.49 times higher for erythromycin, 1.38 times higher for vancomycin, and 1.38 times higher for azithromycin. Antibiotic resistance was mostly associated with the origin of the isolates. These findings underscore the need for ongoing monitoring of antibiotic resistance and enterotoxicity among opportunistic B. cereus s.l. strains, which may pose challenges for public health and veterinary medicine. The results highlight that selective pressure associated with antibiotic use can drive resistance development in bacteria that are not the primary targets of antimicrobial therapy. Full article

Canine vector-borne diseases (CVBDs) pose a global threat to both canine and public health. This study evaluates the prevalence of Anaplasma spp., Ehrlichia spp., Borrelia burgdorferi sensu lato (s.l.), Rickettsia conorii, and Dirofilaria immitis in domestic dogs in Liguria, north-west Italy, a region where data were lacking. From 2013 to 2023, 8584 blood samples from shelter (74%) and owned dogs (26%) were submitted to the Istituto Zooprofilattico Sperimentale of Piemonte, Liguria and Valle d’Aosta (IZSPLV) for serological testing (indirect immunofluorescence and/or rapid immunochromatographic tests). Overall, 18.8% (95% CI: 18.0–19.7) of the dogs tested positive for at least one pathogen, with positivity against R. conorii antigen being the most frequently recorded (24.4%, 95% CI: 23.3–25.5). Lower prevalence levels were observed for Anaplasma spp. (1.82%, 95% CI: 1.47–2.23), Ehrlichia spp., (1.25%, 95% CI: 0.97–1.60), B. burgdorferi s.l. (0.22%, 95% CI: 0.11–0.39), and D. immitis (0.84%, 95% CI: 0.65–1.06). Positive cases for all pathogens increased over time, with prevalence differing significantly between owned and shelter dogs. Topographical factors, land use, and monthly relative humidity appeared to influence the positivity in the dogs. These results update the epidemiology of the investigated CVBDs in Liguria, indicating a widespread exposure to Rickettsia spp. among local dogs. Full article

Southwest China is extremely rich in fungal resources, and a large number of new taxa have been discovered in recent years. In the present study, we examined 26 specimens of the genera Hydnum and Lentaria sensu lato, most of which were obtained in Yunnan Province. Through ITS-nrLSU-tef1 phylogenetic analysis, combined with morphological studies and geographic analyses, five new species were described, viz. H. cremeum (奶油齿菌), H. flavoquamosum (黄鳞齿菌), H. roseoalbum (粉白齿菌), H. roseotangerinum (粉橙齿菌), and L. subalpina (亚高山木瑚菌). Furthermore, we also supplied new information on some known species, including host plants and new distribution records. We re-examined the holotype sequences of two known taxa, H. flabellatum and H. pallidomarginatum, treating H. flabellatum as a synonym of H. pallidomarginatum. Additionally, a key to Lentaria s.l. in China was provided. Full article
