Salinity Stress and Calcium in Pomegranate: Impacts on Growth, Ion Homeostasis, and Photosynthesis

Salinity has significant impacts on crops, a problem that is exacerbated under climate change conditions. For this reason, research is focused on possible ways to mitigate the impacts by adapting cultivation methods such as administering appropriate materials or formulations to plants. Therefore, this study investigated the effects of calcium (Ca2+) supplementation on the growth, physiology, and chemical composition of pomegranate plants (Punica granatum L. cv. ‘Wonderful’) grown under salinity stress. Young self-rooted plants were cultivated in pots containing a sand/perlite (1:1) mixture and irrigated with Hoagland’s nutrient solution amended with NaCl (0, 60, or 120 mM) and CaCl2·2H2O (0 or 10 mM). Salinity significantly reduced the fresh and dry weight of aboveground tissues; photosynthetic performance; chlorophyll content; and potassium (K), calcium (Ca), and magnesium (Mg) concentrations, particularly under high NaCl levels. Sodium (Na) accumulation increased in all plant parts, while nitrogen (N), manganese (Mn), and zinc (Zn) concentrations were elevated in basal leaves. Calcium supplementation mitigated several of these adverse effects, especially under moderate salinity. It helped maintain leaf biomass, supported K+ retention in roots, partially improved chlorophyll concentration, and limited Na+ accumulation in certain tissues. However, Ca2+ application did not consistently reverse the negative impacts of severe salinity (120 mM NaCl), and in some cases, interactions between Ca2+ and other nutrients such as Mg2+ were antagonistic. These findings confirm the inherent salt tolerance of pomegranate and demonstrate that calcium plays a partially protective role under salinity, particularly at moderate stress levels. Further research is needed to optimize Ca2+ use in saline agriculture and enhance sustainable cultivation of pomegranate in salt-affected soils. Full article

B-box (BBX) genes, as zinc finger transcription factors (TFs), play essential roles in regulating plant growth and development. In this study, we identified 23 BBX genes in the pomegranate (Punica granatum L.) genome. These genes were classified into five groups based on the distribution of conserved domains and phylogenetic relationships. Each PgBBX group exhibited similar molecular weights, theoretical isoelectric points (pI), gene structures, and conserved motif distributions compared with BBX members in Arabidopsis and Chinese white pear in corresponding groups. Syntenic analysis revealed segmental duplications of eight PgBBX gene pairs within the pomegranate genome. Additionally, twenty-seven and thirty-one orthologous BBX pairs were identified between PgBBX and AtBBX, and PgBBX and PbBBX, respectively. Promoter analysis revealed the presence of five types of cis-acting elements responding to light, phytohormones, stress, developmental signaling, and potential transcription factors (TFs). GO enrichment analysis confirmed that most PgBBX genes function as TF involved in plant growth and development. RNA-seq data indicated that PgBBX5 was primarily expressed in leaves and flowers, with increased expression in different fruit tissues during ripening. Moreover, PgBBX5 showed a high degree of sequence similarity with anthocyanin-related homologs, including AtBBX24, PhBBX24, FaBBX24, MdCOL4, and PyBBX24. During the ripening of ‘Tunisia’ fruits, PgBBX5 expression was positively correlated with the dynamic changes in anthocyanin content and the expression of key anthocyanin biosynthetic and transport genes. Furthermore, subcellular localization suggested that PgBBX5 encodes a nuclear-localized protein. This study provides a comprehensive characterization of the PgBBX family, offering valuable insights into the mechanisms underlying anthocyanin accumulation during pomegranate fruit ripening. (1) Background. Although Arum spp. are toxic in their raw state, they are sometimes used as food within their native ranges. (2) Methods. We review the available literature in order to provide an overview of its use and detoxification procedures worldwide. (3) Results. The food use of lords-and-ladies was already mentioned by Theophrastus, Dioscorides, Matthioli, Durante, Gerard, and Sirennius. In the references concerning 19th–21st-century use, seven species were identified: A. cyrenaicum, A. discoridis, A. italicum, A. maculatum, A. orientale, A. palaestinum, and A. rupicola. Past or current culinary use of the plant has been recorded in Morocco, Libya, the United Kingdom, the Scilly Islands, Germany, Switzerland, Italy, Romania, Ukraine (including Crimea), Czechia, Slovenia, Croatia, Bosnia-Herzegovina, Albania, Georgia, Türkiye, Syria, Palestine, Lebanon, Israel, Iraq, and Iran. (4) In Europe, rhizomes were used, mainly as a famine food. In SW Asia, the aerial parts remain an important element of local cuisine. Several detoxification procedures are used before consumption, such as prolonged boiling, often involving straining the boiled water and lowering the pH with lemon juice, sumac, citric acid, sorrel leaves, or pomegranate juice. Understanding the ammonium (NH4+) uptake and transport systems, particularly AMT1 genes, is important for plant growth and defense. However, there is a lack of research on identifying and analyzing AMT1 genes in pomegranate, emphasizing the need for further investigation in this area. Five AMT1 genes (PgAMT1-1 to PgAMT1-5) were identified, all of which contain the PF00909 domain, a feature of ammonium transporters. Various characteristics of these genes, including gene length, coding sequence length, and chromosomal locations, were examined. This study evaluated the isoelectric point, hydropathicity, conserved domains, motifs, and synteny of the PgAMT1 proteins. Phylogenetic analysis confirmed the homology of PgAMT1 genes with previously reported AMT in Arabidopsis and tomato. The tissue-specific expression analysis of PgAMT1 genes revealed distinct patterns: PgAMT1-1 and PgAMT1-2 were predominantly expressed in flowers, PgAMT1-3 exhibited notable expression in roots, leaves, and flowers, PgAMT1-4 was primarily expressed in leaf tissue, while the expression of PgAMT1-5 was detected in both leaves and roots. The impact of salt-induced stress on AMT1 gene expression was also examined, revealing that PgAMT1-1, PgAMT1-2, and PgAMT1-4 expression is reduced under increased salt stress. These expression modifications can help regulate NH4+ assimilation in conditions of elevated salinity, maintaining cellular homeostasis and ion balance. This study contributes to the comprehensive identification of the AMT1s gene family in pomegranate; however, further research on the functional characterization of the identified PgAMT1s is needed. Food spoilage is a natural process that influences the quality and safety of food products, negatively affecting their nutritional and organoleptic composition. In these regards, traditional industrial food preservation processes often rely on the use of traditional preservation techniques to extend food shelf life, while ensuring microbiological and chemical stability without compromising the product’s sensory characteristics. However, in recent years, consumers have become increasingly wary of chemical food additives; they often associate their use with potential health risks and negative impact on product appeal. In addition, this is compounded by an increasingly compellent European regulatory framework, as well as efforts in the search of natural and sustainable alternatives for food preservation. In this context, this review explores the potential of natural additives, such as polyphenols, flavonoids, and antioxidants, derived from agro-industrial waste, including fruit peels, vegetable by-products, and seeds. These compounds exhibit strong antioxidant and antimicrobial properties, which not only extend the shelf life of food products but also enhance their safety and quality. Considering vegetable wastes (i.e., pomegranate peels, olive leaves, olive mill wastewater, and grape pomace) as the main by-products from which natural additives can be extracted, this study provides an overview of their efficacy in preventing lipid oxidation and reducing microbial growth, while maintaining sensory properties. This could represent an opportunity both for maintaining food quality and prolonging food shelf life by valorizing by-products to be otherwise disposed of, and also contribute to mitigating the environmental impact associated with the food industry and to optimize food preservation processes. In addition, it highlights the possibility of employing sustainable alternatives to synthetic additives, capable of extending the shelf life of food products while ensuring their safety for human consumption. The use of biological methods to control plant diseases has garnered attention due to their eco-friendly and economically viable nature. Trichoderma spp. are the most ubiquitous fungal saprophytes that can often grow as opportunistic symbionts, are eco-friendly, and are not reported to exert any environmental hazard. Soil-borne pathogens can significantly impact the yield of chilli and tomato crops. The study was conducted to explore the impact of various salts (NaCl, MgCl2, CaCl2, and KCl) and their concentrations (1%, 5%, 10%, and 15%) on the mycelial growth and sporulation of Trichoderma viride followed by its mass multiplication on diverse organic substrates like wheat seeds, wheat husks, mungbean seeds, maize seeds, rice seeds, pea seeds, sorghum seeds, banana peel, apple peel, pomegranate peel, citrus peel, tomato waste, chilli waste, spinach waste, cabbage waste, potato peel, onion peel, cucumber peel, carrot peel, used black tea leaves, used green tea leaves, poultry waste, and cow and goat dung. The study assessed the biocontrol potential of Trichoderma viride against important soil-borne pathogens in chilli (Pythium aphanidermatum, Phytophthora capsici, and Fusarium oxysporum) and tomato (Pythium aphanidermatum, Phytophthora infestans, and F. oxysporum) cropping in the pot and field experiments using saline and alkaline soils. Seed treatment with T. viride significantly enhanced the germination rates of both chilli and tomato crops, with sorghum being the most conducive substrate for mass-multiplying T. viride. The results revealed that lower salt concentrations minimally affected T. viride growth, while higher concentrations hampered both growth and sporulation. Optimal disease control and plant height were observed at a 20 mg concentration of T. viride, consequently applied in vivo using various application methods, i.e., seed treatment, root dip, irrigation, and mixed treatments (all the methods like seed treatment, root dip method, and irrigation method applied together) to manage soil-borne pathogens. Particularly, T. viride application through irrigation and mixed treatments demonstrated strong efficacy. These findings underscore the potential of T. viride application in saline and alkaline soils to manage soil-borne pathogens like Pythium, Phytophthora spp., and Fusarium spp. This study lays the foundation for the practical application of biocontrol agents, like T. viride, in Pakistani agricultural conditions. Moreover, there is a need for further exploration into the genetic mechanisms involved in disease inhibition and plant growth promotion, along with understanding the impact of T. viride on the metabolic pathways of host plants. Pomegranate (Punica granatum) is a well-known fruit tree species and a significant pioneer ecological species on saline–alkali land with moderate resistance to salt stress. To explore its response mechanism to salt stress could provide valuable insights into the molecular and physiological strategies plants employ to adapt and survive in high-salt environments. In this study, changes in physiological parameters and gene expressions were examined following salt treatment. After 72 h of salt treatment, change patterns of SOD and POD differed between high and low salt concentrations. Similar changes were found in the contents of proline and total soluble sugar. RNA-Seq analysis of fifteen samples detected 32,630 genes from the pomegranate genome data. A total of 6571 DEGs, including 374 TFs, were identified across different treatments. Six special modules and 180 hub genes were obtained by WGCNA analysis. Functional annotation highlighted signaling pathways and the accumulation of primary and secondary metabolites as significant pathways. These findings could reveal the salt tolerance mechanism in pomegranate leaves, offering a theoretical foundation for enhancing plant salt tolerance through genetic engineering. Full article

The goal of the research was to determine the impact of fortification with polyphenolic compounds on (i) sensory attractiveness (global satisfaction, appearance, colour, odour, flavour, sweetness, bitterness), (ii) content of polyphenols and colour (L*, a*, b*) after the baking process and (iii) their bioactive potential (antioxidants activity and inhibiting of α-amylase and α-glucosidase enzyme). Fortification was made with extracts of polyphenolic compounds of selected plant raw materials rich in polyphenols from quince (fruits), tilia (flowers), pomegranate (skin), passion fruit (endocarp), sour cherries (leaves), haskap and chokeberry (berries), silver skin (coffee beans), rosehip (seeds). Depending on the nature of the polyphenol extract, flavan-3-ols (monomeric and polymeric), phenolic acid, flavonols and anthocyanins were identified in the product in amounts ranging from 53.7 to 212.6 mg/100 g DM. Cookies’ colour (L*, a*, b*) depended on the type of polyphenol extract used for fortification. Cookies with haskap, chokeberry and sour cherry presented the highest antioxidant potential. Cookies with chokeberry, haskap and rosehip presented high activity in inhibiting α-amylase (65.5, 60.6 and 62.2% of inhibition, respectively), but cookies with haskap, silver skin and quince in inhibiting α-glucosidase activity (23.0, 20.4 and 21.4% of inhibition, respectively). In the sensory evaluation, the most attractive were cookies with rosehip and pomegranate (6.3 and 5.8 score, respectively), but the lowest ratings were given to cookies with passion fruit and silver skin but especially quince cookies, which obtained the lowest desirability (3.7 score). The acceptability of fortified cookies was determined to the least extent by monomeric flavan-3-ols and phenolic acids (in minus in odour/flavour, bitterness, sweetness and global satisfaction), but anthocyanins, polymeric procyanidins and flavonols had the most significant positive impact on consumer acceptance of the assessed features, i.e., global satisfaction, odour/flavour, sweetness and bitterness (positive consumer drivers). The pharmacological effects of pomegranates have been described considering metabolic aspects such as hypoglycemic and hypolipidemic activities. The pomegranate extract has activity on the central nervous system (CNS) as a natural antidepressant and anxiolytic. The chemical composition of pomegranates is complex since the bioactive compounds are multiple secondary metabolites that have been identified in the extracts derived from the peel, seed, flowers, leaves, or in their combination; so, it has not been easy to identify an individual compound as responsible for its observed pharmacological properties. From this point of view, the present review analyzes the effects of crude extracts or fractions of pomegranates and their possible mechanisms of action concerning antidepressant- and anxiolytic-like effects in animal models. Serotonin receptors, estrogen receptors, the peroxisome proliferator-activated receptor gamma (PPARγ), or monoamine oxidase enzymes, as well as potent antioxidant and neuroplasticity properties, have been described as possible mediators involved in the antidepressant- and anxiolytic-like behaviors after pomegranate treatment. The pharmacological effects observed on the CNS in experimental models associated with a specific stress level suggest that pomegranates could simultaneously modulate the stress response by activating several targets. For the present review, scientific evidence was gathered to integrate it and suggest a possible pathway for mediators to be involved in the mechanisms of action of the pomegranate’s antidepressant- and anxiolytic-like effects. Furthermore, the potential benefits are discussed on comorbid conditions with anxiety and depression, such as perimenopause transition and pain. Full article

Alternaria black spot of pomegranate (Punica granatum) was reported for the first time in Italy. In spring 2023, an outbreak of this disease was noticed in commercial pomegranate ‘Wonderful’ orchards of the municipality of Misterbianco (Sicily), following an unusually rainy period. A total of 30 randomly selected Alternaria isolates recovered from typical necrotic spots of leaves and fruits were characterized. Based on the colony morphology on solid agar media (PDA and MEA), isolates were separated into three distinct morphotypes (1, 2, and 3). The first two morphotypes comprised only isolates from fruits, while morphotype 3 comprised only isolates from leaves. Multigene phylogenetic analysis of four DNA regions, including internal transcribed spacer (ITS), translation elongation factor 1-α (EF-1α), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), and a SCAR marker (OPA10–2), identified the isolates of morphotypes 1 and 2 as Alternaria alternata and morphotype 3 isolates as A. arborescens. In pathogenicity tests on unwounded leaves and fruit, the isolates of all three morphotypes produced symptoms on the leaves of three pomegranate cultivars, ‘Acco’, ‘Wonderful’, and ‘Etna’. The symptoms on ‘Acco’ leaves were the least severe. Conversely, the fruits of ‘Acco’ were the most susceptible. The isolates of morphotypes 2 and 3 were not pathogenic on the fruits of ‘Wonderful’ and ‘Etna’. This is the first report of Alternaria black spot in Italy and of A. arborescens associated with Alternaria black spot of pomegranate worldwide. Full article

Quality and safety are one of the main concerns of the European Union in food preservation. Using chemical additives extends the shelf-life of fresh foods but raises consumer’s concerns about the potential long-term carcinogenic effects. Using natural substances derived from agro-industrial by-products, which have significant antimicrobial and antioxidant activities, could extend the shelf-life of fresh foods such as meat. Furthermore, they can provide nutritional improvements without modifying organoleptic properties. This study analyzes the antimicrobial activity of pomegranate peel extract (PPE) and the antioxidant activity of olive leaf extract (OLE), added at concentrations of 10 mg g−1 and 0.25 mg g−1, respectively, to minced poultry and rabbit meat. PPE exhibited in vitro antimicrobial activity against foodborne pathogens starting at 10 mg/well. PPE and OLE determined a reduction in colony count over a storage period of 6 days at 4 °C. Additionally, the combination of PPE and OLE showed antioxidant effects, preserving lipid oxidation and maintaining pH levels. The obtained results demonstrate that PPE and OLE can be recommended as food additives to preserve the quality and extend the shelf-life of meat products. Full article

This study aimed to analyse the impact of a simulated human digestion process on the composition and functional properties of dietary fibres derived from pomegranate-peel, tomato-peel, broccoli-stem and grape-stem by-products. For this purpose, a computer-controlled simulated digestion system consisting of three bioreactors (simulating the stomach, small intestine and colon) was utilised. Non-extractable phenols associated with dietary fibre and their influence on antioxidant capacity and antiproliferative activity were investigated throughout the simulated digestive phases. Additionally, the modifications in oligosaccharide composition, the microbiological population and short-chain fatty acids produced within the digestion media were examined. The type and composition of each dietary fibre significantly influenced its functional properties and behaviour during intestinal transit. Notably, the dietary fibre from the pomegranate peel retained its high phenol content throughout colon digestion, potentially enhancing intestinal health due to its strong antioxidant activity. Similarly, the dietary fibre from broccoli stems and pomegranate peel demonstrated anti-proliferative effects in both the small and the large intestines, prompting significant modifications in colonic microbiology. Moreover, these fibre types promoted the growth of bifidobacteria over lactic acid bacteria. Thus, these results suggest that the dietary fibre from pomegranate peel seems to be a promising functional food ingredient for improving human health. Full article

In modern agricultural production, where a small number of commercial cultivars dominate, the collection, evaluation, and preservation of germplasm are important tasks to reduce the erosion of genes and preserve biodiversity. The aim of this study is to characterize the morphological and fruit chemical properties of the pomegranate germplasm grown on the East Adriatic coast, including the commercial cultivars ‘Hicaznar’, ‘Granada’, and ‘Wonderful’, and to highlight the characteristics with the greatest discriminating power. The characterization of the tree, leaf, flower, arils, seed, and juice was carried out using the UPOV descriptor. The colors of the peel, arils, and juice were analyzed according to the CIEL*a*b* method, total soluble solids were measured using refractometers, and total acidity was determined by titration with 0.1 M NaOH. The research results showed significant diversity between the cultivars, which were grouped into several clusters using an unsupervised analysis technique. Factors such as plant vigor, plant growth habit, predominant number of leaves per node on young shoots, crown type, fruit shape, fruit shape in cross-section, peel weight, total aril weight, aril weight, number of arils per fruit, seed length and width, seed yield, total acidity, TSS/TA ratio, and color parameters of the peel, arils, and juice showed high variability, indicating their strong discriminating power in determining the phenotypic diversity of pomegranate. Nowadays, there is a growing awareness of environmental protection, new findings in the field of sustainable chemistry, the use of biodegradable materials, and the increased use of eco-friendly textile products. For this reason, natural dyes are being used more and more frequently, giving rise to a new way of decorating textiles, namely, plant transfer printing, popularly known as “eco-printing”, in which the shape and/or pigment of a plant is transferred to the textile. In addition, the great interest of the young generation in the application and research into the use of natural dyes can create incentives for cultural and social sustainability through the preservation of national heritage. Plant transfer printing is a method that combines scientific technology and artistic design with corresponding benefits for the eco system. The very fact that the patterns are unique and unpredictable brings out the notion of artistic freedom. In the work, plant transfer printing was carried out on undyed cotton material and on material dyed with pomegranate peels, walnut leaves, coffee, and aleppo pine bark. The influence of the pH value and the capillarity of the fabric, as well as the treatment of the leaves with iron(II) sulphate heptahydrate solution, on the aesthetics of the print and the colour fastness during washing was investigated. Based on the optimised parameters and a sustainable fabric design, the clothing collection “Hamadryad”, inspired by Greek mythology, was realised. Full article

Insights into the PYR/PYL/RCAR Gene Family in Pomegranates (Punica granatum L.): A Genome-Wide Study on Identification, Evolution, and Expression Analysis

The response of plants to abiotic stress is intricately mediated by PYR/PYL/RCARs, key components within the ABA signal transduction pathway. Despite the widespread identification of PYL genes across diverse plant species, the evolutionary history and structural characteristics of these genes within the pomegranate (Punica granatum L.) remained unexplored. In this study, we uncovered, for the first time, 12 PgPYLs from the whole genome dataset of ‘Tunisia’, mapping them onto five chromosomes and categorizing them into three distinct subgroups (Group I, Group II, and Group III) through phylogenetic analysis. Detailed examination of the composition of these genes revealed similar conserved motifs and exon–intron structures among genes within the same subgroup. Fragment duplication emerged as the primary mechanism driving the amplification of the PYL gene family, as evidenced by intra-species collinearity analysis. Furthermore, inter-species collinearity analysis provided insights into potential evolutionary relationships among the identified PgPYL genes. Cis-acting element analysis revealed a rich repertoire of stress and hormone response elements within the promoter region of PgPYLs, emphasizing their putative roles in diverse signaling pathways. Upon treatment with 100 μmol/L ABA, we investigated the expression patterns of the PgPYL gene family, and the qRT-PCR data indicated a significant up-regulation in the majority of PYL genes. This suggested an active involvement of PgPYL genes in the plant’s response to exogenous ABA. Among them, PgPYL1 was chosen as a candidate gene to explore the function of the gene family, and the CDS sequence of PgPYL1 was cloned from pomegranate leaves with a full length of 657 bp, encoding 218 amino acids. Tobacco transient expression analysis demonstrated a consistent trend in the expression levels of pBI121-PgPYL1 and the related genes of the ABA signaling pathway, both of which increased initially before declining. This study not only contributes to the elucidation of the genomic and structural attributes of PgPYL genes but also provides a foundation for understanding their potential functions in stress responses. The identified conserved motifs, evolutionary relationships, and expression patterns under ABA treatment pave the way for further research into the PgPYL gene family’s role in pomegranate biology, offering valuable insights for future studies on genetic improvement and stress resilience in pomegranate cultivation.
