Pseudomonas savastanoi pv. phaseolicola, the causal agent of halo blight of bean, is a useful model for studying bacterial adaptation to plant infection. Although its toxins, effectors, population dynamics, and epidemiology have been investigated for decades, these aspects have often been treated separately. This narrative synthesis reinterprets the available literature within a stage-based infection framework encompassing host-surface colonization, establishment of a compatible apoplastic habitat, evasion of host immune responses, multiplication, and spread to another compatible host. Particular attention was given to studies published after the last major review on this pathovar, whereas older studies were retained when they represented primary reports, classical experimental evidence, or information that has not been superseded. The review shows that successful infection is best understood as involving opportunistic entry, rapid transition from an epiphytic to a pathogenic lifestyle, effector-dependent immune suppression, phaseolotoxin-mediated manipulation of host primary metabolism, the exploitation or establishment of hydrated apoplastic conditions, and environmentally driven dissemination. It also distinguishes experimentally supported mechanisms from unresolved or hypothesis-generating areas, including stomatal reopening, micronutrient acquisition, population-level division of labor, alternative hosts, and symptomless reservoirs. The major synthesis emerging from this review is that the infection biology of P. savastanoi pv. phaseolicola cannot be reduced to individual virulence factors because disease development depends on how these factors are coordinated with host-derived resources, immune constraints, and environmental opportunities. Climate change politics has been largely analyzed through the lenses of a liberal international order. This is the most favorable approach, because liberalism contains a powerful universalistic strand, defends the rights of people, and engages in multilateral negotiations and agreements, which are important to deal with a global issue that requires intra- and intergenerational solidarity. Yet despite robust scientific consensus and decades of international multilateral agreements under the United Nations, global greenhouse-gas atmospheric concentrations continue to increase, and high fossil-fuel dependence persists. One may say that without those negotiations, the situation would be worse, but humanity is increasingly distant from complying with the objective of the United Nations Framework Convention on climate change (UNFCCC). The present work addresses climate change politics under liberal and neorealist international orders and follows the Mearsheimer hypothesis of a transition from a unipolar liberal order to a bipolar neorealistic bounded orders dominated by the US and China. The effect of international orders on sustainability and, more specifically, on climate change politics is analyzed with a methodology based on three structural determinants: (1) the world evolution of climate change variables; (2) primary-energy sources and critical minerals, and (3) climate change responses—mitigation, adaptation and climate geoengineering. The distinct energy and climate policies of the US and China are discussed using these structural determinants. US climate change policy appears to be less driven by climate observation, science and the severity of harmful impacts of climate change than by the vested interests of the fossil-fuel industry. It is argued that solar radiation manipulation (SRM) is a technological fix involving negative side-effects, uncertainties, risks and geopolitical implications, while lacking an agreed international governance framework. Potential deployment is more likely under a neorealistic international order, although it adds further uncertainty and risks without solving the climate change challenge. Full article

Background/Objectives. The intestinal microbiota is a key contributor to postoperative complications, yet direct interventions targeting dysbiosis—antibiotics, probiotics, and synbiotics—have produced inconsistent results. This paradox indicates a fundamental gap in understanding host–microbiota interactions under surgical stress. We aimed to re-examine the causal role of dysbiosis in postoperative pathogenesis and propose a revised therapeutic paradigm centered on host barrier protection. Methods. A narrative literature review was conducted, searching PubMed/MEDLINE, Scopus, and Web of Science for articles published between 2009 and 2025. Reference lists of included publications were additionally screened. Studies in English and Russian were eligible; 107 references were included. Results. We hypothesize that dysbiosis in surgical patients may, at least in part, represent a predictable ecological response to systemic hypoperfusion, pharmacological burden, and ischemia–reperfusion injury, rather than acting solely as an independent pathogenic agent. Microbial shifts, characterized by the depletion of short-chain fatty acid-producing commensals and the expansion of pathobionts, frequently accompany epithelial injury; however, available human data are predominantly observational and do not permit definitive determination of the temporal sequence. This hypothesis provides the conceptual foundation for the proposed therapeutic reorientation. Conclusions. The present findings support the rationale for transitioning from microbiome manipulation to a “host-first” strategy, which prioritizes the restoration of intestinal barrier integrity through the administration of cytoprotective agents and targeted metabolic substrates (glutamine and butyrate). We propose the Gut Resilience Index (GRI) as a theoretical construct to identify patients approaching a critical threshold necessitating rescue therapy. It must be emphasized that both the “host-first” strategy and the GRI remain hypothetical frameworks requiring prospective validation. The most critical next steps include the development and validation of the GRI in prospective cohort studies, as well as randomized controlled trials directly comparing barrier-oriented strategies with standard care. Plant U-box E3 ubiquitin ligases (PUBs) are involved in various biological processes in response to abiotic stress. Recent studies have shown that E3 ubiquitin ligases can regulate the production of important compounds in medicinal plants by targeted degradation of transcription factors. Abscisic acid (ABA), a plant stress response hormone, can cause changes in the content of the medically important monoterpene indole alkaloids (MIAs) in U. rhynchophylla. In this study, we explored the relationship between UrPUB gene expression and MIA content. ABA was applied to tissue-cultured seedlings of U. rhynchophylla, resulting in consistent changes in the content of four MIAs. Seventy-three UrPUB genes were identified and bioinformatically characterized from the genome sequence of U. rhynchophylla. The expression levels of transcription factors involved in regulating the biosynthesis pathway of MIAs and UrPUB genes with higher RNA transcript levels in the roots were determined. Co-expression association analysis revealed that UrPUB17, UrPUB40, UrPUB41, UrPUB44 and UrPUB55 negatively correlate with UrGATA8 and UrWRKY37 under ABA stimulation. Based solely on these correlation data, we hypothesize that these UrPUBs might regulate MIA biosynthesis via ubiquitination of UrGATA8 and UrWRKY37, but direct evidence (protein interaction, ubiquitination, degradation, or genetic manipulation) is lacking. This study identified the UrPUB gene family in the U. rhynchophylla genome and proposes this ubiquitination model as a testable hypothesis, not a demonstrated mechanism. These findings provide new insights into the biological function of the PUB family in response to ABA. Full article

Background: Paranasal sinuses and mastoid air cells have been attributed to multiple functions—such as voice resonance, cranial lightening, and pressure regulation—yet their potential role in local thermal homeostasis remains underappreciated. The thermoregulatory hypothesis, first proposed in the mid-twentieth century, was largely abandoned after the mid-century, when anthropological findings of climate-correlated variation seemed contradictory. Hypothesis: We propose that pneumatized skull regions form a three-component craniofacial biothermal system that maintains thermal stability in the ocular vitreous and vestibular endolymph, two avascular, temperature-sensitive structures that lack intrinsic thermoregulatory capacity. This represents a novel integration that explicitly links paranasal and mastoid pneumatization into a coordinated system that protects sensory organs, distinct from previous brain-cooling hypotheses. Mechanism: The system comprises: (1) passive thermal insulation via air spaces, providing ~15-fold greater thermal resistance than bone; (2) active cold protection via mucosal heat delivery (estimated 2–5 W capacity); and (3) active heat dissipation via evaporative cooling (estimated 0.3–0.5 W capacity). This architecture provides asymmetric protection, with cold buffering exceeding heat dissipation by approximately 5- to 15-fold, consistent with thermodynamic constraints and putative evolutionary priorities. Evidence: Preliminary observations consistent with this hypothesis include the anatomical proximity of pneumatized regions to the vitreous and labyrinth, intranasal selective brain cooling studies, and clinical observations after mastoidectomy showing preserved pressure buffering but reduced vestibular thermal insulation under extreme stimulation. Climate-correlated pneumatization patterns are consistent with bidirectional thermal adaptation. Implications: We present five falsifiable predictions that can be tested with thermographic imaging, pharmacological manipulation, and computational modeling. Validation could inform surgical planning, explain postoperative thermal-sensitivity symptoms, and provide evolutionary insights into craniofacial adaptation. Full article

Host-finding behavior of insect herbivores is a key determinant of herbivory intensity in agricultural and forest ecosystems, which often drives excessive pesticide application for pest control. While host plant apparency theory explains herbivore host detection, and push–pull strategies manipulate this behavior, both produce inconsistent results and remain mechanistically disconnected. Existing frameworks like the Resource Concentration Hypothesis focus mainly on host density, ignoring the multidimensional, context-dependent nature of apparency. Here, we synthesize forest and agricultural research to develop the first unified framework linking these two concepts. We show that host plant apparency is not intrinsic but shaped by plant morphology, non-host identity, and spatial arrangement. Push–pull strategies exploit this relativity by redesigning the chemical and physical apparency landscape. We argue that: (1) push–pull system success requires reducing main crop apparency while enhancing trap crop apparency; (2) trap crops may fail when their dual functions, olfactory attraction or physical interception, are misinterpreted, with profound implications for spatial design; and (3) this integration resolves field contradictions by framing them within a common bottom-up mechanism. Our framework provides a generalizable principle for sustainable pest management: effective control depends on understanding what makes host plants apparent to target pests in their specific local environment. The black soldier fly, Hermetia illucens (Linnaeus, 1758), is a globally farmed insect reared to convert organic waste into useful biomatter and materials. While research on optimizing bioconversion rates and larval development through substrate manipulation has surged, studies on larval behavioral responses to abiotic factors lag. This benchtop-scale study is the first to examine larval behavior in response to reduced airflow. The results suggest third to fifth instar H. illucens larvae, which typically burrow within the substrate and avoid the surface, are significantly more likely to aggregate at the surface of their substrate when in a restricted airflow environment. More than half of third instar and nearly all 5th instar larvae aggregated at the surface when airflow was restricted, compared to less than a quarter under ventilated conditions. This data should stimulate future directions of hypothesis building in larval behavioral and physiological responses to airflow with the goal of using behavior modification to facilitate larval production. Background: Large language models (LLMs) are increasingly considered as adjuncts for differential diagnostic reasoning, yet their sensitivity to gender-attributed cues in the persona prompt—particularly for neglected tropical diseases (NTDs) and in non-English clinical settings—remains poorly characterised. Objective: The objective of this study was to compare the diagnostic accuracy of one proprietary and three open-weight LLMs for Chagas disease (CD) and visceral leishmaniasis (VL) under paired persona-prompt conditions in which the only manipulated variable was the linguistic gender of the simulated medical persona. Methods: This experimental, paired study evaluated ChatGPT-4o, LLaMA 3 70B, Meditron-70B, and Mixtral 8x7B across 12 cases per disease (n = 24) from real records at a Brazilian teaching hospital. The primary outcome was top-five diagnostic accuracy. A committee of five infectious-disease specialists assessed the biological plausibility of all differentials. Paired comparisons used Wilcoxon signed-rank tests; 95% confidence intervals were calculated using the Wilson-score method. Results: ChatGPT-4o achieved the highest accuracy (CD: 100% under both prompts; VL: 83.3–91.7%). LLaMA 3 70B and Mixtral 8x7B showed moderate performance (41.7–83.3%); the medically fine-tuned Meditron-70B exhibited paradoxically poor accuracy (16.7–25.0%) and the lowest committee-rated plausibility scores. A consistent small numerical trend favoured the female prompt across most model–disease combinations (differences of 0–16.7 percentage points), but no comparison reached statistical significance (all p > 0.05). Conclusions: Gender-attributed persona-prompt variation did not produce a systematic effect on LLM diagnostic accuracy for CD or VL. ChatGPT-4o outperformed the three evaluated open-weight alternatives, and medical-domain fine-tuning did not confer the expected advantage. Expert-validated assessment of hypothesis plausibility should complement target-disease accuracy in clinical LLM evaluation studies, particularly for NTDs. Full article

How do ryanodine receptors (RyRs) open simultaneously to trigger the contraction of whole myofibrils within a large skeletal muscle cell? One possible answer is the uniformity of mechanosensitive RyRs, which is mechanically forced by the neighboring environment, including proteins. Here, we review papers addressing this proposed “mechanical sarcoplasmic reticulum (SR) paradigm”. Crystals of the molecular complexes comprising RyR and L-type voltage-gated Ca2+ channels were observed at the T-tubule/SR junction in situ using cryo-electron tomography. Observations of the SR vesicles isolated from rabbit and scallop cross-striated muscles using negative staining and transmission electron microscope raised a hypothesis of dynamic rearrangement of the Ca2+-ATPase (ATPase) molecules in response to cytoplasmic calcium concentration, as follows: (i) At a low calcium concentration where the ratio of operating ATPase molecules to the total molecules is at a submaximal level, the ATPase molecules form, at least in part, their cylindrical crystals in the SR membrane with the help of ATP; this results in the elongation of the SR vesicles. (ii) High concentrations of calcium, at which the ratio of operating ATPase molecules is maximal, reversibly collapse the ATPase crystals to transform the elongated vesicles into round forms comprising tightly attached crystal patches. These data further lead to the idea that the reversible growth of cylindrical ATPase crystals provides a dynamic crystalline network, which acts as an “SR membrane-endoskeletal motor” to manipulate the SR movement. The possibility of interactions between ATPase crystals and neighboring RyR crystals is also discussed. Full article

Insight problems are traditionally presented in verbal or visual form; however, some research suggests that physically interacting with the problem materials may facilitate insight problem solving. In contrast, the more recent literature found no performance improvement with manipulable materials, except for spatial problems (e.g., the “Eight Coins problem”). In Experiment 1, to challenge the hypothesis that the facilitating effect of material manipulation is mainly related to spatial insight problems, two groups of participants were presented with another spatial insight problem, the “Pencil problem” (a new version of the traditional “Match problem”), either in paper format or in a physical interactive condition. The results showed no significant differences in success rates based on presentation format. Therefore, manipulating the materials does not appear to facilitate solving spatial insight problems per se. In accordance with the misunderstanding theory of insight problem solving, the interacting effect may be limited to situations in which manipulation allows individuals to grasp functional aspects that are crucial for restructuring and reaching the solution. To test this hypothesis, we ran a further study (Experiment 2) in which two groups of participants were presented with a “Matchstick Arithmetic problem” in paper format; one group received the standard representation of the task (I = II + II), while the other, a picture of the arithmetic equation with real matchsticks, whose realistic representation indirectly clarified the misunderstanding that caused the ‘problem forming’ and the impasse. There was a significant increase in solutions in the second condition. In this case, the presence of overlapping matchsticks forming the plus operator helps the participants understand that arithmetic symbols, as well as numbers, can be decomposed, thereby overcoming the misunderstanding that lies in the problem’s critical points. This study investigates whether metacognitive prompting for responsible generative AI (GenAI) use can enhance students’ psychological sustainability in AI-assisted learning. Using a face-to-face classroom experiment (N = 148; 74 prompting, 74 control), we examined how metacognitive prompts embedded in a GenAI-assisted academic task influence learning anxiety and academic self-efficacy, and whether anxiety mediates the effect on self-efficacy. Manipulation checks indicated that the prompting condition produced significantly higher metacognitive engagement than the control condition (t(146) = 7.50, p < 0.001, d = 1.23). Hypothesis tests showed that metacognitive prompting reduced learning anxiety (b = −0.68, p < 0.001) and increased academic self-efficacy (b = 0.40, p = 0.008). Learning anxiety was negatively associated with self-efficacy (b = −0.42, p < 0.001). Mediation analyses using bootstrap confidence intervals revealed a significant indirect effect of prompting on self-efficacy via reduced anxiety (ab = 0.26, 95% CI [0.12, 0.43]), indicating partial mediation. These findings suggest that responsible GenAI use can be supported through instructional design. Background/Objectives: Severe acquired brain injury (sABI) disrupts early rehabilitation because arousal fluctuates, trunk control is fragile, and agitation limits therapy tolerance; land-based practice is frequently constrained by fall risk and staffing. We aim to reframe aquatic therapy as a programmable multisensory environment to stabilize arousal and support axial alignment before conventional impairment targets are feasible. Here, programmable denotes the deliberate titration and reporting of water depth, turbulence or perturbation, temperature, body orientation, and flotation and manual support as intervention inputs. Methods: This perspective integrates principles from neurobehavioral assessment, motor control, and immersion physiology to propose the Arousal–Alignment–Action loop as a falsifiable model and to define manipulable aquatic inputs (water depth, turbulence or perturbation, temperature, body orientation, and flotation and manual support) as dosing parameters. We outline a pragmatic testing ladder (within-session micro-experiments, feasibility studies, and embedded evaluations) and a minimal outcomes and confounder set to support cumulative evidence. Results: The framework links state regulation to alignment and goal-directed behavior, specifies predictions that can fail, and highlights boundary conditions (sedation, autonomic instability, pain, recent surgery or wounds, and cervical or cardiopulmonary constraints). A minimal outcome package spanning arousal/responsiveness, trunk control, behavioral dysregulation, participation/tolerance, and basic physiology is proposed, with optional objective adjuncts for mechanism-oriented studies. Conclusions: Treating water as a measurable and titratable medium, rather than a generic modality, may reduce early intensity bottlenecks and improve implementability and comparability of aquatic neurorehabilitation research in medically stable sABI; however, the model is intended as hypothesis-generating until supported by stronger direct clinical evidence. Emerging evidence suggests that certain cestodes, including Taenia solium, may actively modulate the host’s hormonal and immune environment to facilitate their survival. This study aimed to determine whether patients diagnosed with neurocysticercosis (NCC) exhibit immunoendocrine alterations associated with infection. A clinical study was conducted in Honduras, enrolling 11 adult NCC patients (9 female, 2 male) and 11 age- and sex-matched healthy controls. Serum concentrations of seven hormones and two cytokines were evaluated. Compared to controls, NCC patients showed significantly elevated levels of 17β-Estradiol (E2), Progesterone (P4), Androstenedione (A4), Luteinizing Hormone (LH), Follicle-Stimulating Hormone (FSH), Interleukin-6 (IL-6), and Interleukin-10 (IL-10). Conversely, Free testosterone (FT) and Dihydrotestosterone (DHT) levels were significantly reduced. These findings support the hypothesis that T. solium may manipulate host immunoendocrine pathways to promote its establishment and persistence within the central nervous system. Full article

Empirical evidence suggests that affective responses to climate change are predictive of pro-environmental attitudes, intentions, and behavior and may thus contribute to sustainability. However, that evidence is mostly correlational. We present experiments designed to test the mediational hypothesis that exposure to climate change information elicits negative affective responses, which in turn increase pro-environmental intentions. In Study series 1 (8 studies, total n = 819), we used a measurement-of-mediation design. We provided participants with information about the devastating effects of climate change or other non-threatening information. Neither providing that information nor the negative affective responses to that information were associated with participants’ subsequent pro-environmental intentions. In Study 2 (n = 135), we employed a manipulation-of-mediator design, which was a more stringent test of mediation. We asked people to either intensify or suppress their feelings while they were exposed to information about climate change or information unrelated to climate change. Deepening one’s feelings increased negative affect and reduced positive affect but did not significantly raise pro-environmental intentions. Together, these results suggest that climate change information reliably produces affective responses, but these do not translate into pro-environmental intentions in the short term. Full article

Attention can be automatically captured by the distractor that matches the representation of working memory (WM) in search tasks, impairing visual search efficiency and resulting in attentional capture effects. The resource hypothesis of visual search predicts that resource allocation affects attentional capture. However, previous studies have shown partly opposing results inconsistent with this prediction. The purpose of this study is to assess the connection between attentional capture and WM resource allocation. Two experiments were conducted to combine the attentional capture paradigm with continuous delayed-estimation tasks. In Experiment 1, we manipulated the number of memory items between one and two and measured the WM representation precision as well as the magnitude of attentional capture. In Experiment 2, we manipulated resource allocation using a retro-cue task with the presentation of two memory items. In Experiment 1, the results show that when remembering one item, a single-item representation had higher precision compared to the scenario for remembering two items, and it also involved a greater allocation of WM resources. However, there was no significant difference in the magnitude of attentional capture effects between the two conditions. In Experiment 2, the results show that memory precision was higher when the cue pointed to the item compared to when it did not, but there was no significant difference in the magnitude of attentional capture effects between the cued-match and non-cued-match conditions. The findings show that the size of attentional capture effects based on WM is unaffected by the distribution of WM resources. Attentional capture effects may reflect the attention bias of WM representation that occurs in preparation stage of memory-based attentional guidance. Full article
