Even for the strongest human being, maintaining an elevated arm position for an extended duration represents a significant challenge, as fatigue inevitably accumulates over time. The physical strain is further intensified when the individual is engaged in repetitive tasks, particularly those involving the use of tools or heavy equipment. Such activities increase the probability of developing muscle fatigue or injuries due to overuse or improper posture. Over time, this can result in the development of chronic conditions, which may impair the individual’s ability to perform tasks effectively and potentially lead to long-term physical impairment. Exoskeletons play a transformative role by reducing the perceived load on the muscles and providing mechanical support, mitigating the risk of injuries and alleviating the physical burden associated with strenuous activities. In addition to injury prevention, these devices also promise to facilitate the rehabilitation of individuals who have sustained musculoskeletal injuries. This document examines the various types of exoskeletons, investigating their design, functionality, and applications. The objective of this study is to present a comprehensive understanding of the current state of these devices, highlighting advancements in the field and evaluating their real-world impact. Furthermore, it analyzes the crucial insights obtained by other researchers, and by summarizing these findings, this work aims to contribute to the ongoing efforts to enhance exoskeleton performance and expand their accessibility across different sectors, including agriculture, healthcare, industrial work, and beyond. Electromyography (EMG) has emerged as a vital tool in the development of wearable robotic exoskeletons, enabling intuitive and responsive control by capturing neuromuscular signals. This review presents a comprehensive analysis of the EMG signal processing pipeline tailored to exoskeleton applications, spanning signal acquisition, noise mitigation, data preprocessing, feature extraction, and control strategies. Various EMG acquisition methods, including surface, intramuscular, and high-density surface EMG, are evaluated for their applicability in real-time control. The review addresses prevalent signal quality challenges, such as motion artifacts, power-line interference, and crosstalk. It also highlights both traditional filtering techniques and advanced methods, such as wavelet transforms, empirical mode decomposition, and adaptive filtering. Feature extraction techniques are explored to support pattern recognition and motion classification. Machine learning approaches are examined for their roles in pattern recognition-based and hybrid control architectures. This article emphasizes muscle synergy analysis and adaptive control algorithms to enhance personalization and fatigue compensation, followed by the benefits of multimodal sensing and edge computing in addressing the limitations of EMG-only systems. By focusing on EMG-driven strategies through signal processing, machine learning, and sensor fusion innovations, this review bridges gaps in human–machine interaction, offering insights into improving the precision, adaptability, and robustness of next generation exoskeletons. Advances in information technology and the widespread adoption of smartphones have improved human convenience and quality of life by facilitating extensive information sharing. However, the increasing frequency and duration of smartphone use is linked to a high risk of musculoskeletal disorders, particularly manifesting as changes in posture and gait. These alterations can lead to various physical issues, including spinal deformities, reduced gait stability, and increased muscle fatigue. Furthermore, excessive smartphone use can negatively affect mental health, contributing to depression, anxiety, and cognitive impairment. This narrative review primarily aims to systematically examine the effects of smartphone-related posture and gait alterations on physical function and identify associated problems. This study systematically summarized individual studies published between 2009, when smartphones first became widespread, and 2024 that investigated the effects of smartphone-induced posture and gait alterations. Through identifying issues related to these alterations, we aim to propose preventive strategies to avoid further complications. Full article

Objectives: In healthy young adults, thoracic kyphosis can be attributed to a number of factors, including a sedentary lifestyle, stress, poor posture, activity and daily habits, muscle pain, fatigue, and anxiety. In regard to clinical diagnosis and evaluation methods, high-cost radiological measurements and a variety of non-radiological clinical methods are employed. In this study, a decision support system that performs automatic thoracic kyphosis angle measurements has been developed with the objective of avoiding exposure of the human body to radiation and reducing the time required for measurements. Methods: The features were determined with reference to the thoracic kyphosis measurements that were manually marked by the expert on the subjects. The kyphosis angle was calculated by automatically identifying the T1 and T12 points through image segmentation using a convolutional neural network (CNN), which is a type of deep learning algorithm. Results: Intra-class consistency of ICC > 0.95 (p < 0.05) and internal consistency reliability of Cronbach’s α = 0.947 are obtained. Conclusions: The results demonstrate that the proposed algorithm exhibits high intra-class consistency and high internal consistency reliability to provide an automated thoracic kyphosis angle measurement system. In recent years, an increasing number of studies have been carried out in the field of skeletal muscle health and sports nutrition. Selenium functions in the human body through selenoproteins. Selenoproteins play an important role in maintaining skeletal muscle function by delaying exercise fatigue and muscle aging. They mainly regulate skeletal muscle by anti-oxidation, regulating signal pathways, and affecting protein metabolism. Passive back-support exoskeletons commonly employ elastic components to assist users during dynamic tasks. However, these designs are ineffective in providing sustained assistance for prolonged static bending postures, such as those required in surgery, assembly, and farming, where users experience continuous lumbar flexion. To address this limitation, a novel passive back-support exoskeleton inspired by the human spine is proposed in this work. The exoskeleton integrates a five-bar linkage mechanism with vertebrae-mimicking units, allowing for both dynamic flexion–extension movements and rigid support at various flexion angles. During the experiments, subjects are instructed to perform a 30-min forward-bending assembly task under two conditions: with and without wearing the exoskeleton. Compared to the free condition, the electromyography results indicate a 10.1% reduction in integrated EMG (IEMG) and a 9.78% decrease in root mean square (RMS) values of the erector spinae with the exoskeleton. Meanwhile, the metabolic rate is decreased by 11.1%, highlighting the effectiveness of the exoskeleton in mitigating muscle fatigue during prolonged static work. This work provides a promising solution for reducing musculoskeletal strain in occupations requiring sustained forward bending, making it a valuable advancement in passive exoskeleton technology. To address lower limb fatigue in workers engaged in prolonged standing, this study proposes a structural design for a medial-support passive exoskeleton seat. The design incorporates support rods positioned along the medial aspect of the user’s lower limbs and features an adaptive telescopic rod system, enhancing sitting stability and reducing collision risks in workplace environments. Human motion capture technology was used to collect kinematic data of the lower limbs, and a mathematical model of center-of-gravity variation was developed to calculate and optimize the exoskeleton’s structural parameters. Static analysis was performed using ANSYS software (2025 R1) to evaluate the structural integrity of the design. The effectiveness of the exoskeleton seat was validated through surface electromyography (sEMG) experiments, with results showing that the exoskeleton significantly reduces lower limb muscle load by 49.2% to 72.9%. Additionally, force plate experiments demonstrated that the exoskeleton seat improves stability, with a 39.2% reduction in the average displacement of the center of pressure (CoP), confirming its superior postural alignment and balance. The design was also compared with existing exoskeleton chairs, showing comparable or better performance in terms of muscle load reduction, stability, and overall effectiveness. Full article

Industrial workers often engage in repetitive lifting tasks. This type of continual loading on their arms throughout the workday can lead to muscle or tendon injuries. A non-intrusive system designed to assist a worker’s arms would help alleviate strain on their muscles, thereby preventing injury and minimizing productivity losses. The goal of this project is to develop a wearable soft robotic arm enhancement device that supports a worker’s muscles by sharing the load during lifting tasks, thereby increasing their lifting capacity, reducing fatigue, and improving their endurance to help prevent injury. The device should be easy to use and wear, functioning in relative harmony with the user’s own muscles. It should not restrict the user’s range of motion or flexibility. The human arm consists of numerous muscles that work together to enable its movement. However, as a proof of concept, this project focuses on developing a prototype to enhance the biceps brachii muscle, the primary muscle involved in pulling movements during lifting. Key components of the prototype include a soft robotic muscle or actuator analogous to the biceps, a control system for the pneumatic muscle actuator, and a method for securing the soft muscle to the user’s arm. The McKibben-inspired pneumatic muscle was chosen as the soft actuator for the prototype. A hybrid control algorithm, incorporating PID and model-based control methods, was developed. Electromyography (EMG) and pressure sensors were utilized as inputs for the control algorithms. This paper discusses the design strategies for the device and the preliminary results of the feasibility testing. Based on the results, a wearable EMG-controlled soft robotic arm augmentation could effectively enhance the endurance of industrial workers engaged in repetitive lifting tasks. Full article

Prolonged and high-intensity human–robot interaction can cause muscle fatigue. This fatigue leads to changes in both the time domain and frequency domain of the surface electromyography (sEMG) signals, which are closely related to human body movements. Consequently, these changes affect the accuracy and stability of using sEMG signals to recognize human body movements. Although numerous studies have confirmed that the median frequency of sEMG signals decreases as the degree of muscle fatigue increases—and this has been used for classifying fatigue and non-fatigue states— there is still a lack of quantitative characterization of the degree of muscle fatigue. Therefore, this paper proposes a method for quantitatively characterizing the degree of muscle fatigue during periodic exercise, based on the high-frequency components obtained through ensemble empirical mode decomposition (EEMD). Firstly, the sEMG signals of the estimated individuals are subjected to EEMD to obtain the high-frequency components, and the short-time Fourier transform is used to calculate the median frequency (MF) of these high-frequency components. Secondly, the obtained median frequencies are linearly fitted, and based on this, a standardized median frequency distribution range (SMFDR) of sEMG signals under muscle fatigue is established. Finally, a muscle fatigue estimator is proposed to achieve the quantification of the degree of muscle fatigue based on the SMFDR. Experimental validation across five subjects demonstrated that this method effectively quantifies cyclical muscle fatigue, with results revealing the methodology exhibits superiority in identifying multiple fatigue states during cyclical movements under consistent loading conditions. Teleoperation enables robots to perform tasks in dangerous or hard-to-reach environments on behalf of humans, but most methods lack operator immersion and compliance during grasping. To significantly enhance the operator’s sense of immersion and achieve more compliant and adaptive grasping of objects, we introduce a novel teleoperation method for dexterous robotic hands. This method integrates finger-to-finger force and vibrotactile feedback based on the Fuzzy Logic-Dynamic Compliant Primitives (FL-DCP) controller. It employs fuzzy logic theory to identify the stiffness of the object being grasped, facilitating more effective manipulation during teleoperated tasks. Utilizing Dynamic Compliant Primitives, the robotic hand implements adaptive impedance control in torque mode based on stiffness identification. Then the immersive bilateral teleoperation system integrates finger-to-finger force and vibrotactile feedback, with real-time force information from the robotic hand continuously transmitted back to the operator to enhance situational awareness and operational judgment. This bidirectional feedback loop increases the success rate of teleoperation and reduces operator fatigue, improving overall performance. Experimental results show that this bio-inspired method outperforms existing approaches in compliance and adaptability during teleoperation grasping tasks. This method mirrors how human naturally modulate muscle stiffness when interacting with different objects, integrating human-like decision-making and precise robotic control to advance teleoperated systems and pave the way for broader applications in remote environments. This study explores the integration of a custom-designed pneumatic spring into a car-seat cushion and its interaction with a simplified human body model using the Finite Element Method (FEM). A 3D half-symmetry FEM framework, developed from experimental data, ensured computational efficiency and convergence. This research bridged experimental and numerical approaches by analyzing the contact pressure distributions between a seat cushion and a volunteer with representative biometric characteristics. The model incorporated two material groups: (1) human body components (bones and muscles) and (2) seat cushion materials (polyurethane foam, latex, and fabric tape). Mechanical properties were obtained from both the literature and experiments, and simulations were conducted using MSC.Marc software under realistic boundary and initial conditions. The simulation results exhibited strong agreement with experimental data, validating the model’s reliability in predicting contact pressure distribution and optimizing seat cushion designs. Contrary to the conventional notion that uniformly distributed contact pressure inherently enhances comfort, this study emphasizes that the precise localization of pressure plays a crucial role in static and long-term seating ergonomics. Both experimental and simulation results demonstrated that modulating the pneumatic spring’s internal pressure from 0 kPa to 25 kPa altered peak contact pressure by approximately 3.5 kPa (around 20%), significantly influencing pressure redistribution and mitigating high-pressure zones. By validating this FEM-based approach, this study reduces dependence on physical prototyping, lowering design costs, and accelerating the development of ergonomically optimized seating solutions. The findings contribute to a deeper understanding of human–seat interactions, offering a foundation for next-generation automotive seating innovations that enhance comfort, fatigue reduction, and adaptive pressure control. Histidine is an amino acid which plays a critical role in protein synthesis, muscle buffering during anaerobic exercise, and antioxidation. It also acts as a precursor to carnosine, a dipeptide that enhances physical performance by being present in fast-contracting muscle fibers and contributing to buffering capacity. Recent studies have examined other histidine-containing compounds, such as anserine, balenine, and ergothioneine, to assess their potential benefits for physical activity. This narrative review focuses on the literature about the effects of dietary supplementation with these histidine-containing compounds on exercise capacity in animals and humans. The findings indicate that anserine may improve physical performance and reduce fatigue, particularly in quick, repetitive activities. Although balenine has been less extensively studied, it has shown promise in enhancing muscle regeneration and antioxidative defense in animal models. Ergothioneine, a sulfur-containing histidine derivative, displayed antioxidant and anti-inflammatory properties in both animal and human studies, suggesting its potential role in reducing exercise-induced oxidative stress and aiding recovery. The diversity of the presented studies and their limitations do not provide an opportunity to confirm the ergogenic properties of the histidine-containing compounds studied. Nevertheless, supplementation with anserine and ergothioneine shows promise for enhancing physical performance and recovery, though further research is required to better understand their mechanisms and optimize their use in sports and exercise. Background: The nuclear-encoded enzyme polymerase gamma (Pol-γ) is crucial in the replication of the mitochondrial genome (mtDNA), which in turn is vital for mitochondria and hence numerous metabolic processes and energy production in eukaryotic cells. Variants in the POLG gene, which encodes the catalytic subunit of Pol-γ, can significantly impair Pol-γ enzyme function. Pol-γ-associated disorders are referred to as POLG-spectrum disorders (POLG-SDs) and are mainly autosomal-recessively inherited. Clinical manifestations include muscle weakness and fatigue, and severe forms of the disease can lead to premature death in infancy, childhood, and early adulthood, often associated with seizures, liver failure, or intractable epilepsy. Here, we analyzed fibroblasts from a compound heterozygous patient with the established pathogenic variant c.2419C>T; p.(Arg807Cys) and a previously undescribed variant c.678G>C; p.(Gln226His) with a clinical manifestation compatible with POLG-SDs, sensory ataxic neuropathy, and infantile muscular atrophy. We conducted a battery of functional studies for Pol-γ and mitochondrial dysfunction on the patient’s fibroblasts, to test whether the novel variant c.678G>C; p.(Gln226His) may be causative in human disease. Aims/Methods: We analyzed skin-derived fibroblasts in comparison to a first-degree relative (the mother of the patient), an asymptomatic carrier harboring only the established c.2419C>T; p.(Arg807Cys) mutation. Assessments of mitochondrial function included measurements of mtDNA content, mRNA levels of mitochondrial genes, mitochondrial mass, and mitochondrial morphology. Case Presentation and Results: A 13-year-old male presented with symptoms starting at three years of age, including muscle weakness and atrophy in the lower extremities and facial muscles, which later extended to the upper limbs, voice, and back muscles, without further progression. The patient also reported fatigue and muscle pain after physical activity, with no sensory deficits. Extensive diagnostic tests such as electromyography, nerve conduction studies, muscle biopsy, and MRI were unremarkable. Exome sequencing revealed that he carried the compound heterozygous variants in POLG c.678G>C; p.(Gln226His) and c.2419C>T; p.(Arg807Cys), but no other potential genetic pathogenic causes. In comparison to a first-degree relative (his mother) who only carried the c.2419C>T; p.(Arg807Cys) pathogenic mutation, in vitro analyses revealed a significant reduction in mtDNA content (~50%) and mRNA levels of mtDNA-encoded proteins. Mitochondrial mass was reduced by approximately 20%, and mitochondrial interconnectivity within cells was impaired, as determined by fluorescence microscopy and mitochondrial staining. Conclusions: Our findings suggest that the c.678G>C; p.(Gln226His) variant, in conjunction with the c.2419C>T; p.(Arg807Cys) mutation, may compromise mtDNA replication and mitochondrial function and could result in clinically significant mitochondriopathy. As this study is based on one patient compared to a first-degree relative (but with an identical mitochondrial genome), the pathogenicity of c.678G>C; p.(Gln226His) of POLG should be confirmed in future studies, in particular, in conjunction with other POLG-variants. Background/Objectives: The excitability of skeletal muscle is a less-known post-mortem supravital phenomenon in human bodies, and it can be used to estimate the post-mortem interval. We conducted a field study in the Netherlands to investigate the applicability of muscle excitability (SMR) by mechanical stimulation for estimating the post-mortem interval in daily forensic practice. Knowledge concerning the post-mortem cell mechanisms accounting for the post-mortem excitability of skeletal muscle is lacking. Cell mechanisms are the specific intracellular and biochemical processes responsible for post-mortem muscle excitability. Methods: We have studied the theoretical backgrounds of the cell mechanisms that might be responsible for post-mortem muscle excitability, by performing literature research via the databank PubMed. Results: Based on the current available literature, in our opinion the intracellular changes in muscle cells that are responsible for SMR resemble the intracellular processes responsible for muscle fatigue due to energy exhaustion in the living. Conclusions: We hypothesize two pathways, depending on the level of energy in the muscle cell, that could be responsible for post-mortem muscle excitability by mechanical stimulation. Full article

In recent years, the field of wearable sensors has undergone significant evolution, emerging as a pivotal topic of research due to the capacity of such sensors to gather physiological data during various human activities. Transitioning from basic fitness trackers, these sensors are continuously being improved, with the ultimate objective to make compact, sophisticated, highly integrated, and adaptable multi-functional devices that seamlessly connect to clothing or the body, and continuously monitor bodily signals without impeding the wearer’s comfort or well-being. Potentiometric sensors, leveraging a range of different solid contact materials, have emerged as a preferred choice for wearable chemical or biological sensors. Nanomaterials play a pivotal role, offering unique properties, such as high conductivity and surface-to-volume ratios. This article provides a review of recent advancements in wearable potentiometric sensors utilizing various solid contacts, with a particular emphasis on nanomaterials. These sensors are employed for precise ion concentration determinations, notably sodium, potassium, calcium, magnesium, ammonium, and chloride, in human biological fluids. This review highlights two primary applications, that is, (1) the enhancement of athletic performance by continuous monitoring of ion levels in sweat to gauge the athlete’s health status, and (2) the facilitation of clinical diagnosis and preventive healthcare by monitoring the health status of patients, in particular to detect early signs of dehydration, fatigue, and muscle spasms.
