Parkinson’s disease (PD) is the most common age-related movement disorder characterized by the progressive loss of nigrostriatal dopaminergic neurons. To date, PD treatment strategies are mostly based on dopamine replacement medicines, which can alleviate motor symptoms but do not slow down the progression of neurodegeneration. Thus, there is a need for disease-modifying PD therapies. The aim of this work was to evaluate the neuroprotective effects of the novel compound PA96 on dopamine neurons in vivo and in vitro, assess its ability to alleviate motor deficits in MPTP- and haloperidol-based PD models, as well as PK profile and BBB penetration. PA96 was synthesized from (1R,2R,6S)-3-methyl-6-(prop-1-en-2-yl) cyclohex-3-ene-1,2-diol (Prottremin) using the original three-step stereoselective procedure. We found that PA96: (1) supported the survival of cultured näive dopamine neurons; (2) supported the survival of MPP+-challenged dopamine neurons in vitro and in vivo; (3) had chemically appropriate properties (synthesis, solubility, etc.); (4) alleviated motor deficits in MPTP- and haloperidol-based models of PD; (5) penetrated the blood–brain barrier in vivo; and (6) was eliminated from the bloodstream relative rapidly. In conclusion, the present article demonstrates the identification of PA96 as a lead compound for the future development of this compound into a clinically used drug. Full article

Formation of aerosol from biogenic hydrocarbons relies heavily on anthropogenic emissions since they control the availability of species such as sulfate and nitrate, and through them, aerosol acidity (pH). To elucidate the role that acidity and emissions play in regulating Secondary Organic Aerosol (SOA), we utilize the 2013 Southern Oxidant and Aerosol Study (SOAS) dataset to enhance the extensive mechanism of isoprene epoxydiol (IEPOX)-mediated SOA formation implemented in the Community Multiscale Air Quality (CMAQ) model (Pye et al., 2013), which was then used to investigate the impact of potential future emission controls on IEPOX OA. We found that the Henry’s law coefficient for IEPOX was the most impactful parameter that controls aqueous isoprene OA products, and a value of 1.9 × 107 M atm−1 provides the best agreement with measurements. Non-volatile cations (NVCs) were found in higher-than-expected quantities in CMAQ and exerted a significant influence on IEPOX OA by reducing its production by as much as 30% when present. Consistent with previous literature, a strong correlation of isoprene OA with sulfate, and little correlation with acidity or liquid water content, was found. Future reductions in SO2 emissions are found to not affect this correlation and generally act to increase the sensitivity of IEPOX OA to sulfate, even in extreme cases. Full article

Chemical processing of organic material in aqueous atmospheric aerosols and cloudwater is known to form secondary organic aerosols (SOA), although the extent to which each of these processes contributes to total aerosol mass is unclear. In this study, we use GAMMA 5.0, a photochemical box model with coupled gas and aqueous-phase chemistry, to consider the impact of aqueous organic reactions in both aqueous aerosols and clouds on isoprene epoxydiol (IEPOX) SOA over a range of pH for both aqueous phases, including cycling between cloud and aerosol within a single simulation. Low pH aqueous aerosol, in the absence of organic coatings or other morphology which may limit uptake of IEPOX, is found to be an efficient source of IEPOX SOA, consistent with previous work. Cloudwater at pH 4 or lower is also found to be a potentially significant source of IEPOX SOA. This phenomenon is primarily attributed to the relatively high uptake of IEPOX to clouds as a result of higher water content in clouds as compared with aerosol. For more acidic cloudwater, the aqueous organic material is comprised primarily of IEPOX SOA and lower-volatility organic acids. Both cloudwater pH and the time of day or sequence of aerosol-to-cloud or cloud-to-aerosol transitions impacted final aqueous SOA mass and composition in the simulations. The potential significance of cloud processing as a contributor to IEPOX SOA production could account for discrepancies between predicted IEPOX SOA mass from atmospheric models and measured ambient IEPOX SOA mass, or observations of IEPOX SOA in locations where mass transfer limitations are expected in aerosol particles.
