Background: Against the backdrop of the large-scale and intensive development of the livestock industry, enhancing the reproductive efficiency of cattle has become a crucial factor in industrial development. Holstein cows, as the most predominant dairy cattle breed globally, are characterized by high milk yield and excellent milk quality. However, their reproductive efficiency is comprehensively influenced by a variety of complex factors, and improving their reproductive performance faces numerous challenges. The ovary, as the core organ of the female reproductive system, plays a decisive role in embryonic development and pregnancy maintenance. It is not only the site where eggs are produced and developed but it also regulates the cow’s estrous cycle, ovulation process, and the establishment and maintenance of pregnancy by secreting various hormones. The normal functioning of the ovary is crucial for the smooth development of the embryo and the successful maintenance of pregnancy. Methods: Currently, traditional sequencing technologies have obvious limitations in deciphering ovarian function and reproductive regulatory mechanisms. To overcome the bottlenecks of traditional sequencing technologies, this study selected Holstein cows as the research subjects. Ovarian samples were collected from one pregnant and one non-pregnant Holstein cow, and single-nucleus transcriptome sequencing technology was used to conduct an in-depth study on the ovarian cells of Holstein cows. Results: By constructing a cell type-specific molecular atlas of the ovaries, nine different cell types were successfully identified. This study compared the proportions of ovarian cell types under different physiological states and found that the proportion of endothelial cells decreased during pregnancy, while the proportions of granulosa cells and luteal cells increased significantly. In terms of functional enrichment analysis, oocytes during both pregnancy and non-pregnancy play roles in the “cell cycle” and “homologous recombination” pathways. However, non-pregnant oocytes are also involved in the “progesterone-mediated oocyte maturation” pathway. Luteal cells during pregnancy mainly function in the “cortisol synthesis and secretion” and “ovarian steroidogenesis” pathways; non-pregnant luteal cells are mainly enriched in pathway processes such as the “AMPK signaling pathway”, “pyrimidine metabolism”, and “nucleotide metabolism”. Cell communication analysis reveals that there are 51 signaling pathways involved in the pregnant ovary, with endothelial cells, granulosa cells, and luteal cells serving as the core communication hubs. In the non-pregnant ovary, there are 48 pathways, and the interaction between endothelial cells and stromal cells is the dominant mode. Conclusions: This study provides new insights into the regulatory mechanisms of reproductive efficiency in Holstein cows. The differences in the proportions of ovarian cell types, functional pathways, and cell communication patterns under different physiological states, especially the increase in the proportions of granulosa cells and luteal cells during pregnancy and the specificity of related functional pathways, indicate that these cells play a crucial role in the reproductive process of cows. These findings also highlight the importance of ovarian cells in pathways such as “cell cycle”, “homologous recombination”, and “progesterone-mediated oocyte maturation”, as well as the cell communication mechanisms in regulating ovarian function and reproductive performance. The Effect of cdk1 Gene Knockout on Heat Shock-Induced Polyploidization in Loach (Misgurnus anguillicaudatus)

(1) Background: Polyploid fish are highly important in increasing fish production, improving fish quality, and breeding new varieties. The loach (Misgurnus anguillicaudatus), as a naturally polyploid fish, serves as an ideal biological model for investigating the mechanisms of chromosome doubling; (2) Methods: In this study, tetraploidization in diploid loach was induced by heat shock treatment, and, for the first time, the role of the key cell cycle gene cdk1 (cyclin-dependent kinase 1) in chromosome doubling was investigated; (3) Results: The experimental results show that when eggs are fertilized for 20 min and then subjected to a 4 min heat shock treatment at 39–40 °C, this represents the optimal induction condition, resulting in a tetraploid rate of 44%. Meanwhile, the results of the cdk1 knockout model (2n cdk1−/−) constructed using CRISPR/Cas9 showed that the absence of cdk1 significantly increased the chromosome doubling efficiency of the loach. The qPCR analysis revealed that knockout of cdk1 significantly upregulated cyclin genes (ccnb3,ccnc, and ccne1), while inhibiting expression of the separase gene espl1 (p < 0.05); (4) Conclusions: During chromosome doubling in diploid loaches induced by heat shock, knocking out the cdk1 gene can increase the tetraploid induction rate. This effect may occur through downregulation of the espl1 gene. This study offers novel insights into optimizing the induced breeding technology of polyploid fish and deciphering its molecular mechanism, while highlighting the potential application of integrating gene editing with physical induction. Over the years, macromycete fungi have been used as a source of food, part of religious rites and rituals, and as a medicinal remedy. Species with strong health-promoting potential include Hericium erinaceus, Cordyceps militaris, Ganoderma lucidum, Pleurotus ostreatus, Flammulina velutipes, and Inonotus obliquus. These species contain many bioactive compounds, including β-glucans, endo- and exogenous amino acids, polyphenols, terpenoids, sterols, B vitamins, minerals, and lovastatin. The level of some biologically active substances is species-specific, e.g., hericenones and erinacines, which have neuroprotective properties, and supporting the production of nerve growth factor in the brain for Hericium erinaceus. Due to their high health-promoting potential, mushrooms and substances isolated from them have found applications in livestock nutrition, improving their welfare and productivity. This phenomenon may be of particular importance in the nutrition of laying hens and broiler chickens, where an increase in pathogen resistance to antibiotics has been observed in recent years. Gallus gallus domesticus is a key farm animal for meat and egg production, so the search for new compounds to support bird health is important for food safety. Studies conducted to date indicate that feed supplementation with mushrooms has a beneficial effect on, among other things, bird weight gain; bone mineralisation; and meat and egg quality, including the lipid profile and protein content and shell thickness, and promotes the development of beneficial microbiota, thereby increasing immunity. Drosophila suzukii is an invasive pest of many fruit crops worldwide. Employing the Sterile Insect Technique (SIT) could mitigate D. suzukii population growth and crop damage. This study evaluated the efficacy of SIT on commercial fruit, by (1) validating the quality of irradiated sterile males (male mating competitiveness, courtship, and flight performance) in the laboratory, and (2) assessing population suppression and fruit damage reduction in commercial raspberry fields. Treatment with SIT was compared to the grower’s standard chemical insecticide program throughout the season. The principal metrics of efficacy were trap counts of wild adult female D. suzukii in crops and larvae per fruit during harvesting. These metrics together with monitoring of border areas allowed targeting of high-pressure areas with higher releases of sterile males, to maximise efficacy for a given release number. The sterile male D. suzukii were as competitive as their fertile non-irradiated counterparts in laboratory mating competitiveness and flight performance studies while fertility egg-to-pupae recovery was reduced by 99%. In commercial raspberry crops, season-long releases of sterile males significantly suppressed the wild D. suzukii population, compared to the grower standard control strategy; with up to 89% reduction in wild female D. suzukii and 80% decrease in numbers of larvae per harvested fruit. Additionally, relative fruit waste (i.e., percentage of harvested fruits rejected for sale) at harvest was reduced for early, mid and late harvest crops, by up to 58% compared to the grower standard control. SIT has the potential to provide an effective and sustainable strategy for managing D. suzukii in raspberries, increasing marketable yield by reducing adult populations, fruit damage and waste fruit. SIT could therefore serve as a valuable tool for integrated pest management practices in berry production systems. Full article

This study evaluated the production performance and egg quality of Lohmann Brown laying hens fed diets containing different levels of Corn Bran with Solubles (CBS). A total of 144 hens aged 44 weeks were assigned to three treatments in a completely randomized design, with eight replicates per treatment and six birds per replicate. The experimental treatments included diets with CBS inclusion levels of 0%, 5%, and 10%. The experiment lasted 84 days (44 to 55 weeks of age). Data were analyzed via one-way ANOVA, with mean differences evaluated using Tukey’s HSD test (α = 0.05). No significant effects were observed for laying rate, feed intake, feed conversion ratio, or egg mass (p > 0.05). However, egg quality parameters such as shell percentage, shell weight per unit surface area (SWUSA), and yolk color were influenced by the treatments (p < 0.05). Hens fed diets with 5% CBS exhibited higher shell percentage and SWUSA compared to those on the 0% CBS diet. Yolk color intensity increased with higher CBS inclusion levels. In conclusion, incorporating up to 10% CBS in corn–soybean meal diets for laying hens can enhance egg yolk pigmentation. Notably, including 5% CBS improves eggshell quality. Full article

The rising demand for sustainable protein is driving interest in insects as a raw material for advanced food ingredients. This review collates and critically analyses over 300 studies on the conversion of crickets, mealworms, black soldier flies, and other farmed species into powders, protein isolates, oils, and chitosan-rich fibers with targeted techno-functional roles. This survey maps how thermal pre-treatments, blanch–dry–mill routes, enzymatic hydrolysis, and isoelectric solubilization–precipitation preserve or enhance the water- and oil-holding capacity, emulsification, foaming, and gelation, while also mitigating off-flavors, allergenicity, and microbial risks. A meta-analysis shows insect flours can absorb up to 3.2 g of water g−1, stabilize oil-in-water emulsions for 14 days at 4 °C, and form gels with 180 kPa strength, outperforming or matching eggs, soy, or whey in specific applications. Case studies demonstrate a successful incorporation at 5–15% into bakery, meat analogs and dairy alternatives without sensory penalties, and chitin-derived chitosan films extend the bread shelf life by three days. Comparative life-cycle data indicate 45–80% lower greenhouse gas emissions and land use than equivalent animal-derived ingredients. Collectively, the evidence positions insect-based ingredients as versatile, safe, and climate-smart tools to enhance food quality and sustainability, while outlining research gaps in allergen mitigation, consumer acceptance, and regulatory harmonization. This study aimed to investigate the effects of baicalin and citric acid on egg production performance, egg quality, and the intestinal morphology and function of laying hens. A total of 600 Hy-Line Brown laying hens, 59 weeks old, were randomly allocated to four dietary treatments, with 10 replicates per treatment and 15 hens per replicate. The control group was fed a basal diet, while the other three groups were fed the basal diet supplemented with 150 mg/kg baicalin (B), 2000 mg/kg citric acid (CA), or 150 mg/kg baicalin plus 2000 mg/kg citric acid (B + CA), respectively. The experimental period lasted for 12 weeks, and the results indicated that neither the individual addition nor the combined application of baicalin and citric acid had a significant impact on the laying performance. However, compared with the control group, the baicalin and/or citric acid supplementation significantly increased the eggshell strength and Haugh unit. Additionally, the combination of baicalin and citric acid significantly increased the villus height and the villus height/crypt depth ratio in the duodenum and jejunum. It also enhanced the population of beneficial bacteria, such as Lactobacillus and Bifidobacterium, in the cecum and improved the activity of intestinal digestive enzymes, primarily disaccharidases. Furthermore, the addition of baicalin to the diet significantly increased the content of Secretory Immunoglobulin A in the ileum and jejunum after 12 weeks of feeding. These results suggest that the combination of baicalin and citric acid had a synergistic effect on the improvement of egg quality and intestinal morphology and function in laying hens. Overall, our findings provide important insights into the potential benefits of supplementing baicalin and citric acid in the diet of laying hens and may have practical implications for improving egg quality and poultry health status. The objective of this study was to develop a novel sauce formulation in which egg yolk was substituted with pea and soy proteins, in addition to the incorporation of tomato pomace as a functional ingredient. Nine experimental samples (E1–E3, S1–S3, and P1–P3) and three control samples (E0, S0, and P0) were prepared, corresponding to three protein sources (E: egg yolk, S: soy, P: pea), with increasing concentrations of tomato pomace (0, 2, 4, and 6%). The formulations were adjusted proportionally in terms of water and oil to maintain the desired consistency. The analyses performed included: physico-chemical analysis of the sauce (fat content, peroxide value, and CIE L* a* b* color determination), quality assessment using Fourier Transform Infrared Spectroscopy (FT-IR, rheological measurements, and microstructural evaluation. The sample designated P2 demonstrated a notable correlation with favourable parameters, exhibiting intense colouration, elevated protein content, and consistent rheological properties. However, at higher levels of tomato pomace (notably 6%), microstructural instability was observed, which may limit the formulation’s robustness over time. These findings demonstrate that tomato pomace can enhance the functional and structural characteristics of sauce, while also highlighting the importance of optimizing concentration levels to avoid negative impacts on emulsion stability. Overall, the results support the use of tomato pomace and plant proteins in the formulation of sustainable and innovative food products. Full article

Mycotoxins are toxic secondary metabolites produced by various fungi that contaminate livestock feed, posing serious threats to animal health, productivity, and food safety. Although historical research has often examined individual mycotoxins in isolation, real-world conditions typically involve the simultaneous presence of multiple mycotoxins, resulting in additive or synergistic toxic effects that are often more severe than those observed with single toxin exposures. This review comprehensively synthesizes recent findings on multi-mycotoxin contamination in livestock feed, highlighting their physiological effects, mechanisms of action, and implications for regulatory frameworks. Multi-mycotoxin interactions exacerbate oxidative stress, immune suppression, impaired reproduction, and organ damage across species, leading to reduced growth performance, decreased milk and egg production, compromised carcass and wool quality, and increased mortality rates. A major concern is that current international regulatory standards mainly address individual mycotoxins, overlooking the compounded risks of co-occurrence. Global surveillance studies consistently reveal high prevalence rates of mycotoxin mixtures in feedstuffs, especially combinations involving DON, ZEN, AFB1, FB1, and OTA. Understanding these interactions and their underlying cellular mechanisms is critical for improving risk assessment models, formulating integrated mitigation strategies, and safeguarding both livestock productivity and human food security. Food security is defined as a state in which all people at all times have both physical and economic access to sufficient food to meet their dietary needs for a productive and healthy life. The general objective of this work was to assess the situation of food insecurity and its impact on the quantity and quality of food consumption during lockdown in the first wave of the COVID-19 pandemic and to identify the determinants associated with the different food insecurity (FI) categories on a sample of 2227 people (1168 people from Spain and 1059 people from Morocco). Food insecurity (FI) assessed by the Household Food Insecurity Access Scale (HFIAS) were compared in both countries, controlling for the effect of sociodemographic variables, age, gender, marital status, and education level. The mean HFIAS was 0.53 in the Spanish and 3.55 in the Moroccan samples (p < 0.001). Only 2.1% of the Spanish sample were in a situation of severe insecurity against 15.5% in Morocco (p < 0.001). Moroccans with moderate and severe food insecurity decreased their consumption of meat, fish, eggs, nuts, legumes, and fruits. The risk of food insecurity was higher in men than in women, in separated or divorced people, in people with secondary and middle education, and in younger people. Full article

Camelina sativa (CS) is an emerging sustainable oilseed crop with interesting feed application potentialities. The research assessed the potentiality of Camelina sativa (CS) in the diet for free-range laying hens, aiming at reaching a nutritional claim. To this purpose, two feeding groups of hens (n = 100 Livorno hens, n = 25 hens/pen) were farmed outdoor and received either a Control diet, which was a commercial diet for laying hens, or a CS diet. The latter was formulated to include the 5% CS cake and 1% CS oil, replacing conventional feedstuffs. Diets were isoprotein and isoenergy and were available ad libitum throughout the laying period (February–September). At day 1, n = 12 eggs/diet were sampled to assess the initial proximate composition and FA profile. Every 7 days the sampling was repeated to analyze the eggs’ FA profile, up to day 35. At the end of the laying season, n = 6 hens/dietary treatment were slaughtered and subjected to meat quality evaluations. Results highlighted that a 28-day feeding was the period required to obtain 227 mg of C18:3 n-3 and 81 mg of C20:5 n-3 + C22:6 n-3/100 g egg, whereas a further 7 days of feeding trial were ineffective in further enhancing the omega-3 FA content of eggs. CS eggs were comparable to Control ones for overall physical traits, proximate composition, and shelf-life. In addition, at the end of the laying period, the meat of CS hens was also found to be healthier than that of Control ones, thanks to a higher omega-3 FA proportion (p < 0.01), which was, however, not sufficient to reach any nutritional claim. Instead, the proximate composition of CS meat was overall comparable to Control hens. In conclusion, the present research demonstrated that feeding Camelina sativa meal and oil to laying hens is feasible and allows to reach the nutritional claim in eggs “rich in omega-3 FA” after a feeding period of 28 days, without any negative effects on other eggs’ quality trials. Camelina sativa can thus be defined as a promising sustainable feedstuff for the poultry sector for diversification purposes and to enhance the nutritional quality of eggs. Full article

The conservation of poultry biodiversity is a growing global priority, yet it necessarily relies on the scientific valorization of specific local breeds. This study aimed to characterize the lipid composition and cholesterol content of eggs from three native Sicilian chicken breeds (Cornuta, Valplatani, and Siciliana) reared under semi-extensive conditions, in order to evaluate their nutritional potential and support biodiversity preservation strategies. A total of 170 eggs from 11 farms were analyzed. Fatty acid composition and nutritional indices (atherogenic index, thrombogenic index, n-6/n-3 ratio, HH index) were determined according to ISO and AOAC standards. Results showed that Cornuta eggs exhibited the most favorable lipid profile, with the lowest saturated fatty acid (SFA) content (38.55%), the lowest n-6/n-3 ratio (7.35), and the best values for AI (0.52), TI (1.22), and HH (2.02), compared to Valplatani and Siciliana. Conversely, the lowest cholesterol content was found in Siciliana eggs (1463.58 mg/kg), significantly lower than Cornuta (1789 mg/kg; p < 0.05). Although no commercial hybrids were included, the literature data were used for contextual comparison. These findings suggest that native breeds may produce eggs with functional nutritional properties, supporting both healthier food choices and local genetic conservation. Moreover, this study provides a replicable framework for the nutritional valorization of underutilized poultry breeds, reinforcing the role of biodiversity in sustainable food systems. Full article

Duck egg quality classification is critical in farms, hatcheries, and salted egg processing plants, where cracked eggs must be identified before further processing or distribution. However, duck eggs present a unique challenge due to their white eggshells, which make cracks difficult to detect visually. In current practice, human inspectors use standard white light for crack detection, and many researchers have focused primarily on improving detection algorithms without addressing lighting limitations. Therefore, this paper presents duck egg crack detection using an adaptive convolutional neural network (CNN) model ensemble with multi-light channels. We began by developing a portable crack detection system capable of controlling various light sources to determine the optimal lighting conditions for crack visibility. A total of 23,904 images were collected and evenly distributed across four lighting channels (red, green, blue, and white), with 1494 images per channel. The dataset was then split into 836 images for training, 209 images for validation, and 449 images for testing per lighting condition. To enhance image quality prior to model training, several image pre-processing techniques were applied, including normalization, histogram equalization (HE), and contrast-limited adaptive histogram equalization (CLAHE). The Adaptive MobileNetV2 was employed to evaluate the performance of crack detection under different lighting and pre-processing conditions. The results indicated that, under red lighting, the model achieved 100.00% accuracy, precision, recall, and F1-score across almost all pre-processing methods. Under green lighting, the highest accuracy of 99.80% was achieved using the image normalization method. For blue lighting, the model reached 100.00% accuracy with the HE method. Under white lighting, the highest accuracy of 99.83% was achieved using both the original and HE methods. Full article

Microbial contamination of hatching eggs often leads to reduced hatchability and poor chick quality. As trimethylamine (TMA), a metabolite derived from dietary choline, has antimicrobial properties, increasing yolk TMA contents may increase bacterial resistance to eggs; however, the effects of TMA concentrations on chick quality remain unknown. Hence, this study was conducted to determine the effects of yolk TMA concentrations on the hatchability and chick quality of dwarf hens with different FMO3 genotypes. Hens (n = 140) were divided into control and experimental groups; the latter received choline chloride (2800 mg/kg) to elevate their yolk TMA concentrations. The TMA content, Pasgar score, hatchability, and post-hatching performance were evaluated. The results showed that choline supplementation significantly increased TMA concentrations in hens with AT and TT genotypes. Higher yolk TMA concentrations (≥4 µg/g) correlated with improved Pasgar scores and reduced abnormalities in vitality, navel, and yolk sac absorption. Hatchability peaked at 6.49 µg/g TMA, suggesting a threshold effect. Although the growth rate remained unaffected, chick mortality decreased in the high-TMA group. Therefore, moderate TMA concentrations can enhance egg antimicrobial defenses and improve reproductive performance. This strategy provides a biologically grounded alternative to traditional chemical disinfection in hatcheries. Full article

Hemp (Cannabis sativa L.) products have gained attention in poultry nutrition for their rich content of polyunsaturated fatty acids (PUFAs), bioactive compounds, and potential functional benefits. However, findings on their impact on laying-hen performance, egg quality, and yolk fatty acid profiles have been inconsistent. This meta-analysis aimed to evaluate the effects of dietary hemp products on laying-hen performance, egg quality traits, and yolk fatty acid composition, while exploring potential sources of heterogeneity across studies. A comprehensive literature search identified 21 studies that met the inclusion criteria. A random-effects model was used to calculate standardized mean differences (SMDs) with 95% confidence intervals (CIs) for various outcomes, including production performance, egg quality, and yolk fatty acid profiles. Subgroup and meta-regression analyses assessed the influence of factors such as inclusion level, hen age, and hemp product type. The results showed that hemp supplementation had no significant effect on hen-day production, egg mass, feed conversion ratio, or feed intake. However, yolk redness (SMD = 4.40; 95% CI: 2.46, 6.33; p < 0.001) and yellowness (SMD = 4.45; 95% CI: 2.75, 6.16; p < 0.001) were significantly enhanced. Hemp feeding also increased n-3 PUFA levels in egg yolk, including C18:3n3, C20:5n3, and C22:6n3, while reducing saturated and monounsaturated fatty acids. Subgroup analysis indicated that inclusion levels > 10% and hen age ≤ 25 weeks were associated with slight reductions in hen-day production, whereas older hens (>25 weeks) showed increased egg weight. In conclusion, hemp products can enrich yolk pigmentation and n-3 PUFA content in eggs, supporting the production of functional, value-added eggs without compromising the laying-hen performance.
