The pivotal role of the basolateral amygdala (BLA) in the emotional modulation of hippocampal plasticity and memory consolidation is well-established. Specifically, multiple studies have demonstrated that the activation of the noradrenergic (NA) system within the BLA governs these modulatory effects. However, most current evidence has been obtained by direct infusion of synthetic NA or beta-adrenergic agonists. In the present study, we aimed to investigate the effect of endogenous NA release in the BLA, induced by a natural aversive stimulus (coyote urine), on memory consolidation for a low-arousing, hippocampal-dependent task. Our experiments combined a weak object location task (OLT) version with subsequent mild predator odor exposure (POE). To investigate the role of endogenous NA in the BLA in memory modulation, a subset of the animals (Wistar rats) was treated with the non-selective beta-blocker propranolol at the end of the behavioral procedures. Hippocampal tissue was collected 90 min after drug infusion or after the OLT test, which was performed 24 h later. We used the obtained samples to estimate the levels of phosphorylated CREB (pCREB) and activity-regulated cytoskeleton-associated protein (Arc)—two molecular markers of experience-dependent changes in neuronal activity. The result suggests that POE has the potential to become a valuable behavioral paradigm for studying the interaction between BLA and the hippocampus in memory prioritization and selectivity. It has been posited that ingesting a pill constitutes a pivotal action that facilitates the effects of open-label placebos (OLPs: placebos honestly prescribed). In the present OLP experiment, the motor components of a placebo treatment were systematically varied. The participants (n = 183) were randomly allocated to one of four groups that all viewed aversive pictures. The ‘active OLP’ group took a placebo pill with specific instructions concerning the sequence of motor actions for the intake. The ‘usual OLP’ group swallowed the pill (without specific motor instructions), while the third group received an ‘imaginary OLP’ (no pill intake). The fourth group applied cognitive reappraisal (CR; active control group) to reduce emotional distress. The participants rated their affective state as well as the efficacy and plausibility of the treatment approach. Moreover, blood pressure and pulse were recorded as indicators of bodily arousal. The four groups did not differ in their valence ratings and physiological measures. The ‘imaginary OLP’ received higher ratings for both effectiveness and plausibility than the ‘usual OLP’. CR was rated as superior relative to all OLP conditions. In conclusion, reducing emotional distress with OLPs does not necessitate the consumption of a placebo pill. In terms of acceptability and ease of implementation, CR stands as a well-established alternative. Full article

Insects are a conventional food in many cultures worldwide. Why therefore are they treated with aversion by the majority of people in Western countries? The aim of this work is to understand cultural norms and historical practices related to insects that can explain why in the countries of the West it is so difficult to accept them as a foodstuff, and why the disgust that they arouse is more common than in other regions of the world. Insects in Western countries are associated with negative experiences resulting from cultural and religious beliefs, traditions, stories, myths and individual experiences. Although there are species of insect that have positive associations, the high number of negative terms popularized the negative image of these creatures in people’s minds, as a result of which the concept of insects as a foodstuff arouses disgust in the culture of the West. Understanding the aversion to insects will contribute to the broader understanding of consumer attitudes, cultural influences on consumption, or potential shifts in food choices. It also can help develop strategies or methods that will assist in changing this reluctance and encourage the utilization of insects as a food source. Full article

To ensure the optimal living conditions of farm animals, it is essential to understand how their senses work and the way in which they perceive their environment. Most animals have a different hearing range compared to humans; thus, some aversive sounds may go unnoticed by caretakers. The auditory pathways may act through the nervous system on the cardiovascular, gastrointestinal, endocrine, and immune systems. Therefore, noise may lead to behavioral activation (arousal), pain, and sleep disorders. Sounds on farms may be produced by machines, humans, or animals themselves. It is worth noting that vocalization may be very informative to the breeder as it is an expression of an emotional state. This information can be highly beneficial in maintaining a high level of livestock welfare. Moreover, understanding learning theory, conditioning, and the potential benefits of certain sounds can guide the deliberate use of techniques in farm management to reduce the aversiveness of certain events. Given the COVID-19 pandemic and the resulting social distancing measures with inevitable telecommuting, capturing user emotions is essential as it affects both satisfaction and task performance. Therefore, the purpose of this study was to analyze emotions and task performance in terms of dislike and personalized decision-making in indoor spaces. To facilitate experiments with participants, a mixed-reality environment was utilized with the Pleasure, Arousal, Dominance (PAD) test and cognitive tests. The results of the experiment conducted on 30 subjects identified that aroused and discontented emotions dominated in non-preferred spaces, but pleased, important, and autonomous emotions arose in personalized spaces, as determined through sentimental analysis and statistical methods. Although negative emotions were present in the aversion space, attention and execution abilities were high compared to the personalized space, but working memory was low. By conducting stepwise regression analysis, it was found that working in a visually unfavorable space, which caused an increase in controlled or controlling emotions, improved short-term work efficiency. In addition, important emotions did not have a positive effect on any task performance. However, with pleased and contented emotions in a personalized indoor space, long-term work efficiency was increased, as explained by the Yerkes-Dodson law. Full article

Microsaccades are at the interface between basic oculomotor phenomena and complex processes of cognitive functioning, and they also have been a challenge for subtle experimentation and adequate statistical analysis. In the second part of the special thematic issue (for the first part see Martinez-Conde, Engbert, & Groner, 2020) the authors present a series of articles which demonstrate that microsaccades are still an interesting and rewarding area of scientific research the forefront of research in many areas of sensory, perceptual, and cognitive processes. In their article “Pupillary and microsaccadic responses to cognitive effort and emotional arousal during complex decision making” Krejtz, Żurawska, Duchowski, & Wichary (2020) investigate pupillary and microsaccadic responses to information processing during multi-attribute decision making under affective priming. The participants were randomly assigned into three affective priming conditions (neutral, aversive, and erotic) and instructed to make discriminative decisions. As hypothesized by the authors, the results showed microsaccadic rate inhibition and pupillary dilation, depending on cognitive effort prior to decision and moderated by affective priming. Aversive priming increased pupillary and microsaccadic responses to information processing effort. The results indicate that pupillary response is more influenced by affective priming than microsaccadic rate. The results are discussed in the light of neuropsychological mechanisms of pupillary and microsaccadic behavior. In the article “Microsaccadic rate signatures correlate under monocular and binocular stimulation conditions” Essig, Leube, Rifai, & Wahl (2020) investigate microsaccades with respect to their directional distribution and rate under monocular and binocular conditions. In both stimulation conditions participants fixated a Gabor patch presented randomly in orientation of 45° or 135° over a wide range of spatial frequencies. Microsaccades were mostly horizontally oriented regardless of the spatial frequency of the grating. This outcome was consistent between both stimulation conditions. This study found that the microsaccadic rate signature curve correlates between both stimulation conditions, therefore extending the use of microsaccades to clinical applications, since parameters as contrast sensitivity, have frequently been measured monocularly in the clinical studies. The study “Microsaccades during high speed continuous visual search” by Martin, Davis, Riesenhuber, & Thorpe (2020) provides an analysis of the microsaccades occurring during visual search, targeting to small faces pasted either into cluttered background photos or into a simple gray background. Participants were instructed to target singular 3-degree upright or inverted faces in changing scenes. As soon as the participant’s gaze reached the target face, a new face was displayed in a different random location. Regardless of the experimental context (e.g., background scene, no background scene), or target eccentricity (from 4 to 20 degrees of visual angle), The authors found that the microsaccade rate dropped to near zero levels within 12 ms. There were almost never any microsaccades after stimulus onset and before the first saccade to the face. In about 20% of the trials, there was a single microsaccade that occurred almost immediately after the preceding saccade’s offset. The authors argue that a single feedforward pass through the visual hierarchy of processing a stimulus is needed to effectuate prolonged continuous visual search and provide evidence that microsaccades can serve perceptual functions like correcting saccades or effectuating task-oriented goals during continuous visual search. While many studies have characterized the eye movements during visual fixation, including microsaccades, in most cases only horizontal and vertical components have been recorded and analyzed. Little is known about the torsional component of microsaccades. In the study “Torsional component of microsaccades during fixation and quick phases during optokinetic stimulation” Sadeghpour & Otero-Millan (2020) recorded eye movements around the three axes of rotation during fixation and torsional optokinetic stimulus. The authors found that the average amplitude of the torsional component of microsaccades during fixation was 0.34 ± 0.07 degrees with velocities following a main sequence with a slope comparable to the horizontal and vertical components. The size of the torsional displacement during microsaccades was correlated with the horizontal but not the vertical component. In the presence of an optokinetic stimulus a nystagmus was induced producing more frequent and larger torsional quick phases compared to microsaccades produced during fixation of a stationary stimulus. The torsional component and the vertical vergence component of quick phases increased with higher velocities. In previous research, microsaccades have been interpreted as psychophysiological indicators of task load. So far, it is still under debate how different types of task demands are influencing microsaccade rate. In their article “The interplay between task difficulty and microsaccade rate: Evidence for the critical role of visual load” Schneider et al. (1921) examined the relation between visual load, mental load and microsaccade rate. The participants carried out a continuous performance task (n-back) in which visual task load (letters vs. abstract figures) and mental task load (1-back to 4-back) were manipulated as within-subjects variables. Eye tracking data, performance data as well as subjective workload were recorded. Data analysis revealed an increased level of microsaccade rate for stimuli of high visual demand (i.e., abstract figures), while mental demand (n-back-level) did not modulate microsaccade rate. The authors concluded that microsaccade rate reflects visual load of a task rather than its mental load. This conclusion is in accordance with the proposition of Krueger et al. (2019) “Microsaccades distinguish looking from seeing”, linking sensory with cognitive phenomena. The present special thematic issue adds several new interesting facets to the research landscape around microsaccades. They still remain an attractive focus of interdisciplinary research and transdisciplinary applications. Thus, as already noted in the first part of this special thematic issue, research on microsaccades will not only endure, but keep evolving as the knowledge base expands. Full article

Cholinergic muscarinic stimulation of vast areas of the limbic brain induced a well-documented polydipsia in laboratory rats. This excessive water-drinking behavior has not received any convincing biological and physiological interpretation for the last 50 years. This review offers such an interpretation and suggests that cholinergically induced drinking response, mostly by carbachol, is associated with activation of the ascending mesolimbic cholinergic system that serves for initiation of emotional aversive arousal of the organism. The ascending cholinergic system originates from the laterodorsal tegmental nucleus, has a diffuse nature, and affects numerous subcortical limbic structures. It is proposed that the carbachol-induced drinking response is related to the state of anxiety and does not serve the regulation of thirst. Instead, the response is anxiety-induced polydipsia that might occur as a soothing procedure that decreases the aversiveness of the negative emotional state induced by carbachol. It is concluded that carbachol-induced water-drinking behavior is a rewarding process that contributes to alleviating the feeling of anxiety by bringing some relief from the cholinergically induced aversive state, and it is a homologue to anxiety-driven polydipsia in humans. Arousal and distress are often important factors in problematic behaviours, and endogenous corticosteroids are important mediators in the associated stress responses. Exogenous corticosteroid treatments have been reported to change behaviour in human patients and laboratory animals, with similar changes also noted in pet dogs. These behaviours not only potentially adversely impact the welfare of the dogs, but also the quality of life of their owners. Indeed, corticosteroids can bias sensitivity towards aversion in dogs. A decrease in behaviours associated with positive affective states, such as play and exploratory behaviours, together with an increase in aggression and barking have also been described in dogs. According to the available literature, human patients with pre-existing psychiatric disorders are more at risk of developing behavioural side effects due to corticosteroid treatments. It is reasonable to consider that the same may happen in dogs with pre-existing behavioural problems. In this paper, the possible behavioural side effects of exogenous corticosteroids are summarised to help inform and support veterinarians prescribing these drugs. Full article

Proactive aggression (PA) is a planned and unprovoked form of aggression that is most often enacted for personal gain or in anticipation of a reward. Frequently described as “cold-blooded” or goal oriented, PA is thought to be associated with low autonomic arousal. With this view in mind, we performed a scoping review of the biological correlates of PA and identified 74 relevant articles. Physiological findings indicated a robust association between PA and reduced resting heart rate, and to a lesser extent a relationship between PA and decreased heart rate and skin conductance reactivity, perhaps indicating dampened sympathetic function. The twin literature identified PA as a heritable trait, but little evidence implicates specific genes in the pathogenesis of PA. Neuroimaging studies of PA pinpoint impaired amygdala function in the assessment and conditioning of aversive stimuli, which may influence the establishment of behavioral patterns. Nodes of the default mode network were identified as possible neural correlates of PA, suggesting that altered function of this network may be involved in the genesis of PA. Given the overlap of PA with reactive aggression and the overall behavioral complexity of PA, it is clear that multiple endophenotypes of PA exist. This comprehensive review surveys the most salient neurobiologically informed research on PA. Full article

The ability to rapidly process others’ emotional signals is crucial for adaptive social interactions. However, to date it is still unclear how observing emotional facial expressions affects the reactivity of the human motor cortex. To provide insights on this issue, we employed single-pulse transcranial magnetic stimulation (TMS) to investigate corticospinal motor excitability. Healthy participants observed happy, fearful and neutral pictures of facial expressions while receiving TMS over the left or right motor cortex at 150 and 300 ms after picture onset. In the early phase (150 ms), we observed an enhancement of corticospinal excitability for the observation of happy and fearful emotional faces compared to neutral expressions specifically in the right hemisphere. Interindividual differences in the disposition to experience aversive feelings (personal distress) in interpersonal emotional contexts predicted the early increase in corticospinal excitability for emotional faces. No differences in corticospinal excitability were observed at the later time (300 ms) or in the left M1. These findings support the notion that emotion perception primes the body for action and highlights the role of the right hemisphere in implementing a rapid and transient facilitatory response to emotional arousing stimuli, such as emotional facial expressions. Ultrasonic vocalizations are among the oldest evolutionarily forms of animal communication. In order to study the communication patterns in an aversive social situation, we used a behavioral model in which one animal, the observer, is witnessing as his cagemate, the demonstrator, is experiencing a series of mild electrical foot shocks. We studied the effect of the foot shock experience on the observer and the influence of a warning sound (emitted shortly before the shock) on USV communication. These experiments revealed that such a warning seems to increase the arousal level, which differentiates the responses depending on previous experience. This can be identified by the emission of characteristic, short 22 kHz calls of a duration below 100 ms. Two rats emitted calls that overlapped in time. Analysis of these overlaps revealed that in ‘warned’ pairs with a naive observer, 22 kHz calls were mixed with 50 kHz calls. This fact, combined with a high fraction of very high-pitched 50 kHz calls (over 75 kHz), suggests the presence of the phenomenon of social buffering. Pure 22 kHz overlaps were mostly found in ‘warned’ pairs with an experienced observer, suggesting a possible fear contagion with distress sharing. The results show the importance of dividing 22 kHz calls into long and short categories. Full article

This review summarizes all reported and suspected functions of ultrasonic vocalizations in infant and adult rats. The review leads to the conclusion that all types of ultrasonic vocalizations subserving all functions are vocal expressions of emotional arousal initiated by the activity of the reticular core of the brainstem. The emotional arousal is dichotomic in nature and is initiated by two opposite-in-function ascending reticular systems that are separate from the cognitive reticular activating system. The mesolimbic cholinergic system initiates the aversive state of anxiety with concomitant emission of 22 kHz calls, while the mesolimbic dopaminergic system initiates the appetitive state of hedonia with concomitant emission of 50 kHz vocalizations. These two mutually exclusive arousal systems prepare the animal for two different behavioral outcomes. The transition from broadband infant isolation calls to the well-structured adult types of vocalizations is explained, and the social importance of adult rat vocal communication is emphasized. The association of 22 kHz and 50 kHz vocalizations with aversive and appetitive states, respectively, was utilized in numerous quantitatively measured preclinical models of physiological, psychological, neurological, neuropsychiatric, and neurodevelopmental investigations. The present review should help in understanding and the interpretation of these models in biomedical research. Full article

Post-Traumatic Stress Disorder (PTSD) is a complex disorder involving dysregulation of stress-related hormones and neurotransmitter systems. Research focused on the endocannabinoid system (eCBS) for anxiety and stress regulation, cognitive and emotional responses modulation and aversive memories extinction, leading to the hypothesis that it could represent a possible alternative treatment target for PTSD. In this systematic review, we summarize evidence about the efficacy and safety of medicinal cannabidiol (CBD), Δ9-tetrahydrocannabinol (Δ9-THC), and nabilone in PTSD treatment. The PRISMA statement guidelines were followed. A systematic literature search was conducted in MEDLINE/PubMed, Scopus and Web of Science by two independent researchers, who also performed data extraction and quality assessment. Among the initial 495 papers, 234 were screened for eligibility and 10 were included. Studies suggested that different medicinal cannabinoids at distinct doses and formulations could represent promising treatment strategies for the improvement of overall PTSD symptomatology as well as specific symptom domains (e.g., sleep disorders, arousal disturbances, suicidal thoughts), also influencing quality of life, pain and social impact. Although there is a robust rationale for treatment with drugs that target the eCBS and the results are promising, further studies are needed to investigate the safety and efficacy profile of their prolonged use. Various studies suggested alterations in pain perception in psychiatric disorders, such as borderline personality disorder (BPD) and major depression (MD). We previously investigated affective components of pain perception in BPD compared to healthy controls (HC) by increasing aversive stimulus intensities using repetitive peripheral magnetic stimulation (rPMS) and observed alterations in emotional rather than somatosensory components in BPD. However, conclusions on disorder specific alterations in these components of pain perception are often limited due to comorbid depression and medication in BPD. Here, we compared 10 patients with BPD and comorbid MD, 12 patients with MD without BPD, and 12 HC. We applied unpleasant somatosensory stimuli with increasing intensities by rPMS and assessed pain threshold (PT), cutaneous sensation, emotional valence, and arousal by a Self-Assessments Manikins scale. PTs in BPD were significantly higher compared to HC. The somatosensory discrimination of stimulus intensities did not differ between groups. Though elevated rPMS intensities led to increased subjective aversion and arousal in MD and HC, these emotional responses among intensity levels remained unchanged in BPD. Our data give further evidence for disorder-specific alterations in emotional components of pain perception in BPD with an absent emotional modulation among varying aversive intensity levels. A large body of literature documents the sensitivity of pupil response to cognitive load (e.g., Krejtz et al., 2018) and emotional arousal (Bradley et al., 2008). Recent empirical evidence also showed that microsaccade characteristics and dynamics can be modulated by mental fatigue and cognitive load (e.g., Dalmaso et al., 2017). Very little is known about the sensitivity of microsaccadic characteristics to emotional arousal. The present paper demonstrates in a controlled experiment pupillary and microsaccadic responses to information processing during multi-attribute decision making under affective priming. Twenty-one psychology students were randomly assigned into three affective priming conditions (neutral, aversive, and erotic). Participants were tasked to make several discriminative decisions based on acquired cues. In line with the expectations, results showed microsaccadic rate inhibition and pupillary dilation depending on cognitive effort (number of acquired cues) prior to decision. These effects were moderated by affective priming. Aversive priming strengthened pupillary and microsaccadic response to information processing effort. In general, results suggest that pupillary response is more biased by affective priming than microsaccadic rate. The results are discussed in the light of neuropsychological mechanisms of pupillary and microsaccadic behavior generation. Full article
