Reliable quality of epicutaneous patch test (PT) preparations is a prerequisite for establishing a robust diagnosis in patients with suspected allergic contact dermatitis due to delayed-type sensitization. It is difficult to identify potential quality issues in daily practice, since confirmatory methods are lacking and assessment of PT-relevance is predominantly based on patients’ history and exposure. The quality of PT products can be affected, e.g., by the properties of the active substance, an insufficient development of the PT preparation or issues during manufacturing. Resulting quality deficiencies can cause both false-negative and false-positive test results. As PT preparations are medicinal products according to Directive 2001/83/EC, they require a marketing authorization (MA) entailing assessment of quality, safety and efficacy by the competent authorities. The corresponding product dossier is the basis for MA. It is continuously updated, e.g., upon change of a source material supplier, ensuring comparability of the respective product over time. Compliance with regulatory requirements is a crucial foundation for sustainable quality to prevent product deficiencies, ensuring reliable test results in practice. Harmonization across the EU is important to ensure the widespread availability of high-quality PT products. This review presents the MA requirements of PT preparations in the EU, as well as challenges previously reported by physicians. Full article

Background/Objectives: Hypersensitivity reactions (HSRs) to iodinated contrast media (ICM), both immediate and non-immediate, pose clinical challenges despite using low-osmolality agents. This review aims to summarize current diagnostic approaches, cross-reactivity patterns, and the debated role of premedication. Methods: A narrative review was conducted using PubMed (2014–2024), selecting studies on ICM-related HSRs, focusing on skin and in vitro testing, drug provocation tests (DPTs), cross-reactivity, and premedication. Results: Skin tests show limited sensitivity, especially for non-immediate reactions. Cross-reactivity among ICMs is common but unpredictable. DPTs are the diagnostic gold standard but lack standardized protocols. Premedication is frequently used, though its efficacy remains uncertain. Conclusions: The management of ICM hypersensitivity is limited by diagnostic gaps and insufficient evidence on premedication. Standardized protocols and prospective studies are needed to improve patient safety and guide clinical decisions. Full article

The presence of dead broilers within a flock can be significant vectors for disease transmission and negatively impact the overall welfare of the remaining broilers. This study introduced a dead broiler detection method that leverages the fact that dead broilers remain stationary within the flock in videos. Dead broilers were identified through the analysis of the historical movement information of each broiler in the video. Firstly, the frame difference method was utilized to capture key frames in the video. An enhanced segmentation network, YOLOv8-SP, was then developed to obtain the mask coordinates of each broiler, and an optical flow estimation method was employed to generate optical flow maps and evaluate their movement. An average optical flow intensity (AOFI) index of broilers was defined and calculated to evaluate the motion level of each broiler in each key frame. With the AOFI threshold, broilers in the key frames were classified into candidate dead broilers and active live broilers. Ultimately, the identification of dead broilers was achieved by analyzing the frequency of each broiler being judged as a candidate death in all key frames within the video. We incorporated the parallelized patch-aware attention (PPA) module into the backbone network and improved the overlaps function with the custom power transform (PT) function. The box and mask segmentation mAP of the YOLOv8-SP model increased by 1.9% and 1.8%, respectively. The model’s target recognition performance for small targets and partially occluded targets was effectively improved. False and missed detections of dead broilers occurred in 4 of the 30 broiler testing videos, and the accuracy of the dead broiler identification algorithm proposed in this study was 86.7%. Hypopigmentation disorders due to an underproduction of the pigment melanin by melanocytes cause uneven skin coloration, while in hair follicles they cause grey hair. There is a need for novel materials which can stimulate melanogenesis in the skin and hair for personal care use. While titanium dioxide, gold and silver nanoparticles have been extensively used for applications in cosmetic and personal-care products (PCP), the use of relatively inert platinum nanoparticles (PtNPs) has remained underappreciated. PtNPs have been reported to be a mimetic of the enzyme catechol oxidase with small size PtNPs reported to exhibit a higher catechol oxidase activity in a cell-free system, but no testing has been conducted in melanocytes to date. Herein, we have investigated if PtNPs of two sizes (SPtNP: 5 nm; LPtNP: 50 nm) might have an effect on melanogenesis. To this end, we have used MNT-1 human melanoma cells and primary human melanocytes from moderately-pigmented skin (HEMn-MP). Both SPtNP and LPtNP were nontoxic over a concentration range 6.25–25 μg/mL, hence these concentrations were used in further experiments. Both PtNPs stimulated higher extracellular melanin levels than control; SPtNP at concentrations 12.5 and 25 μg/mL significantly stimulated higher levels of extracellular melanin as compared to similar concentrations of LPtNP in MNT-1 cells, in the absence of ROS generation. The effects of PtNPs on melanin secretion were reversible upon removal of PtNPs from the culture medium. The results of primary particle size-specific augmentation of extracellular melanin by SPtNPs were also validated in HEMn-MP cells. Our results thus provide a proof-of-principle that SPtNP might hold potential as a candidate for the treatment of white skin patches, for sunless skin-tanning and for use in anti-greying hair products in cosmetics. Background and Aim: Nickel (Ni)-rich foods can induce allergic contact mucositis (ACM) with irritable bowel syndrome (IBS)-like symptoms in predisposed subjects. Ni ACM has a high prevalence (>30%) in the general population and can be diagnosed by a Ni oral mucosa patch test (omPT). Many celiac disease (CD) patients on a gluten-free diet (GFD) often show a recrudescence of gastrointestinal and extraintestinal symptoms, although serological and histological remission has been achieved. Since a GFD often results in higher loads of ingested alimentary Ni (e.g., corn), we hypothesized that it would lead to a consequent intestinal sensitization to Ni in predisposed subjects. We wanted to (1) study Ni ACM prevalence in still symptomatic CD patients on a GFD and (2) study the effects of a low-Ni diet (LNiD) on their recurrent symptoms. Material and Methods: We recruited 102 consecutive CD patients (74 female, 28 male; age range 18–65 years, mean age 42.3 ± 7.4) on a GFD since at least 12 months, in current serological and histological remission (Marsh–Oberhuber type 0–I) who complained of relapsing gastrointestinal and/or extraintestinal symptoms. Inclusion criteria: presence of at least three gastrointestinal symptoms with a score ≥5 on the modified Gastrointestinal Symptom Rating Scale (GSRS) questionnaire. Exclusion criteria: IgE-mediated food allergy; history of past or current cancer; inflammatory bowel diseases; infectious diseases including Helicobacter pylori; lactose intolerance. All patients enrolled underwent Ni omPT and followed a LNiD for 3 months. A 24 symptoms questionnaire (GSRS modified according to the Salerno Experts’ Criteria, with 15 gastrointestinal and 9 extraintestinal symptoms) was administered at T0 (free diet), T1 (GFD, CD remission), T2 (recurrence of symptoms despite GFD), and T3 (GFD + LNiD) for comparisons. Comparisons were performed using Wilcoxon signed-rank test. RESULTS: Twenty patients (all female, age range 23–65 years, mean age 39.1 ± 2.9) out of 102 (19.6%) were finally included. All 20 patients enrolled (100%) showed positive Ni omPT, confirming an Ni ACM diagnosis. A correct GFD (T0 vs. T1) induced the improvement of 19 out of the total 24 (79.2%) symptoms, and 14 out of 24 (58.3%) were statistically significant (p-value < 0.0083 according to Bonferroni correction). Prolonged GFD (T1 vs. T2) revealed the worsening of 20 out of the total 24 (83.3%) symptoms, and 10 out of 24 (41.7%) were statistically significant. LNiD (T2 vs. T3) determined an improvement of 20 out of the total 24 (83.4%) symptoms, and in 10 out of 24 (41.7%) symptoms the improvement was statistically significant. Conclusions: Our data suggest that the recrudescence of gastrointestinal and extraintestinal symptoms observed in CD subjects during GFD may be due to the increase in alimentary Ni intake, once gluten contamination and persisting villous atrophy are excluded. Ni overload can induce Ni ACM, which can be diagnosed by a specific Ni omPT. Improvement of symptoms occurs after a proper LNiD. These encouraging data should be confirmed with larger studies. Alimentary nickel (Ni) may result in allergic contact mucositis (ACM), whose prevalence is >30% and may present with IBS-like and extra-intestinal symptoms. These symptoms are also frequent in endometriosis, and Ni allergic contact dermatitis has already been observed in endometriosis. Therefore, intestinal and extra-intestinal symptoms in endometriosis may depend on a Ni ACM, and a low-Ni diet could improve symptoms. We studied the prevalence of Ni ACM in endometriosis and focused on the effects of a low-Ni diet on gastrointestinal, extra-intestinal, and gynecological symptoms. We recruited 84 women with endometriosis, symptomatic for gastrointestinal disorders. Thirty-one out of 84 patients completed the study. They underwent Ni oral mucosa patch test (omPT), questionnaire for intestinal/extra-intestinal/gynecological symptoms, and a low-Ni diet. Clinical evaluation was performed at baseline (T0) and after three months (T1). Twenty-eight out 31 (90.3%) patients showed Ni omPT positive results, with Ni ACM diagnosis, whereas three out of 31 (9.7%) patients showed negative Ni omPT. After three months of low-Ni diet, all gastrointestinal, extra-intestinal and gynecological symptoms showed a statistically significant reduction. Ni ACM has a high prevalence in endometriosis and a low-Ni diet may be recommended in this condition to reduce gastrointestinal, extra-intestinal and gynecological symptoms. Full article

Canopy layers form essential structural components, affecting stand productivity and wildlife habitats. Airborne laser scanning (ALS) provides horizontal and vertical information on canopy structure simultaneously. Existing approaches to assess canopy layering often require prior information about stand characteristics or rely on pre-defined height thresholds. We developed a multi-scale method using ALS data with point densities >10 pts/m2 to determine the number and vertical extent of canopy layers (canopylayer, canopylength), seasonal variations in the topmost canopy layer (canopytype), as well as small-scale heterogeneities in the canopy (canopyheterogeneity). We first tested and developed the method on a small forest patch (800 ha) and afterwards tested transferability and robustness of the method on a larger patch (180,000 ha). We validated the approach using an extensive set of ground data, achieving overall accuracies >77% for canopytype and canopyheterogeneity, and >62% for canopylayer and canopylength. We conclude that our method provides a robust characterization of canopy layering supporting automated canopy structure monitoring. Full article

Outlining patches dominated by different plants in wetland vegetation provides information on species succession, microhabitat patterns, wetland health and ecosystem services. Aerial photogrammetry and hyperspectral imaging are the usual data acquisition methods but the application of airborne laser scanning (ALS) as a standalone tool also holds promises for this field since it can be used to quantify 3-dimensional vegetation structure. Lake Balaton is a large shallow lake in western Hungary with shore wetlands that have been in decline since the 1970s. In August 2010, an ALS survey of the shores of Lake Balaton was completed with 1 pt/m2 discrete echo recording. The resulting ALS dataset was processed to several output rasters describing vegetation and terrain properties, creating a sufficient number of independent variables for each raster cell to allow for basic multivariate classification. An expert-generated decision tree algorithm was applied to outline wetland areas, and within these, patches dominated by Typha sp. Carex sp., and Phragmites australis. Reed health was mapped into four categories: healthy, stressed, ruderal and die-back. The output map was tested against a set of 775 geo-tagged ground photographs and had a user’s accuracy of > 97% for detecting non-wetland features (trees, artificial surfaces and low density Scirpus stands), > 72% for dominant genus detection and > 80% for most reed health categories (with 62% for one category). Overall classification accuracy was 82.5%, Cohen’s Kappa 0.80, which is similar to some hyperspectral or multispectral-ALS fusion studies. Compared to hyperspectral imaging, the processing chain of ALS can be automated in a similar way but relies directly on differences in vegetation structure and actively sensed reflectance and is thus probably more robust. The data acquisition parameters are similar to the national surveys of several European countries, suggesting that these existing datasets could be used for vegetation mapping and monitoring.
