Acid mine drainage (AMD) is an environmental concern that needs to be addressed by some mining industries because of its high concentrations of metals and acidity that destroy affected ecosystems. Its formation typically persists beyond the operating life of a mine site. Its management is even more challenging for sites that are abandoned without rehabilitation. In this study, a legacy copper–gold mine located in Sto. Niño, Tublay, Benguet, Philippines, generating a copper- and manganese-rich AMD (Cu, maximum 17.2 mg/L; Mn, maximum 2.90 mg/L) at pH 4.59 (minimum) was investigated. With its remote location inhabited by the indigenous people local community (IPLC), a novel limestone-based hybrid passive treatment system that combines a limestone leach bed (LLB) and a controlled modular packed bed reactor (CMPB) has been developed from the laboratory and successfully deployed in the field while investigating the effective hydraulic retention time (HRT), particle size, and redox conditions (oxic and anoxic) in removing Cu and Mn and increasing pH. Laboratory-scale and pilot-scale systems using simulated and actual AMD, respectively, revealed that a 15 h HRT and both oxic and anoxic conditions were effective in treating the AMD. Considering these results and unsteady conditions of the stream in the legacy mine, a hybrid multi-stage limestone leach bed and packed bed were deployed having variable particle size (5 mm to 100 mm) and HRT. Regular monitoring of the system showed the effective removal of Cu (88.5%) and Mn (66.83%) as well as the increase of pH (6.26), addressing the threat of AMD in the area. Improvement of the lifespan of the system needs to be addressed, as issues of Cu-armoring were observed, resulting in reduced performance over time. Nonetheless, the study presents a novel technique in implementing passive treatment systems beyond the typical treatment trains reported in the literature. Full article

The tyrosine protein kinase (TXK) gene, as a member of the non-receptor tyrosine kinase Tec family, plays a vital role in signal transduction mediation. Phospholipase C epsilon 1 (PLCE1), a membrane-associated enzyme, is of paramount importance for the differentiation of myoblasts and the normal functioning of muscle tissue. In recent years, both of these genes have been reported to be associated with the economic traits of animals. This study aimed to investigate the relationship between single nucleotide polymorphisms (SNPs) in the TXK and PLCE1 genes and growth traits in Ashidan yaks and to search for potential molecular marker loci that can influence Ashidan yak breeding. A cGPS liquid microarray was utilized to genotype 232 Ashidan yaks and to analyze correlations between two SNP loci in the TXK and PLCE1 genes and yak body weight, body height, body length, and chest circumference at different periods. The results indicated that the g.55,999,531C>T locus of the TXK gene and the g.342,350T>G locus of the PLCE1 gene were significantly correlated with the growth traits of Ashidan yaks. Among these, individuals with the CC genotype at the g.55,999,531C>T locus showed a significantly higher body length at 6 months old compared to TT individuals, and those with the CT genotype at 12 months old had a significantly higher chest circumference than TT individuals. At the g.342,350T>G locus, the body height of GG genotype individuals at 18 months of age was significantly higher than that of TT genotype individuals and TG genotype individuals. The above findings can be used as theoretical support for the subsequent improvement of Ashidan yak breeding. Full article

Mangroves provide numerous ecological, social, and economic benefits that include carbon sequestration, habitat for biodiversity, food, recreation and leisure, income, and coastal resilience. In this regard, mangrove-based carbon market projects (MbCMP), involving mangrove conservation, protection, and restoration, are a nature-based solution (NbS) for climate change mitigation. Despite the proliferation of blue carbon projects, a highly publicized need for local community participation by developers, and existing project implementation standards, local communities are usually left out for several reasons, such as a lack of capacity to engage in business-to-business (B2B) market agreements and communication gaps. Local communities need to be engaged and supported at all stages of the MbCMP development process to enable them to protect their ecological, economic, and social interests as custodians of such a critical ecosystem. In this paper, we provided 15 strategic considerations and recommendations to engage and secure the interests of local communities in the growing mangrove carbon market trade. The 15 considerations are grouped into four recommendation categories: (i) project development and community engagement, (ii) capacity building and educational activities, (iii) transparency in resource allocation and distribution, and (iv) partnerships with local entities and long-term monitoring. We expect our study to increase local participation and community-level ecological, social, and economic benefits from MbCMP by incorporating equitable benefit-sharing mechanisms in a B2B conservation-agreement model. Aminoglycosides (AGs) represent a prominent class of antibiotics widely employed for the treatment of various bacterial infections. Their widespread use has led to the emergence of antibiotic-resistant strains of bacteria, highlighting the need for analytical methods that allow the simple and reliable determination of these drugs in pharmaceutical formulations and biological samples. In this study, a simple, robust and easy-to-use analytical method for the simultaneous determination of five common aminoglycosides was developed with the aim to be widely applicable in routine laboratories. With this purpose, different approaches based on liquid chromatography with direct UV spectrophotometric detection methods were investigated: on the one hand, the use of stationary phases based on hydrophilic interactions (HILIC); on the other hand, the use of reversed-phases in the presence of an ion-pairing reagent (IP-LC). The results obtained by HILIC did not allow for an effective separation of aminoglycosides suitable for subsequent spectrophotometric UV detection. However, the use of IP-LC with a C18 stationary phase and a mobile phase based on tetraborate buffer at pH 9.0 in the presence of octanesulfonate, as an ion-pair reagent, provided adequate separation for all five aminoglycosides while facilitating the use of UV spectrophotometric detection. The method thus developed, IP-LC-UV, was optimized and applied to the quality control of pharmaceutical formulations with two or more aminoglycosides. Furthermore, it is demonstrated here that this methodology is also suitable for more complex matrices, such as serum, which expands its field of application to therapeutic drug monitoring, which is crucial for aminoglycosides, with a therapeutic index ca. 50%. Dengue is an arboviral disease that has spread globally and become a major public health concern. A small proportion of patients may progress from symptomatic dengue fever (DF) to dengue hemorrhagic fever (DHF) or dengue shock syndrome (DSS). Findings from a previous genome-wide association study (GWAS) demonstrated that variations in the major histocompatibility complex (MHC) class I chain-related B (MICB) and the phospholipase C epsilon 1 (PLCE1) genes were related to DSS in a Vietnamese population. This study investigated associations of variations in MICB (rs3132468) and PLCE1 (rs3740360, rs3765524) with dengue severity and thrombocytopenia in both the Indonesian and Taiwanese populations. We sampled 160 patients from the Indonesian population and 273 patients from the Taiwanese population. None of the patients had DSS in the Taiwanese population. Based on age demographics, we found that dengue is more prevalent among younger individuals in the Indonesian population, whereas it has a greater impact on adults in the Taiwanese population. Our results showed the association between MICB rs3132468 and DSS. In addition, an association was identified between PLCE1 rs3740360 and DHF in secondary dengue in Indonesian patients. However, there is no association of MICB or PLCE1 variants with thrombocytopenia. This study highlights the value of genetic testing, which might be included in the clinical pathway for specific patients who can be protected from severe dengue. Full article

Conservationists recognize that protected areas (PAs) have limited prospects without the involvement and support of local people. As a governance strategy, community participation is to implement the coordinated development of communities and PAs. However, the effects of community participation on national park governance have rarely been tested. Therefore, the present study used a mixed-method approach that is derived from the International Union for Conservation of Nature (IUCN) green list of protected and conserved areas (PCA) conservation outcomes framework, calibrated to the indigenous peoples and local communities’ (IPLCs) self-assessments about the outcomes of community participation on national park governance to explore the community participation effects. Our results show that management efficiency controls governance outcomes. Potatso National Park’s transformation from the tourism development model to national park is still ongoing, and there exists quite a few problems. We conclude that a successful national park governance as envisaged by the “ecological civilization” paradigm requires a balance of government regulation, participation of various stakeholders in decision-making and discussion, compensation, as well as sustainable access to environmental resources by the affected populations. Blended finance aims to unlock additional private finance for the sustainable development goals (SDGs), however, it has not yet reached the anticipated scale to deliver on SDG 15: Life on Land. So far, blended finance approaches have not been fully adapted to the context where conservation activities take place, for example on communal lands—a common tenure arrangement for conservation in southern Africa. This study identifies opportunities, barriers, and risks to up-scaling private finance for nature in the context of community-based natural resource management (CBNRM) in southern Africa. It considers the feasibility and desirability of relevant revenue streams towards achieving long-term financial sustainability in conservation landscapes, including sustainable wildlife economies and payment for ecosystem services (PES), and involving indigenous peoples and local communities (IPLCs) particularly within CBNRM tenure arrangements. It concludes that a ‘CBNRM investment guarantee’ or similar would be transformational for hundreds of thousands of conservation enterprises and their beneficiaries regionally, but currently no such tailored de-risking mechanism exists. The role of Indigenous peoples and local communities (IPLCs) in sustainably using and managing natural resources is becoming broadly recognised within some international platforms (e.g., the UN Permanent Forum on Indigenous Issues and Intergovernmental Platform on Biodiversity and Ecosystem Services). However, the support for IPLCs to continue managing their land is either completely absent or scanty. This paper presents the value of only four ecosystem services, estimated at USD 1.16 trillion per year, that are delivered from IPLCs managed lands alone (excluding coastal, marine, and other resources). These four ecosystem services (ES), i.e., carbon sequestration, biocontrol, air, and water regulation offer offsite benefits to the wider regional and global populations yet without returns to the IPLCs themselves except for facing more climate and natural disaster-related challenges mainly caused by the actions of mainstream society. It further outlines key challenges and advocates for establishing stewardship mechanisms to promote and support IPLCs land management practices that will effectively help in protecting and preserving biodiversity, water, and other natural resources on Earth to sustain and enhance human well-being. In this paper, we review the role of the norpA-encoded phospholipase C in light and thermal entrainment of the circadian clock in Drosophila melanogaster. We extend our discussion to the role of norpA in the thermo-sensitive splicing of the per 3′ UTR, which has significant implications for seasonal adaptations of circadian behaviour. We use the norpA mutant-generated enhancement of per splicing and the corresponding advance that it produces in the morning (M) and evening (E) locomotor component to dissect out the neurons that are contributing to this norpA phenotype using GAL4/UAS. We initially confirmed, by immunocytochemistry and in situ hybridisation in adult brains, that norpA expression is mostly concentrated in the eyes, but we were unable to unequivocally reveal norpA expression in the canonical clock cells using these methods. In larval brains, we did see some evidence for co-expression of NORPA with PDF in clock neurons. Nevertheless, downregulation of norpA in clock neurons did generate behavioural advances in adults, with the eyes playing a significant role in the norpA seasonal phenotype at high temperatures, whereas the more dorsally located CRYPTOCHROME-positive clock neurons are the likely candidates for generating the norpA behavioural effects in the cold. We further show that knockdown of the related plc21C encoded phospholipase in clock neurons does not alter per splicing nor generate any of the behavioural advances seen with norpA. Our results with downregulating norpA and plc21C implicate the rhodopsins Rh2/Rh3/Rh4 in the eyes as mediating per 3′ UTR splicing at higher temperatures and indicate that the CRY-positive LNds, also known as ‘evening’ cells are likely mediating the low-temperature seasonal effects on behaviour via altering per 3′UTR splicing. Background and Objectives: Congenital nephrotic syndrome (CNS), a genetic disease caused by mutations in genes on autosomes, usually occurs in the first three months after birth. A number of genetic mutations in genes, which encode for the components of the glomerular filtration barrier have been identified. We investigated mutations in NPHS1, NPHS2, PLCE1 (NPHS3), and WT1 genes that relate to the disease in Vietnamese patients. Materials and Methods: We performed genetic analysis of two unrelated patients, who were diagnosed with CNS in the Vietnam National Children’s Hospital with different disease status. The entire coding region and adjacent splice sites of these genes were amplified and sequenced using the Sanger method. The sequencing data were analyzed and compared with the NPHS1, NPHS2, PLCE1, and WT1 gene sequences published in Ensembl (ENSG00000161270, ENSG00000116218, ENSG00000138193, and ENSG00000184937, respectively) using BioEdit software to detect mutations. Results: We detected a new variant p.Ser607Arg and two other (p.Glu117Lys and p.Ser1105Ser) in the NPHS1 gene, as well as two variants (p.Arg548Leu, p.Pro1575Arg) in the PLCE1 gene. No mutations were detected in the NPHS2 and WT1 genes. Patient 1, who presented a heterozygous genotype of p.Ser1105Ser and p.Arg548Leu had a mild disease status but patient 2, who presented a homozygous genotype of these alleles, had a severe phenotype. Conclusions: These results suggest that variants p.Ser1105Ser (in NPHS1 gene) and p.Arg548Leu (in PLCE1 gene) in the homozygous form might play a role in the development of the disease in patients.
