Eucomis autumnalis is a bulbous ornamental species with ethnobotanical relevance. In vitro cultures offer a sustainable tool for biomass propagation and metabolite production. This study investigates the effects of nine LED light spectra: red (R), blue (B), red–blue (RB), RB with green (RBG), yellow (RBY), far-red (RBfR), ultraviolet (RBUV), white (WLED), and fluorescent light (Fl, control), on the morphogenesis, polyphenol production, and antioxidant potential of E. autumnalis shoot cultures. Cultures were maintained on MS medium with 5 µM BA and 0.5 µM NAA. HPLC-DAD analysis identified 11 phenolic acids and 4 flavonoids, including eucomic acid, characteristic of the genus. Light quality impacted compound-specific accumulation and antioxidant activity, with responses varying among compounds and treatments. R and B light increased catechin, gentisic acid and hesperidin (289, 195, 245 mg/100 g DW), while UV suppressed flavonoids by ca. 2-fold for catechin and flavanones compared to other lights. RBG and RBfR induced the highest eucomic acid accumulation (424 mg/100 g DW), ferulic acid and epicatechin, correlating strongly with ABTS•+ activity (18–19% higher than other lights; r > 0.6–0.8). These findings highlight LED spectral modulation as a tool to enhance the phytochemical quality of E. autumnalis in vitro and provide a foundation for future biotechnological applications. Grape pomace is a major by-product of winemaking and a rich source of phenolic compounds with antioxidant potential. The Carménère variety, emblematic of Chilean viticulture, remains underutilized despite its high anthocyanin and flavanol content. This study aimed to develop a cost-effective method to recover and stabilize bioactive compounds from Carménère grape pomace. Five extracts were obtained using ethanol–water mixtures (0–100%) and characterized by HPLC-DAD and antioxidant assays (DPPH, FRAP, ORAC-FL). The 80% ethanol extract (EET-80) showed the highest antioxidant capacity (FRAP: 2909.3 ± 37.6; ORAC-FL: 1864.3 ± 157.8 µmol TE/g dw) and was selected for microencapsulation via spray drying using maltodextrin. This scalable technique protects thermosensitive compounds and enhances their applicability. The optimized 1:50 extract-to-carrier ratio achieved high encapsulation efficiency (85.7 ± 0.7%). In Caco-2 cells, the microencapsulated extract (5–250 µg/mL) showed no alteration in metabolic activity and significantly reduced intracellular ROS levels (65% inhibition at 250 µg/mL). Solvent polarity selectively influenced polyphenol recovery—50% ethanol favored catechin (581.1 µg/g) and epicatechin (1788.3 µg/g), while 80% ethanol enhanced malvidin-3-O-glucoside (118.0 µg/g). These findings support the valorization of Carménère grape pomace as a sustainable source of antioxidants and highlight the role of microencapsulation in improving extract stability and functionality. The search for natural sources of bioactive compounds with health-promoting properties has intensified in recent years. Among these, Tannat grape pomace (TGP), a primary byproduct of winemaking, stands out for its high phenolic content, although its bioactivity may be affected during gastrointestinal digestion. This study aimed to evaluate the impact of in vitro digestion on the antioxidant (ABTS, ORAC-FL, intracellular ROS inhibition), anti-diabetic (α-glucosidase inhibition), anti-obesity (lipase inhibition), and anti-inflammatory (NO inhibition) properties of five sugar-free biscuits formulated with varying percentages of TGP and sucralose. No significant differences were observed in the bioaccessible fractions (BFs, representing the compounds potentially released in the small intestine) between control biscuits and those enriched with TGP, suggesting limited release of phenolics at this stage. Conversely, the colonic fractions (CFs, simulating the material reaching the colon) from biscuits with higher TGP content exhibited greater bioactivities. HPLC-DAD-MS analysis of the CF from the biscuit containing 20% TGP and 4% sucralose revealed a high content of procyanidin trimers, indicating the persistence of these specific phenolic compounds after in vitro digestion. These findings suggest that TGP-enriched biscuits may deliver health benefits at the colonic level and support their potential application in the formulation of functional foods. Further microbiota and in vivo studies should be assessed to confirm the latter. Full article

Citrus limon (L.) Burm. f. is a horticultural crop known for its abundance of valuable secondary metabolites, including flavonoids, which are found in its fruits and leaves. Our previous research has shown that treating C. limon with fulvic acid (FA) can enhance the levels of vitamin C, total acid, total sugar, total flavonoids, and phenols in its fruits as well as the volatiles and total flavonoids in its leaves. In this current study, we established a method to analyze eight specific flavonoids in lemon leaves and evaluated the impact of irrigation with FA on the content of these flavonoids over a six-month period using HPLC-DAD/MS analysis. Moreover, we investigated the potential mechanisms of FA through ELISA and q-PCR methods. The results indicated that FA increased the contents of four flavonoids, namely, isoorientin, eriocitrin, vitexin, and rutin, and promoted the activity and gene expression of phenylalanineammonialyase (PAL), 4-coumaric acid coenzyme A ligase (4CL), chalcone synthase (CHS), flavonoid 3′-hydroxylase (F3′H), and flavonol synthase (FLS). Furthermore, the relationship between flavonoid content and the activities of biosynthetic enzymes was analyzed using orthogonal partial least squares discriminant analysis (OPLS-DA), which revealed a positive correlation between seven flavonoid levels and the activity of five biosynthetic enzymes under FA treatment. Various strategies have been used to increase the efficiency of secondary metabolite production in Salvia plants. This report is the first to examine the spontaneous development of Salvia bulleyana shoots transformed by Agrobacterium rhizogenes on hairy roots and the influence of light conditions on the phytochemical profile of this shoot culture. The transformed shoots were cultivated on solid MS medium with 0.1 mg/L of IAA (indole-3-acetic acid) and 1 mg/L of m-Top (meta-topolin), and their transgenic characteristic was confirmed by PCR-based detection of the rolB and rolC genes in the target plant genome. This study assessed the phytochemical, morphological, and physiological responses of the shoot culture under stimulation by light-emitting diodes (LEDs) with different wavelengths (white, WL; blue, B; red, RL; and red/blue, ML) and under fluorescent lamps (FL, control). Eleven polyphenols identified as phenolic acids and their derivatives were detected via ultrahigh-performance liquid chromatography with diode-array detection coupled to electrospray ionization tandem mass spectrometry (UPLC-DAD/ESI-MS) in the plant material, and their content was determined using high-performance liquid chromatography (HPLC). Rosmarinic acid was the predominant compound in the analyzed extracts. The mixed red and blue LEDs gave the highest levels of polyphenol and rosmarinic acid accumulation (respectively, 24.3 mg/g of DW and 20.0 mg/g of DW), reaching two times greater concentrations of polyphenols and three times greater rosmarinic acid levels compared to the aerial parts of two-year-old intact plants. Similar to WL, ML also stimulated regeneration ability and biomass accumulation effectively. However, the highest total photosynthetic pigment production (1.13 mg/g of DW for total chlorophyll and 0.231 mg/g of DW for carotenoids) was found in the shoots cultivated under RL followed by BL, while the culture exposed to BL was characterized as having the highest antioxidant enzyme activities. Chocolate is made from cocoa, which is rich in (poly)phenols that have a high antioxidant capacity and are associated with the prevention of chronic diseases. In this study, a new production process was evaluated in order to obtain a dark chocolate enriched in (poly)phenols using a cocoa powder with an improved flavanol profile. The antioxidant capacity (Oxygen Radical Absorbance Capacity (ORAC) assay) and the flavanol profile (HPLC-DAD and HPLC-FL) was determined. The analysis of the enriched chocolate showed that the total flavan-3-ols (monomers) content was 4 mg/g representing a 3-fold higher than that quantified in the conventional one. Total levels of dimers (procyanidin B1 and B2) were 2.4-fold higher in the enriched chocolate than in the conventional, with a total content of 6 mg/g. The total flavanol content (flavan-3-ols and procyanidins) in the enriched chocolate was increased by 39% compared to the conventional one which led to a 56% increase in the antioxidant capacity. The new flavanol-enriched dark chocolate is expected to provide greater beneficial effect to consumers. Moreover, the amount of flavonols provided by a single dose (ca. 200 mg per 10 g) would allow the use of a health claim on cardiovascular function, a fact of interest for the cocoa industry. The aim of this research is to demonstrate the concept and ability for the fast and preliminary screening of complex food and environmental samples for the presence of endocrine disrupters fractions, consisting of low-molecular mass micropollutants, particularly various bisphenols (A, B, C, E, F, S, Z, AF, AP, BP and FL). The developed analytical protocol for this research requires two main steps: (i) optimized solid phase extraction (SPE) for selective isolation, purification and pre-concentration of target fraction, and (ii) selective temperature-dependent inclusion chromatography for samples analysis via a HPLC-UV-VisDAD system using isocratic elution and internal standard quantification approach. The chromatographic experiment revealed that both β-CD and its hydroxypropyl derivative strongly interact with selected bisphenols. This is in contrast to the steroids and PAHs molecules investigated previously, where a strong interaction with β-cyclodextrin was observed. Integrated areas derived from acquired chromatographic profiles for each individual sample were used as the simple classification variable enabling samples comparison. We demonstrated that the proposed analytical protocol allows for fast estimation of EDC fractions in various daily use products, food and environmental samples. The materials of interest were selected due to the presence in surface water ecosystems of their residues, and finally, in raw wastewater including rice bags, plastic bags, cloths, sanitary towels, fish baits and various plastic foils from food products. Treated sewage water released directly to the environment from a municipal treatment plant (Jamno, Koszalin) was also investigated. It has been demonstrated that a whole range of low-molecular mass compounds, which may be detected using UV-Vis detector, can easily be emitted from various in daily use products. The presence of micropollutants in treated wastewater, water ecosystems and plastic waste utilization via technological wastewater treatment processes must be addressed, especially in terms of microplastic-based pollutants acting as endocrine disrupters. It is hoped that the proposed simple analytical protocol will be useful for fast sample classification or selection prior to advanced targeted analysis involving the more accurate quantification of specific analytes using e.g., mass spectrometry detectors. Virgin olive oils from the fruits of Croatian autochthonous varieties Oblica, Lastovka and Levantinka were characterized for the first time. Headspace volatiles were analyzed by HS-SPME/GC-FID/MS. The main volatiles were C6 compounds. The most abundant was (E)-hex-2-enal (62.60%–69.20%). (Z)-Hex-3-enal was not found in Lastovka oil, while Levantinka oil did not contain hexanal. Tocopherols, chlorophylls and carotenoids were determined by HPLC-FL. Levantinka oil was characterized by the highest α-tocopherol level (222.00 mg/kg). Total phenolic contents (TPs), as well as antioxidant activity (DPPH assay) of the oils hydrophilic fractions (HFs) were assessed by spectroscopic methods. The antioxidant activity of Oblica oil HF was the most pronounced (0.91 mmol TEAC/kg) and the HF contained the highest TPs amount (212.21 mg/kg). HFs phenolic composition was determined by HPLC-DAD. The main identified phenols were secoiridoids dominated in Oblica oil: decarboxymethyl ligstroside aglycone (p-HPEA-EDA up to 158.5 mg/kg), oleuropein aglycone (3,4-HPEA-EA up to 96.4 mg/kg) and decarboxymethyl oleuropein aglycon (3,4-DHPEA-EDA up to 93.5 mg/kg). Virgin olive oils (VOOs) obtained from the fruits of Croatian autochthonous varieties Mašnjača and Krvavica were extensively characterized for the first time. Investigated oils were compared with the oil obtained from Italian variety Leccino, grown and processed under the same conditions. Headspace volatile profile, tocopherols, chlorophylls, carotenoids and total phenolic content, peroxide value, % acidity, K232, K270 as well as antioxidant activity (DPPH) of the oils’ hydrophilic fractions (HFs) including their phenolic composition were assessed by means of HS-SPME/GC-MS, HPLC-FL, HPLC-DAD and spectrophotometric methods, respectively. Most of the studied quality parameters varied between the cultivars. The main volatile compounds detected in all tested olive oils were the C6 compounds derived from polyunsaturated fatty acids through the lipoxygenase pathway. Krvavica oil was characterized by hexanal (8.8%–9.4%). Leccino oil contained the highest percentage of (E)-hex-2-enal (73.4%–74.0%), whereas (Z)-hex-3-enal (21.9%–25.0%) and (E)-hex-2-enal (27.6%–28.9%) dominated in Mašnjača oil. Leccino oil contained the highest amount of tocopherols (312.4 mg/kg), chlorophylls (7.3 mg/kg), carotenoids (4.2 mg/kg) and total phenols (246.6 mg/kg). The HF of Leccino oil showed the highest antioxidant capacity (1.3 mmol TEAC/kg), while the HFs of Mašnjača and Krvavica oils exhibited the activity of 0.5 mmol TEAC/kg. Full article
