The oil-bearing rose (Rosa damascena Mill.), traditionally cultivated in Bulgaria for centuries, and the rose oil produced from it are of major cultural and economic importance. Its distinctive fragrance and rich aromatic profile are highly valued worldwide. In this study, a set of 15 start codon-targeted (SCoT) molecular markers was used to evaluate the genetic diversity and relationships of 38 rose accessions. The analyzed materials included Bulgarian-bred R. damascena cultivars, a locally improved population (‘Population 5’), three oil-bearing species (Rosa alba L., Rosa gallica L., and Rosa centifolia L.), Romanian heritage roses, and an unidentified rose genotype from an old Bulgarian plantation (Rosa sp.). The SCoT primers yielded a cumulative count of 238 bands, with an average of 12.9 bands per primer. The range of diversity markers, such as PIC (0.20–0.78), number of different alleles (1.5–2.00), Shannon’s information index (0.24–0.69), and gene diversity (0.15–0.50), provided evidence of genetic differences among the examined accessions. Analysis of Molecular Variance (AMOVA) revealed higher genetic variation within groups (61%) than among the groups (39%). Multivariate analyses (UPGMA, PCoA, and STRUCTURE) resolved the accessions into major genetic clusters corresponding to their taxonomic identity or breeding history. The unidentified Rosa sp. formed a distinct genetic group, showing closer affinity to R. gallica. The locally improved R. damascena ‘Population 5’ exhibited higher genetic diversity than the Bulgarian cultivars. Overall, our results demonstrate the effectiveness of SCoT markers and the value of local and traditional rose germplasm as reservoirs of genetic diversity. The study provides a molecular framework to support breeding, conservation, and sustainable management of oil-bearing rose genetic resources. Full article

This study develops and empirically validates a framework integrating cultural factors into sustainable supply chain management (SSCM) for emerging economies. We introduce the Cultural Affinity Index (CAI), a multi-dimensional construct quantifying cultural compatibility between supply chain partners based on language compatibility, regional trust, trade networks, and historical trade patterns. Using publicly available data from UN COMTRADE, the World Bank, and Hofstede Insights, we analyze 850 supplier–manufacturer dyads across five Southeast Asian countries (2019–2023). Through Monte Carlo simulation with empirically calibrated parameters, we demonstrate that high cultural affinity (CAI > 0.7) shows positive associations with economic performance (+18.0%), environmental compliance (+12%), and social sustainability (+32%) compared to baseline scenarios. We test both linear and interaction models, finding that language compatibility and regional trust exhibit synergistic effects (β = 0.15, p < 0.01). Multi-objective optimization reveals Pareto-optimal solutions achieving simultaneous improvements across all triple bottom line dimensions. Sensitivity analysis confirms robustness across varying cultural weights (±20%) and institutional contexts. The framework’s effectiveness varies by institutional quality, with stronger associations in weaker institutional environments (correlation = −0.92). While focused on manufacturing, we discuss adaptations for service sectors. This research provides both theoretical contributions to the SSCM literature and practical tools for organizations managing culturally diverse supply chains in emerging markets. Full article

The WHO considers the Epidermal Growth Factor Receptor (EGFR) one of the key biomarkers of glioblastoma (GB). EGFR can be identified and targeted using molecular recognition elements (MoREs), like aptamers and aptamer–drug conjugates (ApDCs). Understanding the kinetics of anti-EGFR ApDC interactions with EGFR as well as the kinetics of their internalization into the cells is a crucial step for the further development of anti-EGFR ApDCs. For the first time, a novel approach was implemented to study real-time kinetics by measuring the cellular index (CI) using impedance (xCELLigence). Doxorubicin (DOX) was used as an indicator drug. Because DOX intercalates into the DNA double helix, aptamer–DOX non-covalent complexes were obtained. For the anti-EGFR DNA aptamer GR20, an additional duplex was constructed by synthesizing the extra region (GR20h) and via hybridization with the complementary oligonucleotide (h’) to form a duplex (hh’), thus creating the aptamer construct with complementary oligonucleotide (ACCO) GR20hh’. The original HPLC method quantified the assembly efficiency of an ACCO. The ACCO GR20hh’ retained affinity for the recombinant extracellular domain of EGFR, as measured using Biolayer Interferometry (BLI). According to cytofluorimetry, the ACCO GR20hh’ interacts with cells of continuous culture from GB patient (CCGBP) surgical samples. The DOX–ACCO GR20hh’ complexes are more efficiently internalized by EGFR+ cells lines A-431 and CCGBP 107 than DOX alone. Full article

As a result of an ongoing research survey of microfungi in Yunnan, China, several saprobic ascomycetes were collected from various host substrates. Preliminary morphological analyses identified a few of these taxa as Periconia species. We obtained DNA sequence data of the Periconia species from pure cultures and investigated their phylogenetic affinities. Phylogenetic analyses of a combined LSU, ITS, SSU and tef1-α sequence dataset demonstrated that five isolates of Periconia formed well-resolved subclades within Periconiaceae. Accordingly, three new Periconia species are introduced viz. P. artemisiae, P. chimonanthi and P. thysanolaenae, and new host and geographical records of P. byssoides and P. pseudobyssoides, are also reported from dead branches of Prunus armeniaca and Scrophularia ningpoensis. Periconia celtidis formed a monophyletic clade with P. byssoides in the present phylogenetic analyses. Results of the pairwise homoplasy index (PHI) test indicated significant recombination between P. byssoides and P. celtidis. Therefore, P. celtidis has been synonymized under P. byssoides. In addition, we re-illustrated and studied the type specimen of the sexual genus Bambusistroma. As a type species of Bambusistroma, B. didymosporum features similar morphology to the sexual morph of Periconia homothallica and P. pseudodigitata. We therefore synonymize Bambusistroma under Periconia based on morphological and phylogenetic evidence. Furthermore, our new isolates produced brown conidia of asexual morph in agar media typical of the genus Noosia. Based on morphological comparison with Periconia in vitro and phylogenetic status of Noosia, we also treat Noosia as a synonym of Periconia. Detailed descriptions and illustrations of three novel taxa and two new records of Periconia byssoides and P. pseudobyssoides as well as the illustration of P. didymosporum comb. nov. are provided. An updated phylogenetic tree of Periconiaceae using maximum likelihood and Bayesian inference analyses is constructed. Generic circumscription of Periconia is amended. Full article

Two optimization strategies, codon usage modification and glycine supplementation, were adopted to improve the extracellular production of Bacillus sp. NR5 UPM β-cyclodextrin glycosyltransferase (CGT-BS) in recombinant Escherichia coli. Several rare codons were eliminated and replaced with the ones favored by E. coli cells, resulting in an increased codon adaptation index (CAI) from 0.67 to 0.78. The cultivation of the codon modified recombinant E. coli following optimization of glycine supplementation enhanced the secretion of β-CGTase activity up to 2.2-fold at 12 h of cultivation as compared to the control. β-CGTase secreted into the culture medium by the transformant reached 65.524 U/mL at post-induction temperature of 37 °C with addition of 1.2 mM glycine and induced at 2 h of cultivation. A 20.1-fold purity of the recombinant β-CGTase was obtained when purified through a combination of diafiltration and nickel-nitrilotriacetic acid (Ni-NTA) affinity chromatography. This combined strategy doubled the extracellular β-CGTase production when compared to the single approach, hence offering the potential of enhancing the expression of extracellular enzymes, particularly β-CGTase by the recombinant E. coli. Affinity towards diversity (ATD) is the tendency to appreciate the dynamic variety of biophysical and socio-cultural scenarios. ATD may reveal a human predisposition to conserve the diversity necessary to guarantee the preservation of human and natural ecosystems. This study expands upon previous studies which employed only self-report measures by introducing a version of the Implicit Association Test (IAT) and by using a mediational model to test whether ATD through sustainable (pro-environmental and pro-social) behaviors leads to greater wellbeing. We recruited 119 Mexican students to complete a computerized IAT task and respond to ATD, wellbeing, and sustainable behavior self-report questionnaires. Reliability analyses of the scales utilized showed acceptable indexes. A mediational model was tested to examine whether sustainable behaviors mediate the relationship between ATD and wellbeing. Results demonstrated that affinity towards diversity indirectly predicted wellbeing via the mediating effects of pro-environmental behaviors and altruism. Despite finding an association between self-reported affinity towards biodiversity and IAT reaction times, an association was not found with D scores, highlighting the need for further research. These findings suggest that those with increased inclination towards diversity are more prone to engage in environmentally (both physical and social) protective behaviors and to experience psychological wellbeing.
