This study investigates the use of Si and CdTe-based Timepix3 detectors for photovoltaic energy conversion using solar X-rays and other high-energy electromagnetic radiation in space. As space missions increasingly rely on miniaturized platforms like CubeSats, power generation in compact and radiation-prone environments remains a critical challenge. Conventional solar panels are limited by size and spectral sensitivity, prompting the need for alternative energy harvesting solutions—particularly in the high-energy X-ray domain. A novel CubeSat-compatible payload design incorporates a UV-visible filter to isolate incoming X-rays, which are then absorbed by semiconductor detectors to generate electric current through ionization. Laboratory calibration was performed using Fe-55, Ba-133, and Am-241 sources to compare spectral response and clustering behaviour. CdTe consistently outperformed Si in detection efficiency, spectral resolution, and cluster density due to its higher atomic number and material density. Equalization techniques further improved pixel threshold uniformity, enhancing spectroscopic reliability. In addition to experimental validation, simulations were conducted to quantify the expected energy conversion performance under orbital conditions. Under quiet-Sun conditions at 500 km LEO, CdTe absorbed up to 1.59 µW/cm2 compared to 0.69 µW/cm2 for Si, with spectral power density peaking between 10 and 20 keV. The photon absorption efficiency curves confirmed CdTe’s superior stopping power across the 1–100 keV range. Under solar flare conditions, absorbed power increased dramatically, up to 159 µW/cm2 for X-class and 15.9 µW/cm2 for C-class flares with CdTe sensors. A time-based energy model showed that a 10 min X-class flare could yield nearly 1 mJ/cm2 of harvested energy. These results validate the concept of a compact photovoltaic payload capable of converting high-energy solar radiation into electrical power, with dual-use potential for both energy harvesting and radiation monitoring aboard small satellite platforms. We report the results of an ab initio study of the linear and ring structures of cadmium telluride clusters [CdTe]n (CdnTen) n ≤ 10 within the generalized gradient approximation (GGA) and Purdue–Burke–Ernzerhof (PBE) parameterization with Hubbard corrections (GGA+U). We optimized the linear and ring isomers for each size to obtain the lowest-energy structures and to understand their growth behavior. The cases of n < 8 for ring-type structures and n = 6 and 9 for linear-type structures were found to be the most favorable. All observed clusters with a linear structure were found to have a small highest-occupied–lowest-unoccupied molecular orbital (HOMO–LUMO) gap. The CdTe clusters with ring structure showed larger values of the HOMO–LUMO gaps than the band gap value for the bulk crystal. Structural and electronic properties like bond length, the HOMO–LUMO gap, binding energy, and electronegativity were analyzed. Full article

The traceability of quality mineral fingerprints in the viscera of Tan sheep from northwest China was studied. Twenty-five mineral elements in the heart and liver samples of Tan sheep were determined using an inductively coupled plasma mass spectrometer (ICP-MS), and the characteristics of the mineral elements in the visceral tissues of the Tan sheep were further analyzed in combination with a principal component analysis (PCA), hierarchical cluster analysis (HCA), and linear discriminant analysis (LDA) to establish a discriminant model and verify it. The results show that 11 elements (137Ba, 43Ca, 63Cu, 56Fe, 39K, 31P, 60Ni, 78Se, 118Sn, 125Te, and 66Zn) in the Tan sheep heart samples had significant differences among different regions (p < 0.05), and the results of the LDA show that the accuracy rate of the return-generation examination was 85.70%, and the accuracy rate of the hand-over-fork examination was 87.50%; 10 elements (111Cd, 59Co, 52Cr, 56Fe, 39K, 55Mn, 95Mo, 23Na, 121Sb, and 78Se) in the Tan sheep liver samples had significant differences among different regions (p < 0.05), and the results of the LDA showed that the accuracy rate of the return-generation examination was 96.30%, and the accuracy rate of the hand-over-fork examination was 86.25%. This indicates that the multi-element analysis has potential for determining the origin of Tan sheep viscera in certain regions. Full article

A new approach for the botanical origin determination of monofloral bee honey is developed. The methodology combines mineral content and physicochemical parameters determination with intelligent statistics such as self-organizing maps (SOMs). A total of 62 monofloral bee honey samples were analysed, including 31 linden, 14 rapeseed, 13 sunflower, and 4 acacia. All of them were harvested in 2018 and 2019 from trusted beekeepers, after confirming their botanical origin, using melissopalynological analysis. Nine physicochemical parameters were determined, including colour, water content, pH, electrical conductivity, hydroxymethylfurfural content, diastase activity, specific optical rotation, invertase activity, and proline. The content of thirty chemical elements (Ag, Al, As, B, Ba, Bi, Ca, Cd, Co, Cr, Cs, Cu, Fe, Ga, In, K, Li, Mg, Mn, Na, Ni, P, Pb, Rb, S, Se, Sr, Te, V, and Zn) was measured using ICP-OES, ICP-MS, and FAAS as instrumental techniques. The visualisation of the SOMs shows an excellent separation of honey samples in five well-defined clusters—linden, rapeseed, acacia, sunflower, and polyfloral honey—using the following set of 16 descriptors: diastase activity, hydroxymethylfurfural content, invertase activity, pH, specific optical rotation, water content, Al, B, Cr, Cs, K, Na, Ni, Rb, V, and Zn. Major, minor and trace elements in wines from Greece were determined by inductively coupled plasma–mass spectrometry (ICP–MS). The concentrations of 44 elements (Na, Mg, P, K, Ca, Cu, Co, Cr, Zn, Sn, Fe, Mn, Li, Be, B, V, Sr, Ba, Al, Ag, Ni, As, Sn, Hg, Pb, Sb, Cd, Ti, Ga, Zr, Nb, Pd, Te, La, Sm, Ho, Tm, Yb, W, Os, Au, Tl, Th, U) in 90 white and red wines from six different regions in Greece for two consecutive vinification years, 2017 and 2018, were determined. Results for the elements aforementioned were evaluated by multivariate statistical methods, such as discriminant analysis and cluster analysis, and the wines were discriminated according to wine variety and geographical origin. Due to the specific choice of the analytes for multivariate statistical investigation, a prediction rate by cross-validation of 98% could be achieved. The aim of this study was not only to reveal specific relationships between the wine samples or between the chemical variables in order to classify the wines from different regions and varieties according to their elemental profile (wine authentication), but also to observe the annual fluctuation in the mineral content of the studied wine samples. Cluster of differentiation (CD)-73 plays pivotal roles in the regulation of immune reactions via the production of extracellular adenosine, and the overexpression of CD73 is associated with worse outcomes in several types of cancers. Here, we identified 167 esophageal squamous cell carcinoma (ESCC) patients who underwent esophagectomy, including 64 and 103 patients with high and low expression levels of CD73, respectively. Univariate and multivariate analyses showed high expression of CD73 was an independent prognostic factor for worse disease-free survival and overall survival. In addition, we selected another cohort consisting of 38 ESCC patients receiving nivolumab or pembrolizumab and found that treatment response and survival benefit to immunotherapy were strongly correlated with the expression levels of CD73/programmed death ligand 1. Moreover, the transwell assay revealed knockdown of CD73 in two ESCC cell lines, TE1 and KYSE30, exhibited significantly reduced abilities of cell invasion and migration. CD73 silencing also showed that the protein expression levels of CD73, vimentin, and snail were downregulated, while those of E-cadherin were upregulated in Western blotting. The findings of our study indicate CD73 may be an independent prognostic factor for ESCC patients who underwent esophagectomy. Furthermore, it may be associated with the patient responses to immunotherapy. Full article

The purpose of this study was to investigate the tissue regenerating and biomechanical properties of processed eggshell membrane powder (PEP) for use in 3D-scaffolds. PEP is a low-cost, natural biomaterial with beneficial bioactive properties. Most importantly, this material is available as a by-product of the chicken egg processing (breaking) industry on a large scale, and it could have potential as a low-cost ingredient for therapeutic scaffolds. Scaffolds consisting of collagen alone and collagen combined with PEP were produced and analyzed for their mechanical properties and the growth of primary fibroblasts and skeletal muscle cells. Mechanical testing revealed that a PEP/collagen-based scaffold increased the mechanical hardness of the scaffold compared with a pure collagen scaffold. Scanning electron microscopy (SEM) demonstrated an interconnected porous structure for both scaffolds, and that the PEP was evenly distributed in dense clusters within the scaffold. Fibroblast and skeletal muscle cells attached, were viable and able to proliferate for 1 and 2 weeks in both scaffolds. The cell types retained their phenotypic properties expressing phenotype markers of fibroblasts (TE7, alpha-smooth muscle actin) and skeletal muscle (CD56) visualized by immunostaining. mRNA expression of the skeletal muscle markers myoD, myogenin, and fibroblasts marker (SMA) together with extracellular matrix components supported viable phenotypes and matrix-producing cells in both types of scaffolds. In conclusion, PEP is a promising low-cost, natural biomaterial for use in combination with collagen as a scaffold for 3D-tissue engineering to improve the mechanical properties and promote cellular adhesion and growth of regenerating cells. The Oktyabrsk PGE-Cu-Ni deposit is one of the largest resources in the Norilsk–Talnakh ore district, Russia, and it is viewed as an ore giant on a global scale. It contains three types of ores: massive, disseminated and veinlet-disseminated. The two former ore types were formed by a liquation process, whereas the latter was associated with fluid-induced selective metasomatic replacement of metamorphosed wall rocks. One of the major ore minerals in all ore types is chalcopyrite. In this study, we determined concentrations of trace elements in this mineral using laser ablation inductively coupled plasma mass spectrometry. It appeared that standard geochemical tools, such as plotting the data in the form of diagrams of normalized concentrations, binary and ternary plots, do not allow one to distinguish chalcopyrite from visually and genetically different ore types. In contrast, more advanced statistical methods such as cluster analysis show different groupings of elements for each ore type. Based on the element clustering, a classification tree was suggested, which allowed for the differentiation of massive, disseminated and veinlet-disseminated ore types of the Oktyabrsk deposit by Se, Te, Cd and Pb concentrations in chalcopyrite with a success rate of 86%. The general feature is that chalcopyrite of veinlet-disseminated ore is poorer in these elements compared to chalcopyrite of the two other ore types. Chalcopyrite of massive ore is poorer in Se and Te when compared to chalcopyrite of disseminated ore. Full article

T cells engineered with chimeric antigen receptors (CARs) show great promise in the treatment of some cancers. Modifying T cells to express CARs generally relies on T-cell transduction using viral vectors carrying a transgene, resulting in semi-random DNA integration within the T-cell genome. While this approach has proven successful and is used in generating the Food and Drug Administration (FDA, USA) approved B-lymphocyte antigen CD19-specific CAR T cells, it is possible the transgene could integrate into a locus that would lead to malignant transformation of the engineered T cells. In addition, manufacturing viral vectors is time-consuming and expensive. One way to overcome these challenges is site-specific gene integration, which can be achieved through clustered regularly interspaced short palindromic repeat (CRISPR) mediated editing and non-viral DNA, which serves as a template for homology-directed repair (HDR). This non-viral gene editing approach provides a rapid, highly specific, and inexpensive way to engineer T cells. Here, we describe an optimized protocol for the site-specific knock-in of a large transgene in primary human T cells using non-viral double stranded DNA as a repair template. As proof-of-principle, we targeted the T-cell receptor alpha constant (TRAC) locus for insertion of a large transgene containing green fluorescence protein (GFP) and interleukin-15 (IL-15). To optimize the knock-in conditions we tested template DNA concentration, homology arm length, cell number, and knock-in efficiency over time. We then applied these established guidelines to target the TRAC or interleukin-13 (IL-13) locus for the knock-in of synthetic molecules, such as a CAR, bispecific T-cell engager (BiTE), and other transgenes. While integration efficiency depends on the targeted gene locus and selected transgene, this optimized protocol reliably generates the desired insertion at rates upwards of 20%. Thus, it should serve as a good starting point for investigators who are interested in knocking in transgenes into specific loci. Full article

In the Bazhenov Formation, framboidal clusters and nodular pyrite formed in the dysoxic–anoxic interface within organic-rich sediments. Some nodule-like pyritized bituminous layers and pyrite nodules are similar to pyritized microbial mat fragments by the typical fine laminated structure. Framboidal pyrite of the Bazhenov Formation is enriched in redox-sensitive elements such as Mo, V, Au, Cu, Pb, Ag, Ni, Se, and Zn in comparison with the host shales and nodular pyrite. Nodular pyrite has higher concentrations of As and Sb, only. Strong positive correlations that can be interpreted as nano-inclusions of organic matter (Mo, V, Au), sphalerite (Zn, Cd, Hg, Sn, In, Ga, Ge), galena (Pb, Bi, Sb, Te, Ag, Tl), chalcopyrite (Cu, Se) and tennantite (Cu, As, Sb, Bi, Te, Ag, Tl) and/or the substitution of Co, Ni, As and Sb into the pyrite. On the global scale, pyrite of the Bazhenov Formation is very similar to pyrite from highly metalliferous bituminous black shales, associated, as a rule, with gas and oil-and-gas deposits. Enrichment with Mo and lower Co and heavy metals indicate a higher influence of seawater during formation of pyrite from the Bazhenov Formation in comparison to different styles of ore deposits. Transitional elements such as Zn and Cu in pyrite of the Bazhenov Formation has resulted from either a unique combination of the erosion of Cu–Zn massive sulfide deposits of the Ural Mountains from one side and the simultaneous manifestation of organic-rich gas seep activity in the West Siberian Sea from another direction. Theoretical electronic spectra and natural circular dichroism (CD) spectra of (+)-(S,S)-bis(2-methylbutyl)chalcogenides, Ch[CH2CH(CH3)C2H5]2 (Ch = S, Se, and Te), were calculated by the symmetry adapted cluster (SAC) and SAC-configuration interaction (SAC-CI) methods. Whereas the calculated CD spectrum for each stable conformation itself did not reproduce the corresponding experimental one, their Boltzmann-averaged spectra showed good agreement with the experimental results. We provided the assignment for each spectral band according to our calculation results. For the telluride compound, temperature dependence of the CD spectra was experimentally observed due to variation in the Boltzmann factor, and our calculations reproduced it qualitatively. The spectral features that we could not reproduce can be attributed to triplet transitions through the spin-orbit interaction effects as well as accuracy incompleteness on the calculation conditions.
