In dynamic equilibria, benzylsilicon pyridine-2-olate (BnSi(pyO)3, L) and benzaldehyde react with addition of the Si(pyO) moieties to the PhHC=O carbonyl group and formation of compounds BnSi(–O–C(H,Ph)–N(2-pyridone))x(pyO)3−x (L, L’’, and L’’’, for x = 1, 2, 3, respectively). Addition of CuCl to a solution containing L, L, L’’, and L’’’ results in the formation of BnSi(pyO)3CuCl (LCuCl), shifting the equilibrium towards L with liberation of benzaldehyde. In THF as a solvent, the reaction of L in the presence of excess CuCl affords the complex LCuClCuCl. Upon dissolving in chloroform, it transforms into LCuCl with precipitation of CuCl. The solid state structure of LCuClCuCl features both the monomeric complex with CuClCuCl pattern and a dimer thereof with CuClCu(Cl)2CuClCu pattern and a central Cu2Cl2 four-membered ring. This dimer of LCuClCuCl is the first crystallographically characterized representative of this Cu(I)-only Cu4Cl4 motif. The reaction of LCuCl and silver tosylate (AgOTos) in THF affords LCuOTos with precipitation of AgCl, whereas LAgOTos was obtained from L and AgOTos. In the crystal structure, LAgOTos features tetracoordinate Ag(I) in a distorted tetrahedral AgN3O coordination sphere and a short Ag···Si trans-annular contact (3.3245(7) Å). 109Ag NMR spectroscopy indicates a change in the coordination in solution, with δ 109Ag = +551 and +419 ppm in the solid and in CDCl3 solution, respectively. In combination, 29Si NMR spectroscopy indicates changes in the Si coordination sphere, with δ 29Si = −74.2 and −66.5 ppm in the solid and in CDCl3 solution, respectively. Conversion of LAgOTos with tetraethylammonium chloride results in the precipitation of AgCl with release of L. Full article

In this work, we present new methods of synthesis of different molecules including a 2-pyridone nucleus. First, we prepared a series of 1H-free 2-pyridones and N-alkyl 2-pyridones from ethyl cyanoacetate, aromatic aldehydes, various acetophenone derivatives and ammonium acetate or diamino-alkane. These molecules have served as building blocks that, in conjunction with acyl chloride derivatives, glycoside derivatives, etc. have resulted in various heterocyclic hybrid structures carrying a 2-pyridone ring. Moreover, based on the cyano group reactivity of the 2-pyridone ring, we synthesized 5-pyridone 1H-tetrazole in a single step by a cycloaddition reaction [3 + 2] between 3-cyano-2-pyridone nitriles and sodium azide in the presence of metal-free L-proline. We present a systematic review of the methods developed for the synthesis of the aromathecin family of compounds (benz[6,7]indolizino[1,2-b]quinolin-11(13H)-ones) and their derivatives. These methods can be broadly classified into four categories based on the construction of pentacyclic structures: Category 1: by constructing a pyridone moiety (D-ring) on the pyrroloquinoline ring (A/B/C-ring), Category 2: by constructing a pyridine moiety (B-ring) on the pyrroloisoquinolone ring (C/D/E-ring), Category 3: by constructing an indolizidinone moiety (C/D-ring) in a tandem reaction, and Category 4: by constructing a pyrrolidine moiety (C-ring) on the isoquinolone ring (D/E-ring). Five undescribed pentaketide derivatives, (R)-6,8-dihydroxy-4,5-dimethyl-3-methylidene-3,4-dihydro-1H-2-benzopyran-1-one (1), [(3S,4R)-3,8-dihydroxy-6-methoxy-4,5-dimethyl-1-oxo-3,4-dihydro-1H-isochromen-3-yl]methyl acetate (2), (R)-5, 7-dimethoxy-3-((S)-(1-hydroxyethyl)-3,4-dimethylisobenzofuran-1(3H)-one (4b), (S)-7-hydroxy-3-((S)-1-hydroxyethyl)-5-methoxy-3,4-dimethylisobenzofuran 1(3H)-one (5), and a p-hydroxyphenyl-2-pyridone derivative, avellaneanone (6), were isolated together with the previously reported (R)-3-acetyl-7-hydroxy-5-methoxy-3,4-dimethylisobenzofuran-1(3H)-one (3), (R)-7-hydroxy-3-((S)-1-hydroxyethyl)-5-methoxy-3,4-dimethylisobenzofuran-1(3H)-one (4a) and isosclerone (7), from the ethyl acetate extract of a culture of a marine sponge-derived fungus, Hamigera avellanea KUFA0732. The structures of the undescribed compounds were elucidated using 1D and 2D NMR, as well as high-resolution mass spectral analyses. The absolute configurations of the stereogenic carbons in 1, 4b, 5, and 6 were established by X-ray crystallographic analysis. The absolute configurations of C-3 and C-4 in 2 were determined by ROESY correlations and on the basis of their common biosynthetic origin with 1. The crude fungal extract and the isolated compounds 1, 3, 4b, 5, 6, and 7 were assayed for their growth inhibitory activity against various plant pathogenic fungi viz. Alternaria brassicicola, Bipolaris oryzae, Colletotrichum capsici, C. gloeosporiodes, Curvularia oryzae, Fusarium semitectum, Lasiodiplodia theobromae, Phytophthora palmivora, Pyricularia oryzae, Rhizoctonia oryzae and Sclerotium rolfsii. Cannabinoid Receptor 2 (CB2) is a promising target for treating inflammatory diseases. We designed derivatives of 3-carbamoyl-2-pyridone and 1,8-naphthyridin-2(1H)-one-3-carboxamide CB2-selective agonists with reduced lipophilicity. The new compounds were measured for their affinity (radioligand binding) and ability to elicit cyclic adenosine monophosphate (cAMP) signalling and β-arrestin-2 translocation with temporal resolution (BRET-based biosensors). For the 3-carbamoyl-2-pyridone derivatives, we found that modifying the previously reported compound UOSS77 (also known as S-777469) by appending a PEG2-alcohol via a 3-carbomylcyclohexyl carboxamide (UOSS75) lowered lipophilicity, and preserved binding affinity and signalling profile. The 1,8-naphthyridin-2(1H)-one-3-carboxamide UOMM18, containing a cis configuration at the 3-carboxamide cyclohexyl and with an alcohol on the 4-position of the cyclohexyl, had lower lipophilicity but similar CB2 affinity and biological activity to previously reported compounds of this class. Relative to CP55,940, the new compounds acted as partial agonists and did not exhibit signalling bias. Interestingly, while all compounds shared similar temporal trajectories for maximal efficacy, differing temporal trajectories for potency were observed. Consequently, when applied at sub-maximal concentrations, CP55,940 tended to elicit sustained (cAMP) or increasing (arrestin) responses, whereas responses to the new compounds tended to be transient (cAMP) or sustained (arrestin). In future studies, the compounds characterised here may be useful in elucidating the consequences of differential temporal signalling profiles on CB2-mediated physiological responses. Full article

Eight new 4-hydroxy-2-pyridone alkaloids arthpyrones D–K (18), along with two known analogues apiosporamide (9) and arthpyrone B (10), were isolated from a deep-sea-derived fungus Arthrinium sp. UJNMF0008. The structures of the isolated compounds were elucidated on the basis of spectroscopic methods with that of 1 being established by chemical transformation and X-ray diffraction analysis. Compounds 1 and 2 bore an ester functionality linking the pyridone and decalin moieties first reported in this class of metabolites, while 3 and 4 incorporated a rare natural hexa- or tetrahydrobenzofuro[3,2-c]pyridin-3(2H)-one motif. Compounds 36 and 9 exhibited moderate to significant antibacterial activity against Mycobacterium smegmatis and Staphylococcus aureus with IC50 values ranging from 1.66–42.8 μM, while 9 displayed cytotoxicity against two human osteosarcoma cell lines (U2OS and MG63) with IC50 values of 19.3 and 11.7 μM, respectively. Substitution of methanol in [Zn(quin)2(CH3OH)2] (quin denotes an anionic form of quinoline-2-carboxylic acid, also known as quinaldinic acid) with pyridine (Py) or its substituted derivatives, 3,5-lutidine (3,5-Lut), nicotinamide (Nia), 3-hydroxypyridine (3-Py-OH), 3-hydroxymethylpyridine (3-Hmpy), 4-hydroxypyridine (4-Py-OH) and 4-hydroxymethylpyridine (4-Hmpy), afforded a series of novel heteroleptic complexes with compositions [Zn(quin)2(Py)2] (1), [Zn(quin)2(3,5-Lut)2] (2), [Zn(quin)2(Nia)2]·2CH3CN (3), [Zn(quin)2(3-Py-OH)2] (4), [Zn(quin)2(3-Hmpy)2] (5), [Zn(quin)2(4-Pyridone)] (6) (4-Pyridone = a keto tautomer of 4-hydroxypyridine), and [Zn(quin)2(4-Hmpy)2] (7). In all reactions, the {Zn(quin)2} structural fragment with quinaldinate ions bound in a bidentate chelating manner retained its structural integrity. With the exception of [Zn(quin)2(4-Pyridone)] (6), all complexes feature a six-numbered coordination environment of metal ion that may be described as a distorted octahedron. The arrangement of ligands is trans. The coordination sphere of zinc(II) in the 4-pyridone complex consists of only three ligands, two quinaldinates, and one secondary ligand. The metal ion thereby attains a five-numbered coordination environment that is best described as a distorted square-pyramid (τ parameter equals 0.39). The influence of substituents on the pyridine-based ligand over intermolecular interactions in the solid state is investigated. Since pyridine and 3,5-lutidine are not able to form hydrogen-bonding interactions, the solid state structures of their complexes, [Zn(quin)2(Py)2] (1) and [Zn(quin)2(3,5-Lut)2] (2), are governed by π···π stacking, C–H∙∙∙π, and C–H∙∙∙O intermolecular interactions. With other pyridine ligands possessing amide or hydroxyl functional groups, the connectivity patterns in the crystal structures of their complexes are governed by hydrogen bonding interactions. Thermal decomposition studies of novel complexes have shown the formation of zinc oxide as the end product. Full article
