Measuring limb joint angles is crucial for understanding horse conformation, performance, injury diagnosis, and prevention. While Thoroughbred horses have been extensively studied, local Pakistani breeds (e.g., Morna racehorse) have not received scientific attention. This study aimed to quantify normal angles of limb joints in the Morna breed. Limb joint angles of standing horses (n = 50) were quantified using a measuring tape, height stick, protractor scale, and goniometer. The mean and standard deviation (Mean ± SD) values for the forelimb joint angles were 123.02 ± 3.46° for elbow, 171.52 ± 2.39° for knee, and 147.68 ± 5.11° for fetlock. The mean ± SD values for the hindlimb joint angles were 128.62 ± 4.08° for stifle, 160.40 ± 3.89° for hock, and 155.48 ± 2.68° for fetlock. There was a non-significant (p > 0.05) correlation between horse joint angles and, age, body weight, and body condition score (BCS). The elbow and stifle joint angles of Morna align well with characteristics associated with optimal racing performance. However, the hock and fetlock angles differ from jumping Thoroughbred and French trotters. We recommend further research to examine the conformation of the Morna breed, particularly by measuring the lengths of limb segments and correlating it with joint angles. This may provide valuable insights into individual variations within the breed. Full article

Background: This retrospective study aims to examine the patient demographics, survival rates, and treatment methods for small-cell neuroendocrine carcinoma (SCNEC) and large-cell neuroendocrine carcinoma (LCNEC) of prostate origin while also identifying the main differences between common types of prostate cancer with comparative analysis for survival. Methods: Our analysis utilized the Surveillance, Epidemiology, and End Results database (SEER), and data was collected from 2000–2020. Cox proportional hazards and chi-squared analysis were used for statistical analysis. Results: A total of 718 cases of prostate small and large neuroendocrine carcinoma were identified. The median age was 71.5 years, and the median follow-up was 11.0 years (95% confidence interval (95% CI) = 9.2–12.8). Most patients were over the age of 80 years (33.8%) and Caucasian (74.4%). The overall 5-year survival was 8.0% (95% CI = 6.8–9.2). The 5-year OS for Caucasians was 7.3% (95% C.I. 6.0–8.3). For Black Americans, the 5-year OS was 11.9% (95% C.I. 7.3–16.5). For Hispanics, the 5-year OS was 12.2% (95% C.I. 7.7–16.7). The 5-year cause-specific survival (CSS) was 16.2% (95% CI = 14.3–18.1). For treatment modality, the five-year survival for each were as follows: chemotherapy, 3.5% (95% CI = 2.1–4.9); surgery, 18.2% (95% CI = 13.6–22.8); multimodality therapy (surgery and chemotherapy), 4.8% (95% CI = 1.7–7.9); and combination (chemoradiation with surgery), 5.0% (95% CI = 1.0–9.0). The prognostic nomogram created to predict patient survivability matched the findings from the statistical analysis with a statistical difference found in race, income, housing, stage, and nodal status. The nomogram also indicated a slight increase in mortality with tumors of greater size. This analysis showed a slight increase in mortality for patients of Asian race. In addition, there was a significant increase in death for patients with stage 3 tumors, as well as patients who underwent surgery and radiation. Furthermore, we performed propensity score matching for survival differences, and no survival difference was found between SCNEC and LCNEC. Conclusions: Asian patients, larger tumor size, and distant disease were associated with worse long-term clinical outcomes. By leveraging insights from registry-based studies, clinicians can better strategize treatment options, improving patient outcomes in this challenging oncology arena. Full article

Ras-related Rap1A GTPase is implicated in pancreas β-cell insulin secretion and is stimulated by the cAMP sensor Epac2, a guanine exchange factor and activator of Rap1 GTPase. In this study, we examined the differential proteomic profiles of pancreata from C57BL/6 Rap1A-deficient (Null) and control wild-type (WT) mice with nanoLC-ESI-MS/MS to assess targets of Rap1A potentially involved in insulin regulation. We identified 77 overlapping identifier proteins in both groups, with 8 distinct identifier proteins in Null versus 56 distinct identifier proteins in WT mice pancreata. Functional enrichment analysis showed four of the eight Null unique proteins, ERO1-like protein β (Ero1lβ), triosephosphate isomerase (TP1), 14-3-3 protein γ, and kallikrein-1, were exclusively involved in insulin biogenesis, with roles in insulin metabolism. Specifically, the mRNA expression of Ero1lβ and TP1 was significantly (p < 0.05) increased in Null versus WT pancreata. Rap1A deficiency significantly affected glucose tolerance during the first 15–30 min of glucose challenge but showed no impact on insulin sensitivity. Ex vivo glucose-stimulated insulin secretion (GSIS) studies on isolated Null islets showed significantly impaired GSIS. Furthermore, in GSIS-impaired islets, the cAMP-Epac2-Rap1A pathway was significantly compromised compared to the WT. Altogether, these studies underscore an essential role of Rap1A GTPase in pancreas physiological function. Full article

Dentin pulp has a complex function as a major unit in maintaining the vitality of teeth. In this sense, the Wnt/β-Catenin pathway has a vital part in tooth development, maintenance, repair, and regeneration by controlling physiological activities such as growth, differentiation, and migration. This pathway consists of a network of proteins, such as Wnt signaling molecules, which interact with receptors of targeted cells and play a role in development and adult tissue homeostasis. The Wnt signals are specific spatiotemporally, suggesting its intricate mechanism in development, regulation, repair, and regeneration by the formation of tertiary dentin. This review provides an overview of the recent advances in the Wnt/β-Catenin signaling pathway in dentin and pulp regeneration, how different proteins, molecules, and ligands influence this pathway, either upregulating or silencing it, and how it may be used in the future for clinical dentistry, in vital pulp therapy as an effective treatment for dental caries, as an alternative approach for root canal therapy, and to provide a path for therapeutic and regenerative dentistry. Background: Neuroendocrine carcinomas (NECs) of the tubular gastrointestinal tract (GI-NECs) are rare and associated with worse clinical outcomes. This population-based study aims to highlight key demographics, clinicopathological factors, and survival outcomes in the US population. Methods: Data from 10,387 patients with GI-NECs were extracted from the Surveillance, Epidemiology, and End Result (SEER) database from 2000 to 2020. Results: Most patients were >40 years old at the time of presentation with a median age of 63 years old, with almost equal ethnic distribution per US population data. The most common primary tumor site was the small intestine (33.6%). The metastatic spread was localized in 34.8%, regional in 27.8%, and distant in 37.3% of cases, and the liver was the most common site of metastasis (19.9%) in known cases of metastases. Most NEC patients underwent surgery, presenting the highest 5-year overall survival of 73.2% with a 95% confidence interval (CI) (95% CI 72.0–74.4%), while chemotherapy alone had the lowest 5-year survival of 8.0% (95% CI 6.4–10.0%). Compared to men, women had a superior 5-year survival rate of 59.0% (95% CI 57.6–60.5%). On multivariate analysis, age > 65 (HR 2.49, 95% CI 2.36–2.54%, p ≤ 0.001), distant metastasis (HR 2.57, 95% CI 2.52–2.62%, p ≤ 0.001), tumor size > 4 mm (HR 1.98, 95%, CI 1.70–2.31%, p ≤ 0.001), esophageal (HR 1.49, 95% CI 0.86–2.58%, p ≤ 0.001), transverse colon (HR 1.95, 95% CI 1.15–3.33%, p ≤ 0.01), descending colon (HR 2.12, 95% CI 1.12, 3.97%, p = 0.02) anorectal sites, and liver or lung metastases were associated with worse survival. Surgical intervention and tumors located in the small intestine or appendix showed a better prognosis. Conclusion: GI-NECs are a group of rare malignancies associated with a poor prognosis. Therefore, epidemiological studies analyzing national databases may be the best alternative to have a more comprehensive understanding of this condition, assess the impact of current practices, and generate prognosis tools. Since the Palaeozoic era, fire as a potent driver of environmental changes, has dramatically shaped the terrestrial ecosystems. Fire affects soil structure and composition, which in turn affects the floral diversity of an area. This research work aims to examine the impact of fire on vegetation and the physicochemical nature of the soil in fire-affected and fire-free sites across the Mahaban and the surrounding forests, Swabi District, Khyber Pakhtunkhwa, Pakistan. Quadrat quantitative ecological techniques were used for vegetation sampling in fire-free and fire-affected sites. In total, 219 plant species belonging to 173 genera and 70 families were recorded. Among the 219 plant species, 173 species were recorded from fire-free sites and the remaining 122 species were from fire-affected sites. The incidence of fire results in elevated organic matter, nitrogen, phosphorus, and lower calcium carbonate concentrations in the soil. The greatest species richness and evenness were observed across the fire-free sites. Our study concludes that the influence of edaphic and topographic factors on species richness varies between fire-affected and fire-free sites. Fire has significantly altered the nutrient availability in the studied region, and this is confirmed by soil analysis and vegetation research. It is suggested that further research in the field of fire ecology can produce valuable insights. Full article

The composition of microbial communities and the functioning of ecosystems are greatly influenced by the nutrient inputs. Despite this fact, our knowledge regarding the impact of phosphorus (P) inputs on soil P availability and microbial community structures in subtropical acidic soils remains limited. We hypothesized that diverse P inputs, incubation temperatures, and soil types could significantly alter soil P availability and microbial communities. To address this gap, we conducted a laboratory incubation experiment, investigating the effects of biochar and inorganic P amendments on soil available P, soil pH, acid phosphatase enzymes, and bacterial abundance. We employed two different incubation temperatures (15 °C and 25 °C) using acidic paddy soil and red soil from the subtropical Southern China region. Our results indicate a notable increase in soil pH, reaching 37% and 39% at 15 °C and 40% and 40.6% at 25 °C, respectively, following the application of biochar and inorganic P amendments in paddy soil. In the case of red soil, we observed pH increases of 41% and 43% at 15 °C and 44% and 45% at 25 °C after the application of biochar and inorganic P amendment, respectively. The addition of inorganic P amendment resulted in the highest available P contents in paddy soil, reaching 111.47 mg/kg at 15 °C and 100.17 mg/kg at 25 °C, respectively. However, Proteobacteria decreased after inorganic P addition, which showed that P might not be the only limiting nutrient for various bacterial communities. Bacterial diversity and richness indices were found to be higher after biochar application in both soils. Gemmatimonadetes, Acidobacteria, and Actinobacteria were found to be strongly influenced by incubation temperatures, whereas most of the top abundant bacterial phyla, such as Gemmatimonadetes, Actinobacteria, Chloroflexi, Acidobacteria, Planctomycetes, Firmicutes, Patescibacteria, and Bacteroidetes, were highly dependent on soil type. At the genus level, various important P solubilizing genera (Pseudomonas, Bradyrhizobium, Streptomyces jietaisiensis, Massilia) significantly increased after biochar and inorganic P addition in both soils. The correlation analysis proved that P-solubilizing genera were significantly associated with changes in soil pH, as well as soil available P after biochar and inorganic P addition. Conclusively, in a short-term incubation experiment, inorganic P amendment greatly increased the soil pH and available phosphorus contents compared to biochar and control treatments; however, the microbial community was observed to be strongly associated with biochar application, soil type, and incubation temperature. To date, no study has reported the anticoccidial effect of lemon peel powder in broilers. Coccidiosis, caused by Eimeria species, is the prevalent enteric parasitic disease in poultry. Although certain chemical drugs have been used for their control, concerns regarding drug residues and the development of resistance in chickens have arisen among consumers. In this study, a total of 300 Ross 308 broiler chicks were randomly allocated into five groups (five equal replicates of 12 animals). The first group served as the control and did not receive any specific treatment (NC). The second group, referred to as the positive control (PC) group, was deliberately exposed to Eimeria tenella. The third group was challenged with E. tenella and also received treatment with amprolium (1 g/kg) and was designated as AT. The fourth and fifth groups were challenged with E. tenella and simultaneously supplemented with lemon peel powder at a dosage of 3 g/kg (LPP3) and 6 g/kg (LPP6). Sporulated E. tenella oocysts (5 × 104/mL bird) on day 22 of the experiment were administered to the infected broiler chickens. The results indicated that in comparison with the NC, all Eimeria-treated birds exhibited significantly (p < 0.05) lower growth performance. However, a notable improvement was observed when infected birds also received a supplement of LPP3 and LPP6 in their feed. Both LPP3 and LPP6 supplementation significantly (p < 0.05) reduced mortality, lesion scores, and oocyst per gram (OPG) of feces compared with the PC group. Additionally, the histological features of the cecum revealed that villus height, villus width, and crypt depth were partially restored under supplementation with LPP3 and LPP6 in the infected birds. Overall, the results demonstrate that Eimeria-infected birds supplemented with LPP3 and LPP6 exhibited improved growth performance, reduced OPG, lowered intestinal coccidiosis lesion scores, and enhanced intestinal histological features. Mobile edge computing networks (MECNs) based on hierarchical cloud computing have the ability to provide abundant resources to support the next-generation internet of things (IoT) network, which relies on artificial intelligence (AI). To address the instantaneous service and computation demands of IoT entities, AI-based solutions, particularly the deep reinforcement learning (DRL) strategy, have been intensively studied in both the academic and industrial fields. However, there are still many open challenges, namely, the lengthening convergence phenomena of the agent, network dynamics, resource diversity, and mode selection, which need to be tackled. A mixed integer non-linear fractional programming (MINLFP) problem is formulated to maximize computing and radio resources while maintaining quality of service (QoS) for every user’s equipment. We adopt the advanced asynchronous advantage actor-critic (A3C) approach to take full advantage of distributed multi-agent-based solutions for achieving energy efficiency in MECNs. The proposed approach, which employs A3C for computing offloading and resource allocation, is shown through numerical results to significantly reduce energy consumption and improve energy efficiency. This method’s effectiveness is further shown by comparing it to other benchmarks. Full article

Thermal stresses due to long running of spark ignition engine often results in wear and tear of cylinder near the top dead center (TDC). These high thermal stresses at TDC arise due to the high temperature gradient during spark ignition. This situation eventually decreases the life and efficiency of an engine. In this study, the numerical and analytical analysis was carried out on 1298 cc in line four stroke spark ignition (SI) engine having a power output of 63 kW to drop the peak temperature at TDC. to reduce the peak value of temperature, square pin fins were used on the surface of engine cylinder wall near TDC. A parametric study is performed to get an optimal solution for removal of the peak temperature load at TDC. The results showed that the fins with dimension of 4 × 4 × 4 mm3 along with uniform spacing of 2 mm provide the optimum solution. It has been observed that the peak temperature at TDC dropped down considerably from 160 °C to 133 °C (a percentage reduction of 16.87%) for the pin fins case as compared to without the fin case. Furthermore, the heat transfer effectiveness for the optimum case was calculated as 3.32, whereas for numerical and analytical study it was calculated as 3.43. The error recorded between both the values was limited to 3.2%. Full article

Demographics and Clinicopathologic Profile of Pulmonary Sarcomatoid Carcinoma with Survival Analysis and Genomic Landscape

Background: Pulmonary sarcomatoid carcinoma (PSC) is a rare subtype of non-small cell lung cancer (NSCLC) with an aggressive clinical nature and poor prognosis. With novel targeted therapeutics being developed, new ways to effectively treat PSC are emerging. In this study, we analyze demographics, tumor characteristics, treatment modalities, and outcomes of PSC and genetic mutations in PSC. Methods: Data from the Surveillance, Epidemiology, and End Results (SEER) database were reviewed to analyze cases of pulmonary sarcomatoid carcinoma from 2000 to 2018. The molecular data with the most common mutations in PSC were extracted from the Catalogue Of Somatic Mutations in Cancer (COSMIC) database. Results: A total of 5259 patients with PSC were identified. Most patients were between 70 and 79 years of age (32.2%), male (59.1%), and Caucasian (83.7%). The male-to-female ratio was 1.45:1. Most tumors were between 1 and 7 cm in size (69.4%) and poorly differentiated (grade III) (72.9%). The overall 5-year survival was 15.6% (95% confidence interval (95% CI) = 14.4–16.9)), and the cause-specific 5-year survival was 19.7% (95% CI = 18.3–21.1). The five-year survival for those treated with each modality were as follows: chemotherapy, 19.9% (95% CI = 17.7–22.2); surgery, 41.7% (95% CI = 38.9–44.6); radiation, 19.1% (95% CI = 15.1–23.5); and multimodality therapy (surgery and chemoradiation), 24.8% (95% CI = 17.6–32.7). On multivariable analysis, age, male gender, distant stage, tumor size, bone metastasis, brain metastasis, and liver metastasis were associated with increased mortality, and chemotherapy and surgery were associated with reduced mortality (p < 0.001). The best survival outcomes were achieved with surgery. The most common mutations identified in COSMIC data were TP53 31%, ARID1A 23%, NF1 17%, SMARCA4 16%, and KMT2D 9%. Conclusions: PSC is a rare and aggressive subtype of NSCLC, usually affecting Caucasian males between 70 and 79. Male gender, older age, and distant spread were associated with poor clinical outcomes. Treatment with surgery was associated with better survival outcomes. Full article

Heat loss through the building envelope comprises air leaks through the cracks and largely through the windows, which is the weakest link of the thermal envelope. Therefore, it is necessary to devise a systematic approach to analyze the rightful selection of glass for buildings. The investigation is to analyze the energy-saving potential of different glasses and their comparisons to the initial capital cost to find the payback time in terms of energy saving by using two different types of equipment. The quantitative simulation study was completed on the Hourly Analysis Program (HAP) to analyze the annual energy consumption of the HVAC system for seven glasses and two types of chilled water equipment. The results show that the performance glasses with a tint had better efficiency in terms of energy saving, with a payback time of 3–7 months. A comparison of all glasses illustrated that float glass contributes the most to the total cooling load among all glasses, which were 5.04%, 5.7%, 7.6%, and 8.9% for the N, S, E, and W orientations, respectively. Moreover, the lowest contribution of glass to the total cooling load was given by tinted double-glazed glass, which was 2%, 2.3%, 3.0%, and 3.01% for N, S, E, and W orientations, respectively. Background: Salivary gland neoplasms are uncommon in both pediatric and adult populations. Mucoepidermoid carcinoma (MEC) is one of the most common salivary gland tumors and usually presents with atypical clinical features. This study sought to evaluate the demographic and clinical factors affecting outcomes in adults and pediatric populations with MEC that could be used to risk stratification for treatment selection and clinical trial enrollment. Methods: Data on 4507 MEC patients were extracted from the Surveillance Epidemiology and End Result (SEER) database (2000–2018). Patients aged ≤ 18 years were classified into the pediatric population, and those older than 18 years were placed in the adult group. Kaplan–Meier survival curves were created to analyze survival probabilities for various independent factors. Results: The pediatric population comprised 3.7% of the entire cohort, with a predominance of females (51.5%), while the adult population constituted 96.3% of the cohort, with a predominance of female patients (52.2%). Caucasians were the predominant race overall (75.3%), while more African Americans were seen in the pediatric group. In tumor size of <2 cm overall, poorly differentiated tumors with higher metastasis rates were observed more in adults (11.3% and 9.3%) than in the pediatric population (3.0% and 4.8%, p < 0.05). Surgical resection was the most common treatment option (53.9%), making up 63.6% of the pediatric and 53.5% of the adult groups. A combination of surgical resection and radiation was used in 29.8% of the entire cohort while a combination of surgical resection, radiation, and chemotherapy made up only 3.2%. The pediatric group had a lower overall mortality rate (5.5%) than the adult group (28.6%). Females had a higher 5-year survival rate in comparison to males (86.5%, and 73.7%, respectively). Surgical resection led to a more prolonged overall survival and 5-year cancer-specific survival (98.4% (C.I, 93.7–99.6) in the pediatric group and 88.8% (C.I, 87.5–90.0) in the adult group), respectively. Metastasis to the lung, bone, brain, and/or liver was found to have significantly lower survival rates. Multivariate analysis demonstrated that adults (hazard ratio [HR] = 7.4), Asian or Pacific Islander (HR = 0.5), male (HR = 0.8), poorly differentiated histology (HR = 3.8), undifferentiated histology (HR = 4.5), regional spread (HR = 2.1), and distant spread (HR = 3.2) were associated with increased mortality (p < 0.05). Conclusions: Mucoepidermoid carcinoma of the salivary glands primarily affects Whites and is more aggressive in adults than in the pediatric population. Even with surgical resection, the overall survival is poor in the adult population as compared to its pediatric counterparts. Advanced age, larger tumor size, male sex, and lymph node invasion are associated with increased mortality. Full article

Crop improvement is the fundamental goal of plant biologists, and genetic diversity is the base for the survival of plants in nature. In this study, we evaluated 20 wheat lines for morphological and genetic diversity using eight simple sequence repeats markers from Wheat Microsatellite Consortium (WMC). Morphologically, variations were observed among all of the different wheat lines for the studied trait except for single spike weight. The highest values for different agronomic traits were recorded for the different wheat lines. The maximum days to heading were recorded for Borlaug-16 (128.3 ± 2.52 days). Similarly, days to maturity were recorded and were highest in Markaz-19 (182.3 ± 5.13 days), followed by Borlaug-16 (182.0 ± 4.58 days). The highest plant height was observed for Zincol-16 (122.3 ± 2.51 cm), followed by Markaz-19 (120.0 ± 14.79 cm) and Borlaug-16 (119.7 ± 6.8 cm). The productivity measured by 100-grain weight was highest in the case of Zincol-16 (84.0 ± 7.5 g). In contrast, wheat lines Shahkar, Sehar, and Farid-6 showed the lowest values for the traits tested. The results of genetic diversity revealed a total number of 16 alleles at eight SSR markers with an average of 2.00 ± 0.534 alleles per locus. Out of eight SSR markers, one marker (WMC105) was monomorphic, and six were dimorphic, showing two alleles at each locus. The maximum number of alleles (3) was observed for marker WMC78, in which genotypes AC and AA were predominantly found in high-yielding lines Borlaug-2016 and Zincol-2016 that were distantly related to other varieties. Zincol-2016 was also agronomically distinct from the rest of the 19 wheat lines. The results obtained from this study may be of importance for the scientific community to further explore the underlying genetic polymorphism associated high yielding varieties using marker-assisted selection for sustainable agriculture. Kari sheep inhabiting the Chitral district of Pakistan show variation in gestation length. In this study, we have analyzed the genetic differences between the three subtypes of Kari sheep (based on variation in gestation length) using microsatellite markers. Kari sheep samples were collected from their breeding tract and were characterized for gestation length and genetic diversity using microsatellite markers. A total of 78 Kari ewes were grouped into three categories based on gestation length (GL), i.e., Kari-S (with a shorter GL), Kari-M (with a medium GL), and Kari-L (with a longer GL). DNA from these samples was used to amplify 31 ovine-specific microsatellite loci through PCR. Of the total 78 Kari specimens, 24 were grouped in Kari-S (GL = 100.7 ± 1.8), 26 were from the Kari-M subtype (GL = 123.1 ± 1.0), and 28 were Kari-L (GL = 143.8 ± 1.5). Microsatellite analysis revealed an association of genotypes at two marker sites (MAF214 and ILSTS5) with variation in GL. A total of 158 alleles were detected across the 22 polymorphic loci with an average of 7.18 alleles per locus. Unique alleles were found in all three subtypes. The highest number of unique alleles was observed in Kari-L (15), followed by Kari-S (10) and Kari-M (8). The results indicated that Kari-S is a genetically distinct subtype (with higher genetic differentiation and distance) from Kari-M and Kari-L. The genetic uniqueness of Kari-S is important for further exploration of the genetic basis for shorter gestation length, and exploitation of their unique values. Full article
