The authors of the article „Precision medicine in the treatment of locally advanced or metastatic urothelial cancer: new molecular targets and pharmacological therapies“  describe contemporary known modalities of treatment of prostatic cancer. All of them are listed in Table 1. The smoking habit as a risk factor is well described as well as other potential carcinogens. Immunotherapy is a new field, as is mainly based on binding to PD or PD-L1. Antibody-Drug Conjugates are well described and are of interest as a promising new weapon in oncotherapy. The conclusions are sound.

The manuscript “Precision medicine in the treatment of locally advanced or met- 2 astatic urothelial cancer: new molecular targets and pharmaco- 3 logical therapies.”, by Vitiello and collaborators deals with an important issue of molecular mechanisms underlying the onset of the urothelial cancer and available the immunotherapies. The paper is well organized and clearly written and is easy to read it. It gives systematically presentation of immunotherapy studies. A manuscript perfectly fits into the scope of the journal. Hence, the manuscript could be interesting for readers and I recommend it for publication after minor revision.

Minor issues for improvement:

- Schematic illustration of immunotherapy approach would be beneficial to increase quality and attract the audience of the paper.

- In many instances proper explanations of the abbreviations are missing.
