The experiments described in the paper are sound and suitable for publication in this journal after revision.
The authors reported the high degree of polymorphism, then how the method will be effective for preservation and mass propagation of this plants with the high chances of variability among regenerants?
Line 50-51: There are high polymorphism in the regenerated plants how it could be clonal. Better to 'in vitro propagation'
Line 133: g.L−1 . Please delete dot between ‘g’ and ‘L’. This is common mistakes throughout the manuscript.
Line: 161: Please write…..‘in a growth chamber’ instead ‘an artificial climate chamber’
Line 166-167: “commercial mix” Please provide the details.
Line 167: 20-30 oC? Why not a set temperature.
Line 184: Please provide the de tails of ‘lysis buffer’.
Lin 190-196: The measurement was made on in vitro plants or regenerated plants established ex vitro?....….please mention here and also provide the age of the plants.
Line 22: What is mean of SEI2 and SEI5 medium. Please describe in the material methods. The same also in the next lines for other media should be described in the material and methods.
Line 328: A. trifoliate should be italics.
Line 368: A. trifoliate should be italics.

The manuscript requires extensive revision. In my opinion, the topic of the paper is not sufficiently justified, based on previous studies performed. It is suggested to review what is pointed out in the statistical analysis epigraph. In my opinion, greater clarity and precision are required in the writing of the manuscript and specifically in the conclusions of the work.
