Round 1

Reviewer 1 Report

The synthesis of the primaquine-linked derivative of the natural compound celastrol reported by Giovanni Ribaudo and Paolo Coghi is interesting, and the characterization of the structure is well done. The computational studies predicting the compound's interaction with the sarco-endoplasmic reticulum (SR) Ca2+ transport ATPase (SERCA) are interesting, as this is an important target for the development of new therapies against arthritis, a disease that continues to grow. However, the article requires corrections to the molecule names, necessitating italicization of the "N" in all names: the title, line 79, line 91, and so on in supporting info. Similarly, the bibliographic section does not follow the journal's requirements at all. Titles should not be italicized, whereas the journal abbreviation is, and the same applies to the volume number, whereas the date should be bolded and DOI. Line 66: a space is missing between “metabolites” and the ref [11]. Same thing on line 75 between (b) and [15]

In view of the interest of this description, I propose to accept this article on condition that it is carefully proofread and corrected according to molbank's guidelines

Dear Editor Prof. Dr. René T. Boeré,

Enclosed please find the revised version of the original research article entitled "“(2R,4aS,6aS,12bR,14aS,14bR)10-hydroxy-N-(4-((6-methoxyquinolin-8-yl)amino)pentyl)-2,4a,6a,9,12b,14a-hexamethyl-11-oxo-1,2,3,4,4a,5,6,6a,11,12b,13,14,14a,14b-tetradecahydropicene-2-carboxamide" by Yuhan Xie, Kuan Hou Li, Qin Wei, Alessandra Gianoncelli, Giovanni Ribaudo and Paolo Coghi for your consideration. We prepared this improved version following the comments that we received from the Reviewer. The modified parts have been highlighted in the files, and we reported a list of changes below.

We believe that this update version of the manuscript would be suitable for publication as an original research article in Molbank.

Should you have any queries, please do not hesitate to contact me at (853) 68145596 or coghips@must.edu.mo. Thank you for your consideration of our work. I look forward to hearing from you.

Yours Sincerely,

A: We would like to thank the Reviewer for the positive comments that we received and for the suggestions that were very useful to improve the quality of our paper.

R: However, the article requires corrections to the molecule names, necessitating italicization of the "N" in all names: the title, line 79, line 91, and so on in supporting info.

A: Thanks for the comment. The name has been corrected in the files.

R: Similarly, the bibliographic section does not follow the journal's requirements at all. Titles should not be italicized, whereas the journal abbreviation is, and the same applies to the volume number, whereas the date should be bolded and DOI.

A: Thanks for the comment. The bibliographic section was corrected accordingly.

R: Line 66: a space is missing between “metabolites” and the ref [11]. Same thing on line 75 between (b) and [15]

A: Thanks for the comment. We corrected these typos accordingly.

A: We thank the Reviewer for this comment. The article has been double-checked for errors and carefully proofread. In addition, we improved the quality of Figure 2: to facilitate the interpretation of the artwork, we included a slightly closer zoom so the labels can be read more easily.
