This is a very interesting review paper on glyoxalase modulators and their clinical applications. The manuscript is comprehensive (128 references in total) and well written. Tables are very helpful. Graphical part is also attractive. I have no major comments/concerns.

Minor points:

The authors should add a paragraph on future perspective.

Limitations should be also more highlighted as a separate paragraph.

AUTHORS’ RESPONSE: We have added two paragraphs – one on future perspective on each of Glo1 inducers and Glo1 inhibitors (Page 19, lines 3 – 17 and Page 20, lines 7 – 13).
