The collection of such a serial data is not easy.  For this reason, the paper deserves such a credit for carrying out such a study.  Most of analyses are carried out in a very reasonable way.  I agree with the finding of the results may be important to the disease progression.

Very well written scientific manuscript . Proposes solid data for underlying mechanisms with genetic polymorphisms and pathogenesis of CRC. Text editing may need to be done - page 11, line 16 - incomplete sentence - no other statistically. This manuscript will be a major addition to the journal

The study "Intake of red and processed meat, use of non-steroid anti-inflammatory drugs, genetic variants and risk of colorectal cancer; a prospective study of the Danish “Diet, Cancer and Health” cohort" is of a significant interest for public health. The strengths of the present manuscript are the large sample size, the clear presentation of limitations of the study and the possible mechanistic explanations provided in the discussions.

The discussion could be enriched by a short overarching statement about the involvement of TP53 rs1042522 in other cancer types.
