Small extracellular vesicles (sEVs) derived from mesenchymal stem cells have emerged as promising therapeutic agents in regenerative dermatology. This study evaluated the safety and efficacy of Bio-Pulsed avian mesenchymal stem cell-derived sEVs (AMSC-sEVs), topically applied for hair follicle stimulation and skin rejuvenation. Two prospective, single-arm clinical trials were conducted: one involving 30 participants using a hair ampoule over 60 days, and the other involving 30 participants applying a facial essence for 28 days. Objective measurements demonstrated significant improvements in the anagen/telogen hair ratio, reduced shedding, increased collagen density, and reduced wrinkle depth and pigmentation. Small RNA sequencing and qPCR profiling confirmed that Bio-Pulsed AMSC-sEVs were enriched with regenerative microRNAs, such as miR-21-5p and miR-199a-5p, associated with anti-inflammatory and anti-aging effects. No adverse events were reported. These findings suggest that Bio-Pulsed AMSC-sEVs may offer a safe, non-invasive, and cell-free approach to enhance skin and hair regeneration in human subjects. Unveiling the Evolution of MWC 728: Non-Conservative Mass Transfer in an FS CMa Binary

We combine corrected Gaia DR3 astrometry with non-conservative MESA modelling to retrace the evolution of the FS-CMa binary MWC 728. The revised parallax sets the distance at d=1.2±0.1 kpc, leading—after Monte-Carlo error propagation—to luminosities of log(L/L)acc=2.6±0.1 and log(L/L)don=1.5±0.1, corresponding to the accretor and donor, respectively. A fiducial binary track that starts with Mdon=3.6±0.1M, Macc=1.8±0.1M, and P0=21.0±0.2 d reproduces the observations provided the Roche-lobe overflow, which is moderately non-conservative: only 39% of the transferred mass is retained by the accretor, while the remainder leaves the system via (i) a fast isotropic wind from the donor (α=0.01), (ii) isotropic re-emission near the accretor (β=0.45), and (iii) outflow into a circumbinary torus (δ=0.15, lever arm γ=1.3). These channels remove sufficient angular momentum to expand the orbit to the observed Pobs=27.5±0.1 d while sustaining the dusty circumbinary outflow. At t223 Myr, the model matches every current observable: Mdon=1.30±0.05M, Macc=2.67±0.05M, mass ratio q=2.0±0.1, and an ongoing transfer rate of M˙(1±0.3)×106Myr1. MWC 728 thus serves as a benchmark intermediate-mass binary for testing how non-conservative outflows regulate angular-momentum loss and orbital growth. Full article

The incidental finding of small renal masses in CT and MRI examinations can present a diagnostic challenge. Renal cell carcinoma (RCC) and angiomyolipoma (AML) are the most common incidental malignant and benign renal lesions but may present with similar US features. US elastography is a non-invasive technique that can assess tissue elasticity, has shown promising results in many clinical settings, and could be able to differentiate between benign and malignant renal lesions based on tissue stiffness. The purpose of this article is to systematically review the applications of US elastography in the characterization of solid renal masses and to derive and compare the summary estimates of different stiffness values across different lesion subtypes. In December 2022, a systematic search was carried out on the MEDLINE (PubMed) and EMBASE databases to retrieve studies on the application of US elastography in the characterization of solid renal masses. After article selection by three researchers, 14 studies entered qualitative synthesis. A total of 1190 patients were included, and the elastography data of 959 lesions were examined: 317/959 (33%) benign and 642/959 (67%) malignant. Among the malignancies, 590 (91%) were RCC, whereas, among the 317 benign lesions, 244 (77%) were AML. All lesions were classified using a histopathological (biopsy or operative specimen) or imaging (US follow-up/CT/MRI) reference standard. After data extraction and methodological quality evaluation, quantitative synthesis was performed on 12 studies, 4 using strain elastography (SE) and 8 using shear wave elastography (SWE), with single- and double-arm random-effects meta-analyses. Lesion stiffness measured with SE was available in four studies, with an RCC strain ratio higher than the AML strain ratio both in an indirect comparison (Cochran’s Q test p = 0.014) and in a direct comparison (p = 0.021). Conversely, the SWE measurements of RCC and AML stiffness did not significantly differ either at an indirect comparison (p = 0.055) or direct comparison (p = 0.114). Full article

Background: Survivin is an inhibitor of apoptosis protein (IAP), encoded by the Baculoviral IAP Repeat Containing 5 (BIRC5) gene located on q arm (25.3) on chromosome 17. It is expressed in various human cancers and involved in tumor resistance to radiation and chemotherapy. The genetic analysis of the BIRC5 gene and its protein survivin levels in buccal tissue related to oral squamous cell carcinoma (OSCC) in South Indian tobacco chewers has not been studied. Hence, the study was designed to quantify survivin in buccal tissue and its association with pretreatment hematological parameters and to analyze the BIRC5 gene sequence. Method: In a single centric case control study, buccal tissue survivin levels were measured by ELISA. A total of 189 study subjects were categorized into Group 1 (n = 63) habitual tobacco chewers with OSCC, Group 2 (n = 63) habitual tobacco chewers without OSCC, and Group 3 (n = 63) healthy subjects as control. Retrospective hematological data were collected from Group 1 subjects and statistically analyzed. The BIRC5 gene was sequenced and data were analyzed using a bioinformatics tool. Results: Survivin protein mean ± SD in Group 1 was (1670.9 ± 796.21 pg/mL), in Group 2 it was (1096.02 ± 346.17 pg/mL), and in Group 3 it was (397.5 ± 96.1 pg/mL) with significance (p < 0.001). Survivin levels showed significance with cut-off levels of absolute monocyte count (AMC), neutrophil/lymphocyte ratio (NLR), and lymphocyte/monocyte ratio (LMR) at (p = 0.001). The unique variants found only in OSCC patients were T → G in the promoter region, G → C in exon 3, C → A, A → G, G → T, T → G, A → C, G → A in exon 4, C → A, G → T, G → C in the exon 5 region. Conclusions: The tissue survivin level increased in OSCC patients compared to controls; pretreatment AMC, LMR, and NLR may serve as add-on markers along with survivin to measure the progression of OSCC. Unique mutations in the promoter and exons 3–5 were observed in sequence analysis and were associated with survivin concentrations. Full article

Background: The electrical properties of cells and tissues in relation to energy exposure have been investigated, presenting their resistance and capacitance characteristics. The dielectric response to radiofrequency fields exhibits polarization heterogeneity under pathological conditions. The aim of the study was to analyze the differences in changes in resistance and capacitance measurements in the range from 1 kHz to 1 MHz, combined with an assessment of the correlation between the results of electrical impedance spectroscopy (EIS) and inflammatory activation. Methods: In the prospective study, EIS was performed on the non-dominant arm in 29 male patients (median (Q1–Q3) age of 69 (65–72)) with complex coronary artery disease and 10 male patients (median (Q1–Q3) age of 66 (62–69)) of the control group. Blood samples were collected for inflammatory index analysis. Results: The logistic regression analysis revealed a negative correlation with inflammatory indexes, including neutrophil to lymphocyte ratio (NLR) in the CAD group in the frequency of 30 kHz (p = 0.038, r = −0.317) regarding EIS resistance measurements and a positive correlation in CAD group in the frequency of 10 kHz (p = 0.029, r = −0.354) regarding EIS capacitance. Conclusions: The bioelectric characteristics of peripheral tissues measured by resistance and capacitance in EIS differ in patients with coronary artery disease and in the control group. Electrical impedance spectroscopy reveals a statistically significant correlation with inflammatory markers in patients with CAD. Full article

The widely distributed ray-finned fish genus Carassius is very well known due to its unique biological characteristics such as polyploidy, clonality, and/or interspecies hybridization. These biological characteristics have enabled Carassius species to be successfully widespread over relatively short period of evolutionary time. Therefore, this fish model deserves to be the center of attention in the research field. Some studies have already described the Carassius karyotype, but results are inconsistent in the number of morphological categories for individual chromosomes. We investigated three focal species: Carassius auratus, C. carassius and C. gibelio with the aim to describe their standardized diploid karyotypes, and to study their evolutionary relationships using cytogenetic tools. We measured length (q+plength) of each chromosome and calculated centromeric index (i value). We found: (i) The relationship between q+plength and i value showed higher similarity of C. auratus and C. carassius. (ii) The variability of i value within each chromosome expressed by means of the first quartile (Q1) up to the third quartile (Q3) showed higher similarity of C. carassius and C. gibelio. (iii) The fluorescent in situ hybridization (FISH) analysis revealed higher similarity of C. auratus and C. gibelio. (iv) Standardized karyotype formula described using median value (Q2) showed differentiation among all investigated species: C. auratus had 24 metacentric (m), 40 submetacentric (sm), 2 subtelocentric (st), 2 acrocentric (a) and 32 telocentric (T) chromosomes (24m+40sm+2st+2a+32T); C. carassius: 16m+34sm+8st+42T; and C. gibelio: 16m+22sm+10st+2a+50T. (v) We developed R scripts applicable for the description of standardized karyotype for any other species. The diverse results indicated unprecedented complex genomic and chromosomal architecture in the genus Carassius probably influenced by its unique biological characteristics which make the study of evolutionary relationships more difficult than it has been originally postulated. Background: The loss of the Y chromosome in various malignant diseases has been described previously. There are no reliable information on the actual frequency, significance and homogeneity of Y chromosome loss (LoY) in esophageal adenocarcinoma (EAC). Methods: 400 male EAC including lymph-node metastases were analyzed with commercially available Y chromosome specific fluorescence in-situ probes. The results were correlated with molecular and immunohistochemical markers and clinicopathological aspects. Results: The entire cohort (n = 400) showed a singular LoY of one chromosome arm in 1.0% (q-arm) and 2.8% (p-arm), complete LoY in 52.5%. LoY was strongly associated with shortened overall-survival (OS). Patients with preserved Y chromosome had a median OS of 58.8 months, patients with LoY an OS of 19.4 months (p < 0.001). Multivariate analysis showed LoY as an independent prognostic marker with a hazard ratio of 1.835 (95% CI 1.233–2.725). LoY correlated with TP53 mutations (p = 0.003), KRAS amplification (p = 0.004), loss of ARID1a (p = 0.045) and presence of LAG3 (p = 0.018). Conclusions: Loss of the Y chromosome is a very common phenomenon in EAC. The LoY is heterogeneously distributed within the tumor, but corresponding lymph node metastases frequently show homogeneous LoY, indicating a selection and metastasizing advantage with poor prognosis. To date, the male predominance of EAC (7–9:1) is unclear, so genetic explanatory models are favored. The LoY in EAC may be biologically and functionally relevant and additional genomic or functional analyses are needed. Full article
