In Brazil, Trypanosoma vivax was identified between 1970 and 1984 in buffalo, cattle, and sheep in northern Brazil. In the 1990s, the disease was diagnosed in the Pantanal of Mato Grosso. Currently, the disease is considered in enzootic stability in both regions. From 2006 to 2018, outbreaks of the disease associated with the presence of horseflies (Tabanidae) and stable flies (Stomoxys calcitrans) or transmitted by needle and syringe reuse were diagnosed in sheep, goats, and cattle during the rainy season in the semiarid region of Northeast Brazil, a region considered to have enzootic instability. Since 2013, numerous outbreaks have been diagnosed associated with oxytocin inoculation to induce milk letdown in dairy cows. Most outbreaks occur in cattle transported from areas of enzootic instability to areas of stability, or vice versa. Serological, parasitological, and molecular techniques have identified T. vivax in asymptomatic animals in all Brazilian regions. For disease prophylaxis, it is important not to reuse syringes and needles when administering medication and to avoid transporting animals during periods of insect abundance. It is also necessary to avoid the use of oxytocin or adopt safe injection practices, using disposable or disinfected needles and syringes only for cows with ineffective milk ejection. Full article

Breastfeeding is universally recognized for its extensive health benefits for both infants and mothers. However, for some women, the experience of breastfeeding can be complicated by intense negative emotional and physical reactions, including phenomena such as dysphoric milk ejection reflex and breastfeeding aversion/agitation. This review explores the neuroendocrine underpinnings of these conditions, emphasizing the interplay between oxytocin, prolactin, and dopamine. Oxytocin, traditionally viewed as a hormone promoting bonding and emotional regulation, can paradoxically provoke a stress response in vulnerable individuals. Prolactin, a key hormone for lactation and maternal behaviors, is implicated in stress resilience and mood regulation, but its dysregulation may contribute to depressive states. Dopamine, critical for reward processing and emotional stability, may underlie the acute emotional dysregulation seen in dysphoric milk ejection reflex. Together, disturbances in these neurohormonal systems may explain the aversive emotional experiences during breastfeeding. An improved understanding of these mechanisms offers critical insights into maternal mental health during lactation and underscores the importance of supportive clinical approaches for affected women. This retrospective cohort study examined the association between delayed milk ejection (DME), defined as bimodal milk flow, and milk yield in dairy cows milked with an automated milking system (AMS). Additionally, we identified risk factors for DME. Using data from a farm milking approximately 1350 cows, we analyzed 689,484 individual milking records and 194,142 daily cow observations over 350 days with generalized linear mixed models. Cows with DME generally had higher daily milk yields, regardless of lactation number. However, first-lactation cows early in lactation and older cows (≥third lactation) late in lactation produced less milk when experiencing DME. In contrast to the higher daily milk yield, cows produced less milk per milking when experiencing delayed milk ejection. However, more frequent milkings contributed to higher daily milk yield, even with more instances of delayed milk ejection. Risk factors for DME included lactation number, stage of lactation, milking frequency, and milking interval. These findings underscore the complexity of DME in AMS and suggest that optimizing individualized milking protocols could improve milk yield efficiency. Understanding the interplay of cow characteristics and milking management may enhance AMS performance and dairy herd productivity. Full article

Vacuum-assisted and forceps-assisted vaginal births are associated with higher rates of formula supplementation and shorter breastfeeding duration compared to unassisted vaginal births; however, the reasons for this are unclear. Factors such as maternal knowledge, partner support, and parity significantly influence breastfeeding initiation and duration. The prevalence of perineal trauma, neonatal and maternal birth complications, and decreased birth satisfaction is higher after assisted births and may also impact breastfeeding outcomes. Given the limited research on the specific effects of different vaginal birth modes on breastfeeding, this study aimed to examine women’s experiences of establishing breastfeeding after unassisted, vacuum-assisted, and forceps-assisted vaginal birth. A mixed-methods study design was employed using an anonymous online questionnaire, which included binary, multiple choice, and open-ended questions, and Likert scale items. Using social media, we recruited Australian women who had an unassisted, vacuum-assisted, or forceps-assisted birth within the last year. Details of participant demographics, breastfeeding history, initiation and establishment, postpartum mobility, and pain ratings were recorded. Additionally, qualitative data on postpartum recovery and breastfeeding support were analysed using an inductive thematic analysis framework. A total of 565 women were recruited between May and June 2024, of which 488 responses were retained for analysis. Thematic analysis of the qualitative responses identified four central themes that defined women’s experiences of establishing breastfeeding and were similar between unassisted or assisted vaginal birth modes: Experience of Care, Environment, Expectations, and Health Complications. A range of both positive and negative experiences of breastfeeding support, environmental factors, and expectations of the realities of breastfeeding impacted women’s experiences. For many women, various maternal and/or newborn health issues, nipple pain, and latching difficulties made breastfeeding more difficult. Commercial milk formula supplementation during the hospital stay was more prevalent after a forceps-assisted birth when compared to unassisted vaginal birth (41% vs. 17%, respectively; p < 0.001). Further, during the first two weeks at home, commercial milk formula supplementation was more prevalent after both forceps-assisted (26%) and vacuum-assisted (23%) births than after unassisted vaginal birth (8%, p < 0.001). Pain ratings in the early days following birth and in the first two weeks at home were significantly higher for the forceps-assisted group than for the other vaginal birth modes (p ≤ 0.005). Women that had an unassisted vaginal birth with an intact perineum had the lowest pain ratings in the early days and weeks after birth, while pain ratings were similar between women that had a vacuum-assisted birth and those who had an unassisted vaginal birth with a perineal tear or episiotomy (p = 0.05). Early commercial milk formula supplementation is associated with shorter breastfeeding duration, while postpartum pain is known to impede maternal mobility and may partially inhibit the milk ejection reflex, potentially negatively impacting breastfeeding and increasing formula use. Therefore, women who have an instrumental assisted vaginal birth, particularly those who have a forceps-assisted birth, are at greater risk of suboptimal breastfeeding outcomes including short durations of exclusive and any breastfeeding. Improvements to early postpartum pain management, breastfeeding education, and the judicious use of commercial milk formula may improve breastfeeding and subsequent maternal and health outcomes after instrument-assisted vaginal birth. Background: Whilst it is inconvenient and time-intensive, predominantly (PP) and exclusively pumping (EP) mothers rely on breast expression to provide milk for their infants and to ensure continued milk supply, yet these populations are poorly understood. Methods: We assessed and characterised Western Australian PP mothers (n = 93) regarding 24 h milk production (MP) and infant milk intake and demographics, perinatal complications and breastfeeding difficulties, the frequencies of which were compared with published general population frequencies. Pumping efficacy and milk flow parameters during a pumping session in PP mothers (n = 32) were compared with those that pump occasionally (reference group, n = 60). Results: PP mothers had a higher frequency of pregnancy complications and breastfeeding difficulties than the general population. Exclusive pumping did not impact the 24 h MP and the amount of milk available to the infant. PP mothers had more milk ejections, longer active flow duration and lower milk removal efficacy ratios; however, responsiveness to pump (time to first milk ejection), total flow duration, time to stop pumping, percentage of available milk removed and comfort parameters were not different to the reference group. Conclusions: Despite the reported challenges, when an effective pump is used, predominant or exclusive pumping does not negatively affect the 24 h MP and the amount of milk available to the infant. Increasing awareness of the impacts of perinatal and breastfeeding complications on women may assist health professionals in early identification of mothers at high risk for early cessation of breastfeeding, escalating early postpartum intervention and targeted breastfeeding support. The primary objective was to investigate the association between delayed milk ejection (DME) and the average milk flow rate, milking unit-on time, and duration in a low milk flow rate in Holstein dairy cows in a large dairy herd with suboptimal premilking teat stimulation. Our second objective was to study the association between peak lactation milk yield and the occurrence of DME. This longitudinal field study was conducted at a 4300-cow dairy farm with a thrice-daily milking schedule over a 1-week period. We analyzed data from 61,677 cow milking observations from 2937 cows. Delayed milk ejection was defined as present if the 30–60 s milk flow rate was ≤3.1 kg/min. The mean average milk flow rate (MAMF, kg/min), mean milking unit-on time (MMUT, s), and mean duration of a low milk flow rate (MLMF, s) were calculated as the mean values from the 21 milking observations. General linear multivariable models revealed associations of DME with MAMF, MMUT, and MLMF. A multivariable ordinal logistic regression model revealed an association between peak lactation milk yield and DME. Cows with lower peak lactation milk yield had greater odds of exhibiting a higher frequency level of DME. The observed associations between DME and milking performance indices suggest that DME can negatively affect milking and parlor efficiency. Peak lactation milk yield may serve as a proxy to estimate cows’ risk of recurrent DME. Future research is warranted to test if alleviating DME through, for example, a modified milking routine influences the milking performance indices described herein. Full article

Oxytocin, a significant pleiotropic neuropeptide, regulates psychological stress adaptation and social communication, as well as peripheral actions, such as uterine contraction and milk ejection. Recently, a Japanese Kampo medicine called Kamikihito (KKT) has been reported to stimulate oxytocin neurons to induce oxytocin secretion. Two-pore-domain potassium channels (K2P) regulate the resting potential of excitable cells, and their inhibition results in accelerated depolarization that elicits neuronal and endocrine cell activation. We assessed the effects of KKT and 14 of its components on a specific K2P, the potassium channel subfamily K member 2 (TREK-1), which is predominantly expressed in oxytocin neurons in the central nervous system (CNS). KKT inhibited the activity of TREK-1 induced via the channel activator ML335. Six of the 14 components of KKT inhibited TREK-1 activity. Additionally, we identified that 22 of the 41 compounds in the six components exhibited TREK-1 inhibitory effects. In summary, several compounds included in KKT partially activated oxytocin neurons by inhibiting TREK-1. The pharmacological effects of KKT, including antistress effects, may be partially mediated through the oxytocin pathway. Full article

The present review aims to analyze the anatomical and physiological characteristics of the mammary gland and udders of water buffalo by making an anatomofunctional comparison with dairy cattle. It will also discuss the knowledge generated around the physiological regulation of milk ejection in the water buffalo. It was found that buffalo’s average udder depth and width is approximately 20 cm smaller than Bos cattle. One of the main differences with dairy cattle is a longer teat canal length (around 8.25–11.56 cm), which highly influences buffalo milking. In this sense, a narrower teat canal (2.71 ± 0.10 cm) and thicker sphincter muscle are associated with needing higher vacuum levels when using machine milking in buffalo. Moreover, the predominant alveolar fraction of water buffalo storing 90–95% of the entire milk production is another element that can be related to the lower milk yields in buffalo (when compared to Bos cattle) and the requirements for prolonged prestimulation in this species. Considering the anatomical characteristics of water buffalo’s udder could help improve bubaline dairy systems. Full article

While considered as hard milkers, both buffaloes and camels are milked with equipment destined for dairy cows based on external morphological similarities with this species. This work aimed to study similarities and differences in milkability traits between Mediterranean buffaloes and Maghrebi she-camels and to evaluate the effect of parity and lactation stage. A total of 422 milk flow curves recorded with an electronic milkmeter (Lactocorder®) for both species were accessed. Milking characteristics including milk yield per milking, peak milk flow, average milk flow, duration of the main milking phase, duration of total milking, duration of various phases of milk flow, lag time and time to milk ejection, stripping yield, overmilking time and incidence of bimodal milk flow curves were evaluated for both species. Results showed that the values of milk yield per milking, duration of the main milking phase and duration of total milking were higher in buffaloes (3.98 ± 0.10 kg; 4.07 ± 0.11 min; 9.89 ± 0.21 min, respectively) compared to camels (3.51 ± 0.08 kg; 3.05 ± 0.09 min; 3.76 ± 0.09 min, respectively). However, camels had significantly higher peak and average milk flow (2.45 ± 0.07 kg/min and 1.16 ± 0.03 kg/min, respectively). Camels took significantly less time for milk ejection to occur. Only 15.49% of recorded curves were bimodal in buffaloes while 34.93% of bimodal curves were recorded for camels. Overmilking was significantly higher in buffaloes (3.64 ± 0.21 min vs. 0.29 ± 0.02 min). Parity and lactation stage had a significant effect on most studied milking traits suggesting the need for some particular practices with primiparous animals and animals at different levels of lactation for both species. Breastfeeding is a key issue found in ancient sources that resonates with public debates today, affecting women in different parts of the world and of all social classes. The aim of this research was to identify breastfeeding narratives in ancient medical and philosophical texts from the 1st to the 6th century CE that address ethical issues in the medical management and social perception of new mothers. We examined 15 literary sources and one funerary inscription on lactation and critically evaluated the ancient idea of the perfect breastfeeding mother versus the non-breastfeeding mother. We then discussed our historical data in terms of objectivity and significance in relation to contemporary attitudes towards motherhood and lactation, e.g., (1) the cult of the perfect, breastfeeding mother in contemporary lactation education and (2) the onset of conditions which may affect normal breastfeeding, such as dysphoric milk ejection reflex (D-MER), breastfeeding aversion response (BAR) or post-partum depression. The analysis of the results showed that in both ancient and contemporary postnatal health care: (1) good mothering is associated with breastfeeding and (2) alternative feeding methods are acknowledged, but never as the best, natural option. Finally, our analysis shows that public health policies on breastfeeding and mothers’ own knowledge of their bodies are contested between nursing theories, social expectations and economic factors. Oxytocin (OXT) is one of the essential hormones in the birth process; however, estradiol, prolactin, cortisol, relaxin, connexin, and prostaglandin are also present. In addition to parturition, the functions in which OXT is also involved in mammals include the induction of maternal behavior, including imprinting and maternal care, social cognition, and affiliative behavior, which can affect allo-parental care. The present article aimed to analyze the role of OXT and the neurophysiologic regulation of this hormone during parturition, how it can promote or impair maternal behavior and bonding, and its importance in lactation in domestic animals. Breastmilk is thought to influence the infant gut by supplying prebiotics in the form of human milk oligosaccharides and potentially seeding the gut with breastmilk microbes. However, the presence of a breastmilk microbiota and origins of these microbes are still debated. As a pilot study, we assessed the microbes present in expressed breastmilk at six-weeks postpartum using shotgun metagenomic sequencing in a heterogenous cohort of women who delivered by vaginal (n = 8) and caesarean delivery (n = 8). In addition, we estimated the microbial load of breastmilk at six-weeks post-partum with quantitative PCR targeting the 16S rRNA gene. Breastmilk at six-weeks postpartum had a low microbial mass, comparable with PCR no-template and extraction controls. Microbes identified through metagenomic sequencing were largely consistent with skin and oral microbes, with four samples returning no identifiable bacterial sequences. Our results do not provide convincing evidence for the existence of a breastmilk microbiota at six-weeks postpartum. It is more likely that microbes present in breastmilk are sourced by ejection from the infant’s mouth and from surrounding skin, as well as contamination during sampling and processing. Full article

The overall objective of this study was to evaluate the effects that milking temperament (MT) of water buffaloes has on milking traits, welfare indicators, and reproductive outputs. The study was performed on 60 multiparous buffalo cows (6.7 ± 0.6 lactations) at the beginning of their lactation (100 days in milk, DIM). Each buffalo cow was scored by two independent observers using a temperament scoring system (1: extremely calm, 2: calm, 3: alert, 4: reactive, and 5: aggressive), and then grouped as ‘calm’ (scores 1, 2, and 3; n = 42) or ‘nervous’ (scores 4 and 5; n = 18). Additionally, the milk yield at 100 DIM (MY), milking speed (MS), calving interval (CI), age at first calving (AFC), body condition score (BCS), animal-based welfare parameters, and infrared thermography data (IRT) were evaluated. The MT significantly influenced the MY (p = 0.0082), with calmer cows outperforming their nervous counterparts. The MS was significantly influenced by the MT (p = 0.0015), with calmer animals having a higher milk ejection rate. The MT of the cows had no influence on the CI, AFC, or BCS. The evidence from this study suggests that the responsiveness of buffalo cows during milking affects their milk yield and milking speed, with no associations being found for reproduction efficiency indicators or animal-based welfare indicators. The present review aims to integrate the anatomical characteristics of the mammary gland and the neurophysiology of milk ejection to understand the milking capacity of the water buffalo. Since one of the main uses of this species is milk production, this article will analyze the controversies on the use of oxytocin as a stimulant during milking as well as the existing alternatives that farmers apply to promote correct stimulation during milk letdown. According to the available literature, the efficiency of the milking process, the quality of the milk, and the health of the animals are elements that require the consideration of species-specific characteristics to enhance the performance of buffaloes. The incorporation of technological innovations and competitive strategies could contribute to a better understanding of water buffalo in the milk industry. Full article
