Avocado fruits are considered unusual because of the large amounts of oil and seven-carbon (7-C) carbohydrates (mannoheptulose and perseitol) in the fruit’s flesh and skin. The fruit may be held on the tree unripe until required for marketing, and in some producing regions, this may extend past the next flowering period. This prolonged period on the tree is associated with increased oil content and decreased 7-C carbohydrates. There has been relatively less research into soluble hexose sugars and starch. In this research, the inter-relationships between fruit maturation, storage, and ripening have been investigated for both 7-C and six-carbon non-structural carbohydrates using ‘Hass’ fruit harvested from the same trees between 11 and 14 months after flowering. Significant differences were identified in both fruit flesh and skin for most compounds, affected by maturity, storage, and ripening. It is concluded that the non-structural carbohydrate composition of ‘Hass’ fruit is variable, with significant changes occurring associated with maturation, storage, and ripening. The compositions of the flesh and skin tissues are not consistently proportionate. Maturation provides the initial baseline composition from which any further change through storage or ripening can occur. The changes with maturation appear to be associated with the tree’s phenology, with tree-to-tree differences in the timing or degree of change. Full article

Background: Unripe avocados (Persea americana) are naturally enriched in mannoheptulose (MH), which is a candidate caloric restriction mimetic. Objectives: To evaluate the effects of a diet supplement made from unripe avocado on glucose tolerance, and cardiometabolic risk factors in free-living nondiabetic adults with obesity. Methods: In a double-blinded, randomised controlled trial, 60 adults (female n = 47, age 48 ± 13 years, BMI 34.0 ± 2.6 kg/m2) were stratified by sex and randomised to avocado extract (AvX, 10 g finely ground, freeze-dried unripe avocado) or placebo (10 g finely ground cornmeal plus 5% spinach powder) daily, for 12 weeks. The primary outcome was a change in glucose area under the curve (AUC) in response to a 75 g oral glucose tolerance test. A post-hoc analysis was subsequently performed in a subgroup with insulin AUC above the median of baseline values after removal of participants >2 SD from the mean. Results: There were no between-group differences in glucose AUC (p = 0.678), insulin AUC (p = 0.091), or cardiovascular outcomes. In the subgroup analysis, insulin AUC was lower in AxV versus placebo (p = 0.024). Conclusions: Daily consumption of unripe avocado extract enriched in MH did not alter glucose tolerance or insulin sensitivity in nondiabetic adults with obesity, but the data provided preliminary evidence for a benefit in insulin AUC in a subgroup of participants with elevated baseline postprandial insulin levels. Beginning at 16 weeks of age and continuing for 44 weeks, male C57BL/6J were fed either a control (CON) diet; a high-fat (HF) diet (60% unsaturated); or the HF diet containing an extract of unripe avocados (AvX) enriched in the 7-carbon sugar mannoheptulose (MH), designed to act as a glycolytic inhibitor (HF + MH). Compared to the CON diet, mice on the HF diet exhibited higher body weights; body fat; blood lipids; and leptin with reduced adiponectin levels, insulin sensitivity, VO2max, and falls from a rotarod. Mice on the HF + MH diet were completely protected against these changes in the absence of significant diet effects on food intake. Compared to the CON diet, oxidative stress was also increased by the HF diet indicated by higher levels of total reactive oxygen species, superoxide, and peroxynitrite measured in liver samples by electron paramagnetic resonance spectroscopy, whereas the HF + MH diet attenuated these changes. Compared to the CON, the HF diet increased signaling in the mechanistic target of the rapamycin (mTOR) pathway, and the addition of the MH-enriched AvX to this diet attenuated these changes. Beyond generating further interest in the health benefits of avocados, these results draw further new attention to the effects of this rare sugar, MH, as a botanical intervention for preventing obesity. Avocado cv. Hass consumption has expanded worldwide given its nutritional, sensory, and functional attributes. In this work, avocado fruit from two harvests was subjected to hydrothermal treatment (38 °C for 1 h) or left untreated (control) and then stored for 30 and 50 days in a controlled atmosphere (4 kPa O2 and 6 kPa CO2 at 7 °C) (HTCA and CA, respectively) with subsequent ripening at ~20 °C. The fruit was evaluated for primary and secondary metabolites at harvest, after storage, and after reaching edible ripeness. A decrease from harvest to edible ripeness in mannoheptulose and perseitol was observed while β-sitosterol, hydrophilic and lipophilic antioxidant activity (H-AOX, L-AOX), abscisic acid, and total phenolics (composed of p-coumaric and caffeic acids such as aglycones or their derivatives) increased. HTCA fruit at edible ripeness displayed higher contents of mannoheptulose, perseitol, β-sitosterol, L-AOX, caffeic acid, and p-coumaric acid derivatives, while CA fruit presented higher contents of α-tocopherol, H-AOX, and syringic acid glycoside for both harvests and storage times. The results indicate that a hydrothermal treatment prior to CA enables fruit of high nutritional value characterized by enhanced content of phenolic compounds at edible ripeness to reach distant markets. The ability of light-emitting diode (LED) light treatment to reduce the anthracnose decay via its eliciting effects and thus induce resistance in the avocado (Persea americana), was investigated in this study to replace the current postharvest fungicide treatment. In experiment 1, the effect of blue or red LED lights (6 h per day) on the incidence of anthracnose in artificially inoculated (Colletotrichum gloesposorioides) and naturally infected avocados (cv. Fuerte and Hass) at 12–14 °C (simulated market shelf) for 4, 8, 14, and 16 days was investigated. In experiment 2, the effect of blue or red LED lights on the induced defence mechanism, fruit metabolites, antioxidant activity, and percentage of fruit reaching ready-to-eat stage was determined. Exposure to red LED light significantly reduced the anthracnose decay incidence in naturally infected cv. Fuerte on day 12 and in cv. Hass on day 16 compared to the prochloraz fungicide treatment by upregulating the PAL genes and maintaining the epicatechin content. Blue LED light accelerated the ripening in both cultivars, probably due to reduced D-mannoheptulose content. Red LED light exposure for 6 h per day and 12 days storage showed potential to replace the prochloraz treatment with improved ascorbic acid content and antioxidant activity. Full article

There is a growing interest in the use of nutraceuticals for weight management in companion animals. The purpose of this study was to determine the effects of mannoheptulose (MH), a sugar in avocados that inhibits glycolysis, on energy metabolism in adult Beagle dogs. The study was a double-blind, randomized controlled trial where dogs were allocated to a control (CON, n = 10, 10.1 ± 0.4 kg) or MH containing diet (168 mg/kg, n = 10, 10.3 ± 0.4 kg). Blood was collected after an overnight fast and 1 h post-feeding (week 12) to determine serum satiety related hormones and biochemistry. Resting and post-prandial energy expenditure and respiratory quotient were determined by indirect calorimetry (weeks 4 and 8). Physical activity was measured using an accelerometer (weeks 3, 7, 11). Body composition was assessed using dual X-ray absorptiometry (week 12). MH significantly (p < 0.05) increased fasting serum glucagon-like peptide-1 and post-prandial serum ghrelin. MH tended (p < 0.1) to increase fasting serum gastric inhibitory peptide and decrease physical activity. Together, these findings suggest that dietary MH has the ability to promote satiation and lowers daily energy expenditure. Full article
