The risk of zoonotic parasitic infections is closely linked to direct and indirect interactions between animals and humans. The mutual coexistence of species in public spaces predisposes individuals to gastrointestinal parasitosis owing to various social and hygienic-sanitary factors. This study aimed to assess the risk of environmental contamination by gastrointestinal parasites in fecal samples collected from parks and public squares in 18 municipalities located in a microregion of the central plateau of Brazil, correlating the collection with the population size of each municipality. We collected 536 soil fecal samples from 117 randomly selected public areas across a 56,111.874 km2 region. Eggs, cysts, and oocysts were detected using the Willis flotation technique and the Hoffman’s spontaneous sedimentation method. The Hoffman’s analysis revealed that 70.3% of the fecal samples were infected across 91.5% of the sampled areas. Identified parasites included the members of the family Ancylostomatidae (56.5%), Toxocara spp. (6.2%), Trichuris spp. (1.7%), Strongyloides spp. (0.2%), Dipylidium caninum (25.8%), Spirometra spp. (0.4%), Taeniidae (0.2%), Platynosomum fastosum (0.6%), Trematoda (0.2%), Giardia spp. (3.2%), Cystoisospora spp. (5.6%), Sarcocystis spp. (0.2%), and Entamoeba spp. (2.4%). The presence of positive fecal samples in public areas and municipalities correlated with populations of up to 10,000 inhabitants (p = 0.023). Areas contaminated with feces were 63.4% more likely to contain parasites than other areas (odds ratio 1.6336). Favorable environmental factors combined with inadequate sanitary management contribute to a high risk of environmental contamination, representing a significant zoonotic potential and highlighting the need for improved public health policies and preventive measures. P23-Specific IgY Significantly Reduces Diarrhea and Oocyst Shedding in Calves Experimentally Infected with Cryptosporidium parvum

Background/Objectives: Cryptosporidium parvum is a zoonotic enteroparasite causing severe diarrhea in newborn calves, leading to significant economic losses in dairy and beef farming. This study aimed to evaluate whether C. parvum p23-specific IgY antibodies could control neonatal calf diarrhea caused by C. parvum. Methods: A recombinant immunogen comprising the p23 protein fused to the antigen-presenting cell homing (APCH) molecule was expressed using the baculovirus system. Hens were immunized with the APCH-p23 immunogen, and the resulting IgY was spray-dried for treatment use. Eight newborn calves were included in the study and received commercial colostrum within the first 12 h of life. Four calves were treated with 20 g of powdered egg containing IgY (p23-specific IgY titer of 256 in milk) twice daily for 7 days. The remaining four calves received regular non-supplemented milk. All calves were orally infected with 6 million oocysts and monitored for 21 days. Results: Calves treated with p23-specific IgY exhibited significantly reduced diarrhea duration (3.5 vs. 7.5 days; p = 0.0397) and oocyst shedding duration (6.50 vs. 12 days; p = 0.0089). In addition, the total number of excreted oocysts, as measured by the change of the area under the curve (AUC), was significantly reduced in the treated group (14.25 vs. 33.45; p = 0.0117). Although the onset of diarrhea was delayed (3.5 to 6.5 days post-infection; p = 0.1840), and diarrhea severity was reduced (24.25 to 17 AUC; p = 0.1236), both parameters were not statistically significant. Conclusions: P23-specific IgY antibodies effectively reduced the C. parvum-induced duration of diarrhea in experimentally infected calves. These findings highlight the potential of this passive treatment as a promising strategy for controlling neonatal calf diarrhea. Full article

The purpose of this study was to evaluate the influence of environmental pollution and the living conditions of indigenous Ecuadorians on the transmission of enteroparasites in an Andean agricultural area located at high altitude. Environmental pollution was recorded after observation in each community. The parasites were identified by microscopic sediment analysis using physiological saline solution from macerated arthropods, washed vegetables, and human stools, utilizing four coproparasitological techniques (direct examination, Kato–Katz, ether concentration, and Ziehl–Neelsen). The results show that the inadequate disposal of human and animal excreta that contaminate soil and water, incorrect food hygiene, inadequate sanitary infrastructure in houses, a lack of animal veterinary care, and rodent proliferation are important reservoirs of zoonotic parasites. The use of excrement as fertilizer increases the number of flies, which act as mechanical vectors, and vegetables grown in areas with disperse infective parasitic forms act as vehicles that are marketed at the local, regional, and international levels. These analyses verify contamination levels of 52.7% in mechanical vectors, 70.6% in vegetables, and 98.2% in human stools. The agricultural communities analyzed maintained poor hygienic–sanitary and environmental conditions, which had a significant influence on the transmission of enteroparasites that affect human health. Asymptomatic carriage of diarrhoea-causing enteric protist parasites in the general population is poorly understood, particularly in medium- to high-income countries. This molecular epidemiological survey investigates the presence, molecular diversity, and household transmission of Giardia duodenalis, Cryptosporidium spp., Blastocystis sp., and Enterocystozoon bieneusi in schoolchildren aged 2–13 years (n = 74) and their legal guardians (n = 6) in Madrid, Spain. Enteroparasite detection and genotyping was conducted in stool samples by molecular (PCR and Sanger sequencing) methods. Potential associations linked to infections were investigated through epidemiological questionnaires. Giardia duodenalis was the most prevalent enteric parasite found (14%, 95% CI: 7.1–23), followed by Blastocystis sp. (10%, 95% CI: 6.2–22) and Cryptosporidium spp. (3.8%, 95% CI: 0.78–11). None of the participants tested positive for E. bieneusi. Sequence analyses revealed the presence of G. duodenalis assemblage B, sub-assemblage BIV in a single child. The three Cryptosporidium isolates obtained were assigned to C. hominis, two of them belonging to the gp60 subtype IbA10G2. Four Blastocystis subtypes were identified including ST2 (38%, 3/8), ST3 (25%, 2/8), ST4 (25%, 2/8), and ST8 (12%, 1/8). All G. duodenalis and Cryptosporidium isolates were detected in children only. Blastocystis ST3 and ST4 were circulating in members of the same household. Blastocystis carriage rates increased with the age of the participants. Presence of diarrhoea-causing enteric protists was common in apparently healthy children. Full article

Infections by the protist enteroparasites Giardia duodenalis, Cryptosporidium spp., and, to a much lesser extent, Blastocystis sp. are common causes of childhood diarrhoea in low-income countries. This molecular epidemiological study assesses the frequency and molecular diversity of these pathogens in faecal samples from asymptomatic schoolchildren (n = 807) and symptomatic children seeking medical attention (n = 286) in Zambézia province, Mozambique. Detection and molecular characterisation of pathogens was conducted by polymerase chain reaction (PCR)-based methods coupled with Sanger sequencing. Giardia duodenalis was the most prevalent enteric parasite found [41.7%, 95% confidence interval (CI): 38.8–44.7%], followed by Blastocystis sp. (14.1%, 95% CI: 12.1–16.3%), and Cryptosporidium spp. (1.6%, 95% CI: 0.9–2.5%). Sequence analyses revealed the presence of assemblages A (7.0%, 3/43) and B (88.4%, 38/43) within G. duodenalis-positive children. Four Cryptosporidium species were detected, including C. hominis (30.8%; 4/13), C. parvum (30.8%, 4/13), C. felis (30.8%, 4/13), and C. viatorum (7.6%, 1/13). Four Blastocystis subtypes were also identified including ST1 (22.7%; 35/154), ST2 (22.7%; 35/154), ST3 (45.5%; 70/154), and ST4 (9.1%; 14/154). Most of the genotyped samples were from asymptomatic children. This is the first report of C. viatorum and Blastocystis ST4 in Mozambique. Molecular data indicate that anthropic and zoonotic transmission (the latter at an unknown rate) are important spread pathways of diarrhoea-causing pathogens in Mozambique. Full article

Enterospora nucleophila is a microsporidian enteroparasite that infects mainly the intestine of gilthead sea bream (Sparus aurata), leading to an emaciative syndrome. Thus far, the only available information about this infection comes from natural outbreaks in farmed fish. The aim of the present study was to determine whether E. nucleophila could be transmitted horizontally using naturally infected fish as donors, and to establish an experimental in vivo procedure to study this host–parasite model without depending on natural infections. Naïve fish were exposed to the infection by cohabitation, effluent, or intubated either orally or anally with intestinal scrapings of donor fish in four different trials. We succeeded in detecting parasite in naïve fish in all the challenges, but the infection level and the disease signs were always milder than in donor fish. The parasite was found in peripheral blood of naïve fish at 4 weeks post-challenge (wpc) in oral and effluent routes, and up to 12 wpc in the anal transmission trial. Molecular diagnosis detected E. nucleophila in other organs besides intestine, such as gills, liver, stomach or heart, although the intensity was not as high as in the target tissue. The infection tended to disappear through time in all the challenge routes assayed, except in the anal infection route. Full article
