Optimizing Rooting and Growth of Salvia rosmarinus Cuttings in Soilless Systems Affected by Growth Regulators

This study investigated how propagation systems, growth regulators, and hormone formulations interactively affect the rooting and subsequent growth of rosemary (Salvia rosmarinus Spenn) cuttings. A three factorial (3 × 2 × 7) experiment was conducted under a fully controlled greenhouse environment, incorporating three soilless propagation systems (mist, float, aeroponics), two rooting hormone formulations (powder and gel-based IBA), and two growth regulators (paclobutrazol and daminozide) at three concentrations each. Significant differences (p < 0.001) were found in shoot height, root length, and number of lateral roots. The float system combined with powder hormone and no retardants achieved the highest shoot height (mean = 16.7 cm), while aeroponics with powder hormone and daminozide 1000 ppm promoted the greatest root branching (mean = 12.2 lateral roots per cutting). Root length was maximized (mean = 15.9 cm) under float systems with daminozide 1000 ppm. High doses of both growth regulators negatively affected all parameters across systems. Post-transplantation monitoring confirmed that cuttings from float and mist systems treated with powder hormone and low or no growth retardants exhibited superior establishment and net growth over 60 days. These findings demonstrate the critical importance of pairing hormone type, regulator concentration, and propagation system, providing actionable protocols for nursery managers aiming to enhance Salvia rosmarinus propagation in commercial practice. In Eastern Europe, the traditional marketing of cyclamen is in the period from the middle of February to the middle of March. The poor light of winter months and the higher number of plants (because of the reduction in heating costs) often result in elongated plants. To avoid this, it is recommended to use plant growth retardants. At the same time, another problem is that flowers do not rise from the level of rosette due to the unfavorable cultivation conditions. This can be solved with growth stimulants. In the experiment, we tested the effect of growth regulators on the growth of a frequently used variety of cyclamen. We used daminozide and paclobutrazol plant growth retardants for height control and gibberellic acid (GA) and benzyladenine (BAP) as growth promoters for increasing the number of flowers and the length of stems. The results show that daminozide and paclobutrazol are both effective for height control in cyclamen production. Full article

The aim of the study was to examine the effects of application of plant-growth regulators (PGR) on the growth of paulownia plants and evaluate their usefulness in paulownia nurseries. The experiment was carried out on the paulownia interspecific hybrid (Paulownia elongata × Paulownia fortunei) ‘Oxytree’. Micropropagated plants grown in pots were sprayed with PGR solutions. Gibberellins (GA3, GA4+GA7), 6-benzyladenine (BA), triclopyr (TPA), ethephon (ETH), daminozide (DA), prohexadione-Ca (PH), and trinexapac-ethyl (TE) were used. They were included in commercial products (Florgib 245 mg dm−3, Gibb plus 11 SL 5 cm3 dm−3, Globaryll 100 SL 1.5 cm3 dm−3, Topper 10 ST 200 mg dm−3, Agrostym 480 SL 2 cm3 dm−3, B-Nine 85 SG 2 g dm−3, Regalis plus 10 WG 2.5 g dm−3, Moddus 250 EC 1 cm3 dm−3, respectively). None of the studied preparations modified positive correlations between the longitudinal vs. transverse growth of stems and growth of leaves. Not only GA3, GA4+GA7, but also BA (cytokinin) and unexpectedly TE (retardant) stimulated stem elongation and thickening. Furthermore, the effect of TE lasted longer than influence of GAx and BA. Leaves of TE-treated plants were distinguished by a higher chlorophyll a/b ratio, and a lower relative chlorophyll content and efficiency of photosystem II (Fv/Fm, Fv/F0). TPA strongly deformed the stems and leaves of liners. Other regulators (ETH, DA, PH) retarded the diameter growth of stems and leaf expansion. Some results obtained by GAx, BA, and especially by TE treatment, might be beneficial for paulownia grown as ornamental and/or timber tree. Full article

Weed control during common buckwheat cultivation is hindered by the crop’s high sensitivity to agrochemicals. This study evaluates whether biostimulants (Asahi SL, Kelpak SL, B-Nine) could reduce the adverse effect of abiotic stress caused by these substances on buckwheat’s vegetation indices and yield. To this end, a four-factor field experiment was performed according to the 3 4−1 Box–Behnken design on chernozem soil with silt texture at the Experimental Station of the Agricultural University of Krakow (Poland, 50°07′ N, 20°04′ E). The results showed that calcium cyanamide fertilization was effective in reducing the abundance of dicotyledonous weeds by 39% and the dry weight of weeds per unit area by 20% relative to ammonium nitrate-fertilized sites. However, the most effective method of weed control was the application of metazachlor together with clomazone. The mixture of these active substances reduced the abundance of monocotyledonous weeds, dicotyledonous weeds, and dry weight of weeds by 83%, 40.5%, and 36.4%, respectively. The use of herbicides adversely affected the leaf area index (LAI). Nitrophenol treatment of buckwheat grown on soil fertilized with calcium cyanamide resulted in increased achene yield and number of seeds per plant compared to ammonium nitrate fertilization. The application of daminozide on chemically protected plants resulted in improved vegetation indices such as normalized difference vegetation index (NDVI) and soil plant analysis development (SPAD) compared to sites not exposed to herbicides. Daminozide, a plant growth regulator, is an effective inhibitor of the Jumonji domain-containing protein (JMJD) histone demethylase. Herein, four ruthenium(II)/rhenium(I)-daminozide conjugates, with molecular formulas [Ru(N-N)2bpy(4-CH2OH-4′-CH2O-daminozide)](PF6)2 (Ru-1/Ru-2) (N-N = 1,10-phenanthroline (phen, in Ru-1) and 4,7-diphenyl-1,10-phenanthroline (DIP, in Ru-2)) and Re(N-N)(CO)3(PyCH2O-daminozide) (Re-1/Re-2) (Py = pyridine, N-N = phen (in Re-1) and DIP (in Re-2)), were synthesized and characterized. Among these complexes, Ru-2 and Re-2 exhibited higher cytotoxicity against tumor cells than cisplatin. Upregulation of H3K9Me3 expression level was found in human cervical cancer cells (HeLa) treated with Ru-2 and Re-2, indicating that these two complexes can inhibit the activity of JMJD histone demethylase. Further investigation revealed that Re-2 can selectively accumulate in the mitochondria of HeLa cells. Both Ru-2 and Re-2 can cause mitochondrial damage, induce apoptosis, and inhibit cell migration and colony formation of HeLa cells. Overall, these complexes exhibit multiple anticancer functions, including inhibiting JMJD, inducing apoptosis, and inhibiting cell invasion, making them promising candidates for anticancer drugs. Early cone formation is a consistent issue in Fraser fir Christmas tree plantations in the eastern United States. Growers must remove cones by hand, resulting in significant labor costs, as cones degrade the aesthetic value of Christmas trees. In this study, we conducted two series of experiments in collaboration with several Christmas tree farms to determine the effectiveness of applying plant growth regulators (PGRs), specifically GA-inhibitors, in reducing cone formation and shoot growth in Fraser fir. In the first experiments we investigated the effectiveness of paclobutrazol, applied as a soil injection or as a foliar spray in reducing cone formation. The highest rate of soil application (300 mL of paclobutrazol per tree) reduced cumulative coning by approximately 38%. Leader growth control of soil applied paclobutrazol increased with application rate. In the second set of experiments, we compared four GA-inhibitors (paclobutrazol, chlormequat chloride, uniconazole-p, daminozide) applied as foliar sprays on coning and leader growth. Overall, paclobutrazol was the most effective compound for reducing coning and shoot growth. The results confirm earlier findings that application of GA-inhibitors can reduce, but not eliminate, coning. Likewise, PGR application can reduce shoot growth and possibly increase tree density but will not eliminate the need for shearing. Full article

Triticum timopheevii Zhuk. is a tetraploid wheat that is utilized worldwide as a valuable breeding source for wheat improvement. Gene-based biotechnologies can contribute to this field; however, T. timopheevii exhibits recalcitrance and albinism in tissue cultures, making this species of little use for manipulation through genetic engineering and genome editing. This study tested various approaches to increasing in vitro somatic embryogenesis and plant regeneration, while reducing the portion of albinos in cultures derived from immature embryos (IEs) of T. timopheevii. They included (i) adjusting the balance between 2,4-D and daminozide in callus induction medium; (ii) cultivation using various darkness/illumination schedules; and (iii) inclusion of additional concentrations of copper ions in the tissue culture medium. We achieved a 2.5-fold increase in somatic embryogenesis (up to 80%) when 50 mg L−1 daminozide was included in the callus induction medium together with 3 mg L−1 2,4-D. It was found that the dark cultivation for 20–30 days was superior in terms of achieving maximum culture efficiency; moreover, switching to light in under 2 weeks from culture initiation significantly increased the number of albino plants, suppressed somatic embryogenesis, and decreased the regeneration of green plants. Media containing higher levels of copper ions did not have a positive effect on the regeneration of green plants; contrarily, the elevated concentrations caused albinism in plantlets. The results and relevant conclusions of the present study might be valuable for establishing an improved protocol for the regeneration of green plants in tissue cultures of T. timopheevii.
