While the Ankom RF system facilitates efficient high-throughput in vitro fermentation studies, its high cost and limited flexibility constrain its broader applicability. To address these limitations, we developed and validated a low-cost, modular gas monitoring system (FerME), assembled from commercially available components. To evaluate its performance and reproducibility relative to the Ankom RF system (Ankom Technology, Macedon, NY, USA), in vitro rumen fermentation experiments were conducted under strictly controlled and identical conditions. Whole rumen contents were collected approximately 2 h post-feeding from individual mid- or late-lactation dairy cows and immediately transported to the laboratory. Each fermenter received 50 mL of processed rumen fluid, 100 mL of anaerobically prepared artificial saliva buffer, and 1.2 g of the donor cow’s diet. Bottles were sealed with the respective system’s pressure sensors, flushed with CO2, and incubated in a 50 L water bath maintained at 39 °C. FerME (New Taipei City, Taiwan) and Ankom RF fermenters were placed side-by-side to ensure uniform thermal conditions. To assess the effect of filter bag use, an additional trial employed Ankom F57 filter bags (Ankom Technology, Macedon, NY, USA; 25 μm pore size). Trial 1 revealed no significant differences in cumulative gas production, volatile fatty acids (VFAs), NH3-N, or pH between systems (p > 0.05). However, the use of filter bags reduced gas output and increased propionate concentrations (p < 0.05). Trial 2, which employed filter bags in both systems, confirmed comparable results, with the FerME system demonstrating improved precision (CV: 4.8% vs. 13.2%). Gas composition (CH4 + CO2: 76–82%) and fermentation parameters remained consistent across systems (p > 0.05). Importantly, with 12 pressure sensors, the total cost of FerME was about half that of the Ankom RF system. Collectively, these findings demonstrate that FerME is a reliable, low-cost alternative for real-time rumen fermentation monitoring and could be suitable for studies in animal nutrition, methane mitigation, and related applications. Background: The relationship between sports participation and food preferences in adults, as well as the influence of gender, is still unclear. Objective: The objective of this study was to investigate the association between sports participation and individual food preferences and to explore potential gender differences among sports participants in a large group of Italian adults. Methods: This cross-sectional study involved 2665 adults (aged ≥ 18 years) who lead normal lives and underwent a routine lifestyle and dietary assessment at a clinical centre specialising in nutrition, metabolic health, and lifestyle counselling in Rome. Participants completed an online questionnaire on food preferences (19 foods) and sports practice. Multivariate logistic regression models, adjusted for age, sex, and smoking, were used to assess associations. Results: Sports participation was defined as engaging in structured physical activity at least once per week and was reported by 53.5% of subjects (men: 60.1%; women: 49.0%; p < 0.0001). After adjustment, active individuals were significantly more likely to prefer plant-based drinks, low-fat yoghurt, fish, cooked and raw vegetables, fruit, whole grains, tofu, and dark chocolate (all p < 0.05) and less likely to prefer cow’s milk (p = 0.018). Among sport participants, males were more likely to prefer meat (general, white, red, processed) and eggs, while females preferred plant-based drinks. No significant gender differences were observed for dairy products, legumes, or fish. Differences in food preferences were also observed according to the type of sport, with bodybuilders showing higher preference for tofu and dark chocolate. The strongest associations were found in the 25–44 age group. Conclusions: Sports participation is independently associated with specific food preferences, characterised by greater preference for plant-based and fibre-rich foods, and gender differences in food choices persist even among active adults. These findings highlight the need to consider both sports participation and gender when designing nutritional interventions. Full article

This study aimed to evaluate daily patterns of hourly milking frequency (MF) in dairy cows milked with an automatic milking system (AMSs), considering the effects of season, parity order (PO), days in milk (DIM), and milk yield (MY). A retrospective longitudinal study was conducted on a commercial dairy farm in southern Brazil over one year using data from 130 Holstein cows and 94,611 milking events. MF data were analyzed using general linear models. Overall, hourly MF followed a consistent daily pattern, with peaks between 4:00 and 11:00 a.m. and between 2:00 and 6:00 p.m., regardless of season, PO, DIM, or MY category. MF was higher in primiparous (2.84/day, p = 0.0013), early-lactation (<106 DIM; 3.00/day, p < 0.0001), and high-yielding cows (≥45 L/day; 3.09/day, p < 0.0001). High-yielding cows also showed sustained milking activity into the late nighttime. Although seasonal and individual factors significantly affected MF, they had limited influence on the overall daily distribution of milkings. These results suggest stable behavioral patterns within the specific AMS management conditions observed in this study and suggest that adjusting milking permissions and feeding strategies based on cow characteristics may improve system efficiency. Heat stress in dairy cows, caused by high temperature and humidity during summer, has led to significant declines in milk production and severe economic losses for farms. Exosomes—extracellular vesicles carrying bioactive molecules—are critical for intercellular communication and immunity but remain understudied in heat-stressed Holstein cows. In this study, we extracted exosomes from three heat-stressed (HS) cows and three non-heat-stressed (Ctr) cows and employed proteomics to analyze plasma exosomes. We identified a total of 28 upregulated and 18 downregulated proteins in the HS group compared to the control group. Notably, we observed a significant upregulation of key protein groups, including cytoskeletal regulators, signaling mediators, and coagulation factors, alongside the downregulation of HP-25_1. These differentially expressed proteins demonstrate strong potential as heat stress biomarkers. GO and KEGG analyses linked the differentially expressed proteins to actin cytoskeleton regulation and endoplasmic reticulum pathways. Additionally, protein–protein interaction (PPI) analysis revealed the PI3K-Akt signaling pathway as a central node in the cellular response to heat stress. These findings establish plasma exosomes as valuable biospecimens, provide valuable insights into the molecular mechanisms of heat stress response, and may contribute to the development of precision breeding strategies for enhanced thermal resilience in dairy herds. Background: Against the backdrop of the large-scale and intensive development of the livestock industry, enhancing the reproductive efficiency of cattle has become a crucial factor in industrial development. Holstein cows, as the most predominant dairy cattle breed globally, are characterized by high milk yield and excellent milk quality. However, their reproductive efficiency is comprehensively influenced by a variety of complex factors, and improving their reproductive performance faces numerous challenges. The ovary, as the core organ of the female reproductive system, plays a decisive role in embryonic development and pregnancy maintenance. It is not only the site where eggs are produced and developed but it also regulates the cow’s estrous cycle, ovulation process, and the establishment and maintenance of pregnancy by secreting various hormones. The normal functioning of the ovary is crucial for the smooth development of the embryo and the successful maintenance of pregnancy. Methods: Currently, traditional sequencing technologies have obvious limitations in deciphering ovarian function and reproductive regulatory mechanisms. To overcome the bottlenecks of traditional sequencing technologies, this study selected Holstein cows as the research subjects. Ovarian samples were collected from one pregnant and one non-pregnant Holstein cow, and single-nucleus transcriptome sequencing technology was used to conduct an in-depth study on the ovarian cells of Holstein cows. Results: By constructing a cell type-specific molecular atlas of the ovaries, nine different cell types were successfully identified. This study compared the proportions of ovarian cell types under different physiological states and found that the proportion of endothelial cells decreased during pregnancy, while the proportions of granulosa cells and luteal cells increased significantly. In terms of functional enrichment analysis, oocytes during both pregnancy and non-pregnancy play roles in the “cell cycle” and “homologous recombination” pathways. However, non-pregnant oocytes are also involved in the “progesterone-mediated oocyte maturation” pathway. Luteal cells during pregnancy mainly function in the “cortisol synthesis and secretion” and “ovarian steroidogenesis” pathways; non-pregnant luteal cells are mainly enriched in pathway processes such as the “AMPK signaling pathway”, “pyrimidine metabolism”, and “nucleotide metabolism”. Cell communication analysis reveals that there are 51 signaling pathways involved in the pregnant ovary, with endothelial cells, granulosa cells, and luteal cells serving as the core communication hubs. In the non-pregnant ovary, there are 48 pathways, and the interaction between endothelial cells and stromal cells is the dominant mode. Conclusions: This study provides new insights into the regulatory mechanisms of reproductive efficiency in Holstein cows. The differences in the proportions of ovarian cell types, functional pathways, and cell communication patterns under different physiological states, especially the increase in the proportions of granulosa cells and luteal cells during pregnancy and the specificity of related functional pathways, indicate that these cells play a crucial role in the reproductive process of cows. These findings also highlight the importance of ovarian cells in pathways such as “cell cycle”, “homologous recombination”, and “progesterone-mediated oocyte maturation”, as well as the cell communication mechanisms in regulating ovarian function and reproductive performance. Leptospirosis is a globally significant zoonosis affecting animal health, productivity, and the environment. While typically associated with tropical climates, its persistence in semi-arid regions such as La Laguna, Mexico—characterized by low humidity, high temperatures, and limited water sources—remains poorly understood. Although these adverse environmental conditions theoretically limit the survival of Leptospira, high livestock density and synanthropic reservoirs (e.g., rodents) may compensate, facilitating transmission. In this cross-sectional study, blood sera from 445 dairy cows (28 herds: 12 intensive [MI], 16 semi-intensive [MSI] systems) were analyzed via microscopic agglutination testing (MAT) against 10 pathogenic serovars. Urine samples were cultured for active Leptospira detection. Risk factors were assessed through epidemiological surveys and multivariable analysis. This study revealed an overall apparent seroprevalence of 27.0% (95% CI: 22.8–31.1), with significantly higher rates in MSI (54.1%) versus MI (12.2%) herds (p < 0.001) and an estimated true seroprevalence of 56.3% (95% CI: 50.2–62.1) in MSI and 13.1% (95% CI: 8.5–18.7) in MI herds (p < 0.001). The Sejroe serogroup was isolated from urine in both systems, confirming active circulation. In MI herds, rodent presence (OR: 3.6; 95% CI: 1.6–7.9) was identified as a risk factor for Leptospira seropositivity, while first-trimester abortions (OR:10.1; 95% CI: 4.2–24.2) were significantly associated with infection. In MSI herds, risk factors associated with Leptospira seropositivity included co-occurrence with hens (OR: 2.8; 95% CI: 1.5–5.3) and natural breeding (OR: 2.0; 95% CI: 1.1–3.9), whereas mastitis/agalactiae (OR: 2.8; 95% CI: 1.5–5.2) represented a clinical outcome associated with seropositivity. Despite semi-arid conditions, Leptospira maintains transmission in La Laguna, particularly in semi-intensive systems. The coexistence of adapted (Sejroe) and incidental serogroups underscores the need for targeted interventions, such as rodent control in MI systems and poultry management in MSI systems, to mitigate both zoonotic and economic impacts. Due to the feeding system (high-concentrate diet) during the early lactation stage, ruminal pH in dairy cows follows a diurnal pattern and can remain below the critical level of 5.5 for extended periods of the day. This study aimed to evaluate the effect of low ruminal pH on blood concentrations of certain macrominerals, trace minerals, and fat-soluble vitamins and on the oxidative status of dairy cows during the first half of lactation. Fifty-three randomly selected lactating Holstein cows were used; blood and ruminal fluid samples were collected from all cows on days 30, 90 and 150 of lactation. Blood samples were obtained via coccygeal venipuncture, while the ruminal fluid was obtained by rumenocentesis and the pH was measured immediately after collection. Using a threshold pH of 5.5, samples were classified as normal (pH > 5.5) or low pH (pH ≤ 5.5). Serum concentrations of Ca, Mg, K, Cr, Mn, Zn, Se, and vitamins A, D3, E, and K were not significantly affected by ruminal pH, either by days in milk or by their interaction (p > 0.05). Plasma malondialdehyde and reduced glutathione followed the same trend (p > 0.05). Copper concentration was significantly higher (p < 0.05), and Fe concentration tended to be higher in cows with low pH compared to those with normal pH (p = 0.052). On day 150 of lactation, Cu, Fe, and Co concentrations were significantly higher in low-pH cows compared to normal-pH cows (p < 0.05). Low ruminal pH is associated with significant changes in serum concentrations of copper, iron, and cobalt but has no significant effect on the oxidative status of the animals or on the serum concentrations of the macro elements and fat-soluble vitamins studied. This study aimed to determine the factors associated with the occurrence of the first-wave dominant follicle (DF) in the ovary ipsilateral to the corpus luteum (CL) in lactating dairy cows and dairy heifers. A total of 505 estruses were investigated (lactating dairy cows, n = 361; dairy heifers, n = 144). The locations of the preovulatory follicle (PF) and regressed CL were examined at the estrus, and the locations of the first-wave dominant follicle (DF) and newly formed CL were examined seven days after estrus using transrectal ultrasonography. Then, cows were classified into two groups: the first-wave DF in the ovary ipsilateral to the CL (IG) and the first-wave DF in the ovary contralateral to the CL (CG). To evaluate the factors which affect the occurrence of IG and CG, binominal logistic regression analysis was conducted; the location of the PF and regressing CL, season (warm: June–September; cool: October–May), live weight, days in milk at estrus, daily milk production, and body condition score were used as independent variables. The occurrence rate of IG was significantly higher when the PF was located contralateral to the regressing CL (lactating dairy cows, 63.4%; dairy heifers, 58.6%) rather than ipsilateral (lactating dairy cows, 44.9%; dairy heifers, 35.1%). The IG occurrence rate was significantly higher with an increase in daily milk production (<30 kg, 47.3%; 30–40 kg, 55.2%; >40 kg, 60.5%) in lactating dairy cows. In conclusion, the occurrence of IG was associated with relative locations of the PF and regressing CL in lactating dairy cows and dairy heifers and with the level of milk production in lactating dairy cows. The somatic cell count (SCC) and differential somatic cell count (DSCC) are proxies for the udder health of dairy cattle, regarded as the criterion of mastitis identification with healthy, suspicious mastitis, mastitis, and chronic/persistent mastitis. However, SCC and DSCC are tested using flow cytometry, which is expensive and time-consuming, particularly for DSCC analysis. Mid-infrared spectroscopy (MIR) enables qualitative and quantitative analysis of milk constituents with great advantages, being cheap, non-destructive, fast, and high-throughput. The objective of this study is to develop a dairy cattle udder health status diagnostic model of MIR. Data on milk composition, SCC, DSCC, and MIR from 2288 milk samples collected in dairy farms were analyzed using the CombiFoss 7 DC instrument (FOSS, Hilleroed, Denmark). Three MIR spectral preprocessing methods, six modeling algorithms, and three different sets of MIR spectral data were employed in various combinations to develop several diagnostic models for mastitis of dairy cattle. The MIR diagnostic model of effectively identifying the healthy and mastitis cattle was developed using a spectral preprocessing method of difference (DIFF), a modeling algorithm of Random Forest (RF), and 1060 wavenumbers, abbreviated as “DIFF-RF-1060 wavenumbers”, and the AUC reached 1.00 in the training set and 0.80 in the test set. The other MIR diagnostic model of effectively distinguishing mastitis and chronic/persistent mastitis cows was “DIFF-SVM-274 wavenumbers”, with an AUC of 0.87 in the training set and 0.85 in the test set. For more effective use of the model on dairy farms, it is necessary and worthwhile to gather more representative and diverse samples to improve the diagnostic precision and versatility of these models. Dietary tannin supplementation represents a potential strategy to modulate rumen fermentation and enhance lactation performance in dairy cows, though responses remain inconsistent. A 21-day feeding trial was conducted to evaluate the effect of chestnut tannin (CNT) extract on the enteric methane emissions (EME), blood metabolites, and milk production traits in mid-lactation dairy cows. Thirty-six Holstein cows were allocated to three homogeneous treatment groups: control (CNT0, 0 g/d CNT), CNT40 (40 g/d CNT), and CNT80 (80 g/d CNT). Measurements of EME, dry matter intake (DMI), milk yield (MY), and blood and milk parameters were carried out pre- and post-21-day supplementation period. Compared with the no-additive group, the CNT extract reduced methane production, methane yield, and methane intensity in CNT40 and CNT80 (p < 0.001). CNT40 and CNT80 cows exhibited lower blood urea nitrogen (p = 0.019 and p = 0.002) and elevated serum insulin (p = 0.003 and p < 0.001) and growth hormone concentrations (p = 0.046 and p = 0.034), coinciding with reduced aspartate aminotransferase (p = 0.016 and p = 0.045), and lactate dehydrogenase (p = 0.011 and p = 0.008) activities compared to control. However, CNT80 had higher circulating NEFA and BHBA than CNT0 (p = 0.003 and p = 0.004) and CNT40 (p = 0.035 and p = 0.019). The blood glucose, albumin, and total bilirubin concentrations were not affected. MY and fat- and protein-corrected milk (FPCM), MY/DMI, and FPCM/DMI were higher in both CNT40 (p = 0.004, p = 0.003, p = 0.014, p = 0.010) and CNT80 (p = 0.002, p = 0.003, p = 0.008, p = 0.013) cows compared with controls. Feeding CNT80 resulted in higher protein content (p = 0.015) but lower fat percentage in milk (p = 0.004) compared to CNT0. Milk urea nitrogen and somatic cell counts were significantly lower in both CNT40 (p < 0.001, p = 0.009) and CNT80 (p < 0.001 for both) compared to CNT0, while milk lactose did not differ between treatments. These findings demonstrate that chestnut tannin extract effectively mitigates EME while enhancing lactation performance in mid-lactation dairy cows. Full article

This study proposed an online early lameness detection method for dairy cow health management to overcome the inability of wearable sensor-based methods for online detection and low sensitivity to early lameness. Wearable IMU sensors collected acceleration data in stationary and moving states; a threshold discrimination module using variance of motion-direction acceleration was designed to distinguish states within 2 s, enabling rapid data screening. For moving-state windowed data, the InceptionTime network was modified with YOLOConv1D and SeparableConv1D modules plus Dropout, which significantly reduced model parameters and helped mitigate overfitting risk, enhancing generalization on the test set. Typical gait features were fused with deep features automatically learned by the network, enabling accurate discrimination among healthy, mild (early) lameness, and severe lameness. Results showed that the online detection model achieved 80.6% dairy cow health status detection accuracy with 0.8 ms single-decision latency. The recall and F1 score for lameness, including early and severe cases, reached 89.11% and 88.93%, demonstrating potential for early and progressive lameness detection. This study improves lameness detection efficiency and validates the feasibility and practical value of wearable sensor-based gait analysis for dairy cow health management, providing new approaches and technical support for monitoring and early intervention on large-scale farms. The environmental impact of milk production, particularly its share of greenhouse gas (GHG) emissions, is a topic under investigation in various parts of the world. This paper presents an overview of current knowledge on the carbon footprint (CF) of milk production at the farm level, with a particular focus on technological, environmental and organisational factors affecting emission levels. The analysis is based on a review of, inter alia, 46 peer-reviewed publications and 11 environmental reports, legal acts and databases concerning the CF in different regions and under various production systems. This study identifies the main sources of emissions, including enteric fermentation, manure management, and the production and use of feed and fertiliser. It also demonstrates the significant variability of the CF values, which range, on average, from 0.78 to 3.20 kg CO2 eq kg−1 of milk, determined by the farm scale, nutritional strategies, local environmental and economic determinants, and the methodology applied. Moreover, this study stresses that higher production efficiency and integrated farm management could reduce the CF per milk unit, with further intensification having, however, diminishing effects. The application of life cycle assessment (LCA) methods is essential for a reliable assessment and comparison of the CF between systems. Ultimately, an effective CF reduction requires a comprehensive approach that combines improved nutritional practices, efficient use of resources, and implementation of technological innovations adjusted to regional and farm-specific determinants. The solutions presented in this paper may serve as guidelines for practitioners and decision-makers with regard to reducing GHG emissions. Full article

This study aimed to evaluate cow longevity and identify the main culling reasons in dairy herds in Southern Brazil. Two data sets from 26 predominantly confined Holstein herds were analyzed over a 10-year period (2007–2016). The first included 11,150 cows that were culled, died, or sold, and the second comprised 636,739 cows for demographic analysis. The average annual culling rate was 24.2%, mainly due to reproductive disorders (34.0%), mastitis/high somatic cell count (20.4%), and feet and leg problems (17.9%). Involuntary causes represented 89.5% of all culling. The death rate averaged 3.8%, with the most frequent causes being unknown (27.3%), other reasons (25.6%), tick fever (10.2%), and accidents/injuries (10.0%). Larger herds had higher culling rates than smaller ones (26.2% vs. 22.8%; p = 0.04), as did higher-producing herds compared to lower-producing ones (25.7% vs. 22.0%; p = 0.02). Cows with ≥5 calvings were culled more often (p < 0.01) than those in earlier lactations. Culling was lowest (p < 0.02) in spring and highest (p < 0.01) during early (0–60 d) and late (>420 d) lactation. Herds with a higher proportion of older cows had slightly lower milk yields (p < 0.01), indicating longevity does not always enhance productivity. Full article

Milk lactose concentration has been proposed as a noninvasive indicator of metabolic health in dairy cows, particularly during early lactation when metabolic demands are elevated. This study aimed to investigate the relationship between milk lactose levels and physiological, biochemical, and behavioral parameters in early-lactation Holstein cows. Twenty-eight clinically healthy cows were divided into two groups: Group 1 (milk lactose < 4.70%, n = 14) and Group 2 (milk lactose ≥ 4.70%, n = 14). Both groups were monitored over a 21-day period using the Brolis HerdLine in-line milk analyzer (Brolis Sensor Technology, Vilnius, Lithuania) and SmaXtec intraruminal boluses (SmaXtec Animal Care Technology®, Graz, Austria). Parameters including milk yield, milk composition (lactose, fat, protein, and fat-to-protein ratio), blood biomarkers, and behavior were recorded. Cows with higher milk lactose concentrations (≥4.70%) produced significantly more milk (+12.76%) and showed increased water intake (+15.44%), as well as elevated levels of urea (+21.63%), alanine aminotransferase (ALT) (+22.96%), glucose (+4.75%), magnesium (+8.25%), and iron (+13.41%) compared to cows with lower lactose concentrations (<4.70%). A moderate positive correlation was found between milk lactose and urea levels (r = 0.429, p < 0.01), and low but significant correlations were observed with other indicators. These findings support the use of milk lactose concentration as a practical biomarker for assessing metabolic and physiological status in dairy cows, and highlight the value of integrating real-time monitoring technologies in precision livestock management. Inflammatory processes within the body have been hypothesised to be causative agents of various health complications, including reproductive issues. This study investigates the effects of mastitis on ovarian morphometry and fertility outcomes while also comparing the effectiveness of different synchronisation protocols in affected cows. Healthy multiparous Holstein dairy cows, in their second to fifth lactations and with an average milk yield of approximately 12,000 kg in the preceding lactation, were selected for this study. The average milk yield per lactation was approximately 12,000 kg, with an average daily milk yield of 30 kg. The aim of this study was to determine the impact of mastitis on the reproductive cycle, ovarian morphometry, and function in dairy cows, as well as its effect on synchronisation efficiency. This study focused on both clinically healthy cows and those diagnosed with clinical mastitis postpartum. Three different synchronisation protocols—OvSynch, G7G, and Presynch—were evaluated at 60 days in milk (DIM). A total of 110 cows were included in this study, with 53 being clinically healthy and 57 affected by mastitis. The results indicated that inflammation affects ovarian morphometric parameters such as the area and the number of functional structures. In addition, the G7G protocol was observed to result in higher conception rates in cows suffering from mastitis following the second insemination. This study demonstrated that cows that developed mastitis within the first 30 DIM exhibited higher conception rates after the first insemination compared to those in cows that developed mastitis later in lactation.
