Aflatoxin B1 (AFB1) contamination in corn silage poses significant risks to livestock and human health. This study developed a non-destructive detection method for AFB1 using color-sensitive arrays (CSAs). Twenty self-developed CSAs were employed to react with samples, with reflectance spectra collected using a portable spectrometer. Spectral data were optimized through seven preprocessing methods, including Standard Normal Variate (SNV), Multiplicative Scatter Correction (MSC), first-order derivative (1st D), second-order derivative (2nd D), wavelet denoising, and their combinations. Key variables were selected using five feature selection algorithms: Competitive Adaptive Reweighted Sampling (CARS), Principal Component Analysis (PCA), Random Forest (RF), Uninformative Variable Elimination (UVE), and eXtreme Gradient Boosting (XGBoost). Five machine learning models were constructed: Light Gradient Boosting Machine (LightGBM), XGBoost, Support Vector Regression (SVR), RF, and K-Nearest Neighbor (KNN). The results demonstrated significant AFB1-responsive characteristics in three dyes: (2,3,7,8,12,13,17,18-octaethylporphynato)chloromanganese(III) (Mn(OEP)Cl), Bromocresol Green, and Cresol Red. The combined 1st D-PCA-KNN model showed optimal prediction performance, with determination coefficient (Rp2 = 0.87), root mean square error (RMSEP = 0.057), and relative prediction deviation (RPD = 2.773). This method provides an efficient solution for silage AFB1 monitoring. Full article

Herein, a heterogeneous Fenton-like catalyst was designed by immobilizing iron oxide (FeS) and nickel sulfide (NiS) on the surface of β-cyclodextrin (β-CD), creating a NiS/FeS@β-CD composite for degrading triphenylmethane cresol red dye. Varied instruments were used to study the physical and chemical characteristics of the NiS/FeS@β-CD catalyst. The appropriate catalytic conditions of the Fenton-like degradation of cresol red by NiS/FeS@β-CD were identified, clarifying that the higher degradation % fulfilled 99.86% with an adsorption % of 27.44% at a cresol red concentration = 50 mg/L, NiS/FeS@β-CD dose = 0.01 g, pH = 3, processing temperature = 30 °C, H2O2 concentration = 100 mg/L, and H2O2 volume = 1 mL. The kinetic assessments depicted the preference of the second order to represent the Fenton-like degradation of cresol red by NiS/FeS@β-CD. The mechanistic proposition of the adsorption/Fenton-like degradation of cresol red was understood using a quenching test and XPS analysis. Finally, to confirm the durability of NiS/FeS@β-CD, a reusability test was proceeded on the catalyst for five adsorption/Fenton-like degradation runs, with identifying the leaching concentrations of nickel and iron from the catalyst by ICP-OES after each run. (1) Background: There are many cases of human disease caused by the hepatitis A virus contamination of aquatic products, so the development of the rapid detection of hepatitis A virus in aquatic products is crucial. (2) Methods: In this study, we developed three visual loop-mediated isothermal amplification methods for the rapid and intuitive detection of hepatitis A virus in aquatic products. New specific LAMP primers were designed for the HAV-specific VP1 protein shell. (1) HNB dye was added to the LAMP reaction system. After the reaction, the color of the reaction mixture changed from violet to sky blue, showing a positive result. (2) Cresol red dye was added to the LAMP reaction system, and a positive result was indicated by orange, while a negative result was indicated by purple. (3) By labeling FIP with biotin and LF with 6-FAM, the amplified product simultaneously contained biotin and 6-FAM, which bound to the anti-biotin antibody on the gold nanoparticles on the lateral flow dipstick (LFD). Subsequently, biotin was further combined with the anti-fam antibody on the T-line of the test strip to form a positive test result. (3) Results: The three visual LAMP methods were highly specific for HAV. The sensitivity of the visual assay was 2.59 × 100 copies/μL. The positive detection ratio for 155 bivalve shellfish samples was 8.39%, which was the same as that for RT-qPCR. The three visual LAMP methods established in our work have better sensitivity than the international gold standard, and their operation is simple and requires less time. (4) Conclusions: The results can be obtained by eye color comparison and lateral flow dipsticks. Without the use of large-scale instrumentation, the sensitivity is the same as that of RT-qPCR. The test strips are lightweight, small in size, and easy to carry; they are suitable for emergency detection, on-site monitoring, field sampling, or remote farms and other non-laboratory environments for rapid identification. In this study, an innovative odor imaging system capable of detecting adulteration in quince seed edible oils mixed with sunflower oil and sesame oil based on their volatile organic compound (VOC) profiles was developed. The system comprises a colorimetric sensor array (CSA), a data acquisition unit, and a machine learning algorithm for identifying adulterants. The CSA was created using a method that involves applying a mixture of six different pH indicators (methyl violet, chlorophenol red, Nile blue, methyl orange, alizarin, cresol red) onto a Thin Layer Chromatography (TLC) silica gel plate. Subsequently, difference maps were generated by subtracting the “initial” image from the “final” image, with the resulting color changes being converted into digital data, which were then further analyzed using Principal Component Analysis (PCA). Following this, a Support Vector Machine was employed to scrutinize quince seed oil that had been adulterated with varying proportions of sunflower oil and sesame oil. The classifier was progressively supplied with an increasing number of principal components (PCs), starting from one and incrementally increasing up to five. Each time, the classifier was optimized to determine the hyperparameters utilizing a random search algorithm. With one to five PCs, the classification error accounted for a range of 37.18% to 1.29%. According to the results, this novel system is simple, cost-effective, and has potential applications in food quality control and consumer protection. Full article

The present work focuses on the investigation of protective coatings produced on zinc from chitosan (Chit) and an anionic dye, namely cresol red. Cresol red (CR) fulfills the basic requirements to be used as a corrosion inhibitor because it possesses a relatively high molecular weight and includes in its structure oxygen and sulfur atoms as well as aromatic rings. Moreover, it is an anionic compound that can interact with positively charged chitosan to produce reinforced coatings for zinc anti-corrosion protection. The influence of cresol red as a possible corrosion inhibitor for zinc substrates was investigated either in solution or incorporated in Chit coatings. Two preparation methods for the coatings were used: (i) Chit coating impregnation by immersion in the CR solution after Chit deposition on Zn, and (ii) chitosan mixing with the CR solution before applying the dip-coating technique. Potentiodynamic polarization curves were used to determine the kinetic parameters of the corrosion process. Long-term measurements were carried out in wet/dry cyclic conditions by using electrochemical impedance spectroscopy. EIS measurements recorded in 0.2 g/L Na2SO4 at pH = 7 show an important increase in the impedance of the coatings occurring from the first until the fifty-fifth day in a row, in dry–wet cycles. This increase is due to the beneficial effect of CR incorporated in Chitosan and could be, at least partially, related to a consolidation of the Chit coating structure in the presence of CR by crosslinking between Chit and CR molecules. The structure of the coatings was studied, and the interactions between chitosan and cresol red were put into evidence by using FT-IR spectroscopy. Adhesion and wettability measurements were also carried out. The adhesion of Chit incorporating CR on Zn was better than that on glass substrates and reached ~99.99%, suggesting a better affinity of the chitosan coating towards the Zn substrate due to the existence of ZnO on the substrate surface. All the results show that CR could be used on zinc as a corrosion inhibitor incorporated in chitosan at basic pHs, but without taking advantage of its pH-indicating properties, which are lost due to the interactions occurring between the positively charged biopolymer and the negatively charged dye molecule. The preparation method of Chit coating impregnation with CR by immersion in the solution after deposition on Zn led to poorer results than the method in which chitosan was previously mixed with CR before applying the dip-coating technique. Since SARS-CoV-2 is a highly transmissible virus, a rapid and accurate diagnostic method is necessary to prevent virus spread. We aimed to develop and evaluate a new rapid colorimetric reverse transcription loop--mediated isothermal amplification (RT-LAMP) assay for SARS-CoV-2 detection in a single closed tube. Nasopharyngeal and throat swabs collected from at-risk individuals testing for SARS-CoV-2 were used to assess the sensitivity and specificity of a new RT-LAMP assay against a commercial qRT-PCR assay. Total RNA extracts were submitted to the RT-LAMP reaction under optimal conditions and amplified at 65 °C for 30 min using three sets of specific primers targeting the nucleocapsid gene. The reaction was detected using two different indicator dyes, hydroxynaphthol blue (HNB) and cresol red. A total of 82 samples were used for detection with HNB and 94 samples with cresol red, and results were compared with the qRT-PCR assay. The sensitivity of the RT-LAMP-based HNB assay was 92.1% and the specificity was 93.2%. The sensitivity of the RT-LAMP-based cresol red assay was 80.3%, and the specificity was 97%. This colorimetric feature makes this assay highly accessible, low-cost, and user-friendly, which can be deployed for massive scale-up and rapid diagnosis of SARS-CoV-2 infection, particularly in low-resource settings. The red flour beetle, Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae), is a major storage pest that could lead to a wide range of damage. Its secretions have a significant impact on the quality of stored grain and food, leading to serious food safety problems such as grain spoilage and food carcinogenesis. This study investigates new detection techniques for grain storage pests to improve grain insect detection in China. The primary volatile organic chemicals (VOCs) in these secretions are identified using headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography-mass spectrometry (GC-MS). The specific VOCs that are unique to T. castaneum are selected as criteria for determining the presence of T. castaneum in the granary. To obtain more specific VOCs, experiments were designed for the analysis of T. castaneum samples under different extraction times, two types of SPME fibers and two GC-MS devices of different manufacturers. The experimental results showed that 12 VOCs were detected at relatively high levels, seven of which were common and which were not detected in other grains and grain insects. The seven compounds are 1-pentadecene, 2-methyl-p-benzoquinone, 2-ethyl-p-benzoquinone, 1-hexadecene, cis-9-tetradecen-1-ol, m-cresol and paeonol. These seven compounds can be used as volatile markers to identify the presence of T. castaneum, which could serve as a research foundation for the creation of new techniques for T. castaneum monitoring. This study aims to develop a freshness indicator for fish products that changes color to indicate ammonia among volatile base compounds (TVB-N) generated during storage. Through an optimization experiment, we observed the indicator’s color change relative to the ammonia concentration standard, finally selecting cresol red and bromocresol purple for the indicator mixture. In addition, eco-DEHCH and Breathron film were applied to the freshness indicator, considering environmental and economic values. For the storage experiment, Chub mackerel (Scomber japonicus), Spanish mackerel (Scomberomorus niphonius), and Largehead hairtail (Trichiurus lepturus) samples were stored at three different temperatures (4, 10, and 20 °C) for seven days, and we measured pH, TVB-N, total bacterial count, and ammonia content every 24 h. The pH-sensitive sensors’ color changes monitor amine release, especially ammonia, from decomposing fish. The chromatic parameter ∆E value increased significantly with fish product storage periods. We confirmed that when the freshness limit and bacterial spoilage level were reached, the color of the indicator changed from yellow to black and sequentially changed to purple as the storage period increased. Therefore, a developed freshness indicator can be used for determining the quality of fish products quickly and non-destructively by reflecting the freshness and spoilage degree of fish products during storage. Electrochemical removal of organic pollutants represents an attractive methodology in water depollution. The key challenges for researchers comprise finding simple, affordable electrode materials with satisfactory efficiency in all ranges of pollutant concentration. Electrochemical oxidation of a mixture of phenol-based dyes: bromocresol green (BCG), cresol red (CR), and thymol blue (TB), in sulphate medium, at total concentration not exceeding 15 ppm, has been performed using simply prepared, low-cost composite electrodes, based on graphene nanoplatelets (GNP) and metallic oxides (TiO2 and SnO2) loaded on stainless steel substrate: GNP@SS, SnO2/GNP@SS, and TiO2/GNP@SS. Electrodes were characterised by XRD, FTIR, and electrochemical techniques. The degradation kinetics of initial dyes was tracked with UPLC and GC-MS chromatography for 6 h, at a current density of 10 mA/cm2. GC-MS analysis of the degradation products revealed oxidised aromatic compounds as the main products, while TOC analysis confirmed a total mineralisation extent in the range of 30–35%. The proposed degradation mechanism involves the attack of OH-radical, as the main oxidising agent, to the hydroxyl oxygens of dye phenolic rings. Obtained results provide useful information for the further development of affordable laboratory-scale and industrial systems for the complete removal of phenol-based compounds. Full article

Eryptosis is the stress-induced RBC (red blood cell) death mechanism. It is known that eryptosis is largely influenced by plasma and blood composition, and that it is accelerated in patients affected by chronic kidney disease (CKD). The aim of this study is to evaluate the eryptosis rate in healthy RBCs treated with different concentration of IL-6, IL-1β, urea and p-cresol, comparable to plasmatic level of CKD patients, at different time points. We exposed healthy RBCs to increasing concentrations of IL-6, IL-1β, urea and p-cresol. Morphological markers of eryptosis (cell membrane scrambling, cell shrinkage and PS exposure at RBC surface) were evaluated by flow cytometric analyses. The cytotoxic effect of cytokines and uremic toxins were analyzed in vitro on healthy RBCs at 4, 8 and 24 h. Morphology of treated RBCs was dramatically deranged, and the average cell volume was significantly higher in RBCs exposed to higher concentration of all molecules (all, p < 0.001). Furthermore, healthy RBCs incubated with each molecules demonstrated a significant increase in eryptosis. Cytofluorimetric analysis of eryptosis highlighted significantly higher cell death rate in RBCs incubated with a higher concentration of both cytokines compared with RBCs incubated with a lower concentration (all, p < 0.05). In conclusion, our data show that cytokines and uremic toxins have a harmful effect on RBCs viability and trigger eryptosis. Further studies are necessary to validate these results in vivo and to associate abnormal eryptosis with cytokine levels in CKD patients. The eryptosis pathway could, moreover, become a new promising target for anemia management in CKD patients. Full article

Over the years, many materials have been used to restore buildings, paintings, ceramics, and mosaic pieces exhibiting different types of dyes and colour hues. Recently, geopolymers have been used for restoration purposes owing to their high chemical and mechanical resistance. In this work, white metakaolin was used to obtain white geopolymers, cured at 25 and 40 °C, as bulk materials to be coloured with synthetic organic dyes, i.e., bromothymol blue, cresol red, phenolphthalein, and methyl orange. These dyes were added during the fresh paste preparation to obtain dyed geopolymeric solids. Ionic conductivity and pH measurement confirmed the chemical stability of the consolidated materials, while FT-IR analyses were used to follow the geopolymerisation occurrences at different ageing times (from 7 to 56 days). Finally, the colour hues and properties were assessed in the CIELAB colour space before and after immersion in water. Full article

Smoke taint in wine is thought to be caused by smoke-derived volatile phenols (VPs) that are absorbed into grape tissues, trapped as conjugates that are imperceptible by smell, and subsequently released into wines as their free odor-active forms via metabolism by yeasts during fermentation. Blocking VP uptake into grapes would, therefore, be an effective way for vineyards to protect ripening grape crops exposed to smoke. Here, we re-evaluated a biofilm that had previously shown promise in pilot studies in reducing levels of smoke-derived VPs. A suite of nine free and acid-labile VPs were quantitated in Pinot Noir grapes that had been exposed to smoke after being coated with the biofilm one, seven or 14 days earlier. In contrast with earlier studies, our results demonstrated that in all cases, the biofilm treatments led to increased concentrations of both free and total VPs in smoke-exposed grapes, with earlier applications elevating concentrations of some VPs more than the later time points. Tracking VP concentrations through the grape ripening process demonstrated that some (phenol, p/m-cresol, and guaiacol) were not entirely sequestered in grapes as acid-labile conjugates, suggesting the presence of VP storage forms beyond simple glycosides. Free VPs in grapes, though a minor portion of the total, most clearly correlated with concentrations present in the resulting wines. Finally, red table grapes, available year round, were observed to replicate the effects of the biofilm treatments and were capable of transforming most VPs into acid-labile conjugates in under 24 h, indicating that they might be an effective model for rapidly assessing smoke-taint prophylactic products in the laboratory. This study investigated the polymer film composite of polyvinyl alcohol (PVA), trichlorethylene (TCE) and cresol red (CR) dye irradiated with gamma (γ) rays for potential application as radiation dosimetry. The film was prepared via the solvent-casting method with varying concentrations of TCE. Film samples were exposed to radiation from a γ-rays radiation source of 60Cobalt isotope. Color changes before and after γ-rays irradiation were observed, and the optical properties of the polymer films were investigated by spectrophotometry. Results show that increasing the radiation dose physically changed the color of the polymer film, from purple (pH > 8.8) without radiation (0 kGy) to yellow (almost transparent) (2.8 < pH < 7.2) at the highest dose (12 kGy). The concentration of acid formed due to irradiation increased with the increase in irradiation doses and at higher TCE content. The critical doses of PVA-TCE composites decreased linearly with the increase of TCE composition, facilitating an easy calibration process. The dose response at 438 nm increased exponentially with increasing radiation dose, but showed an opposite trend at the 575 nm band. An increase in the TCA concentration indicated a decrease in the absorption edge and an increase in activation energy, but both decreased for all TCE concentrations at higher doses. The energy gap for the direct and the indirect transitions decreased with increasing TCE concentration and γ-rays radiation dose. The results of this study demonstrated the potential application of PVA-TCE-CR polymer film as γ-rays irradiation dosimetry in a useful dose range of 0–12 kGy. Full article

LC-MS-assisted metabolomic profiling of the Red Sea-derived brown algae Sargassum cinereum “Sargassaceae” dereplicated eleven compounds 111. Further phytochemical investigation afforded two new aryl cresol 1213, along with eight known compounds 1421. Both new metabolites, along with 19, showed moderate in vitro antiproliferative activity against HepG2, MCF-7, and Caco-2. Pharmacophore-based virtual screening suggested both 5-LOX and 15-LOX as the most probable target linked to their observed antiproliferative activity. The in vitro enzyme assays revealed 12 and 13 were able to inhibit 5-LOX more preferentially than 15-LOX, while 19 showed a convergent inhibitory activity toward both enzymes. Further in-depth in silico investigation revealed the molecular interactions inside both enzymes’ active sites and explained the varying inhibitory activity for 12 and 13 toward 5-LOX and 15-LOX. Environmental impact associated with odor and gaseous emissions from animal manure is one of the challenges for communities, farmers, and regulatory agencies. Microbe-based manure additives treatments are marketed and used by farmers for mitigation of emissions. However, their performance is difficult to assess objectively. Thus, comprehensive, practical, and low-cost treatments are still in demand. We have been advancing such treatments based on physicochemical principles. The objective of this research was to test the effect of the surficial application of a thin layer (¼ inches; 6.3 mm) of biochar on the mitigation of gaseous emissions (as the percent reduction, % R) from swine manure. Two types of biochar were tested: highly alkaline and porous (HAP) biochar made from corn stover and red oak (RO), both with different pH and morphology. Three 30-day trials were conducted with a layer of HAP and RO (2.0 & 1.65 kg∙m−2, respectively) applied on manure surface, and emissions of ammonia (NH3), hydrogen sulfide (H2S), greenhouse gases (GHG), and odorous volatile organic compounds (VOCs) were measured. The manure and biochar type and properties had an impact on the mitigation effect and its duration. RO significantly reduced NH3 (19–39%) and p-cresol (66–78%). H2S was mitigated (16~23%), but not significantly for all trials. The phenolic VOCs had relatively high % R in most trials but not significantly for all trials. HAP reduced NH3 (4~21%) and H2S (2~22%), but not significantly for all trials. Significant % R for p-cresol (91~97%) and skatole (74~95%) were observed for all trials. The % R for phenol and indole ranged from (60~99%) and (29~94%) but was not significant for all trials. The impact on GHGs, isobutyric acid, and the odor was mixed with some mitigation and generation effects. However, larger-scale experiments are needed to understand how biochar properties and the dose and frequency of application can be optimized to mitigate odor and gaseous emissions from swine manure. The lessons learned can also be applicable to surficial biochar treatment of gaseous emissions from other waste and area sources. Full article
