Ru(II) Complexes with 3,4-Dimethylphenylhydrazine: Exploring In Vitro Anticancer Activity and Protein Affinities

Two new Ru(II) complexes, mononuclear [RuCl26-p-cymene)(3,4-dmph-κN)] (1) and the binuclear complex [{RuCl(η6-p-cymene)}2(μ-Cl)(μ-3,4-dmph-κ2N,N′)]Cl (2; 3,4-dmph = 3,4-dimethylphenylhydrazine), are synthesized and experimentally and theoretically structurally characterized utilizing 1H and 13C NMR and FTIR spectroscopy, as well as DFT calculations. Degradation product of 2, thus ([{RuCl(η6-p-cymene)}2(μ-Cl)(μ-3,4-dmph-κ2N,N′)][RuCl36-p-cymene)] (2b) was characterized with SC-XRD. In the crystals of 2b, the cationic and anionic parts interact through N-H...Cl hydrogen bridges. The spectrofluorimetric measurements proved the spontaneity of the binding processes of both complexes and HSA. Spin probing EPR measurements implied that 1 and 2 decreased the amount of bound 16-doxylstearate and implicated their potential to bind to HSA more strongly than the spin probe. The cytotoxicity assessment of both complexes against the MDA-MB-231 and MIA PaCa-2 cancer cell lines demonstrated a clear dose-dependent decrease in cell viability and no effect on healthy HS-5 cells. Determination of the malondialdehyde and protein carbonyl concentrations indicated that new complexes could offer protective antioxidant benefits in specific cancer contexts. Gel electrophoresis measurements showed the reduction in MMP9 activity and indicated the potential of 1 in limiting the cancer cells’ invasion. The annexin V/PI apoptotic assay results showed that 1 and 2 exhibit different selectivity towards MIA PaCa-2 and MDA-MB-231 cancer cells. A comparative molecular docking analysis of protein binding, specifically targeting acetylcholinesterase (ACHE), matrix metalloproteinase-9 (MMP-9), and human serum albumin (HSA), demonstrated distinct binding interactions for each complex. Heterometallic phosphinidene complexes are appealing species for the construction of novel organophosphorus ligands thanks to the high reactivity expected from the combination of M-P multiple bonding and the intrinsically different electronic and coordination preferences of the distinct metals. In a preliminary study, we found that the heterobimetallic complex [MoReCp(μ-PMes*)(CO)6] (Mes* = 2,4,6-C6H2tBu3) reacted with CN(p-C6H4OMe) via [2+1]-cycloaddition to form a novel azaphosphallene complex. We have now examined in detail the reactions of the above complex and those of its MoMn analogue with different isocyanides, which turned out to be strongly dependent on experimental conditions and on the size of the substituent at the isocyanide. All the products formed follow from one or several of the following reaction pathways: (i) CO substitution by CNR; (ii) addition of CNR at the group 7 metal centre; and (iii) [2+1] cycloaddition of isocyanide at a Mo=P bond to form azaphosphallene groups, with the former process being dominant in reactions at room temperature and for the Mn system. In contrast, low-temperature reactions of the Re system favoured the addition processes, with the [2+1] cycloaddition at Mo=P bonds only taking place at substrates without metal-metal bonds and when the size of the CNR group does not cause unbearable steric clashes when placed in between the Cp and Mes* groups. Complexes displaying multiple bonds between different metal atoms have considerable synthetic potential because of the combination of the high electronic and coordinative unsaturation associated to multiple bonds with the intrinsic polarity of heterometallic bonds but their number is scarce and its chemistry has been relatively little explored. In a preliminary study, our attempted synthesis of the unsaturated hydrides [MoMCp(μ-H)(μ-PR2)(CO)5] from anions [MoMCp(μ-PR2)(CO)5] and (NH4)PF6 yielded instead the ammonia complexes [MoMCp(μ-H)(μ-PR2)(CO)5(NH3)] (M = Mn, R = Ph; M = Re, R = Cy). We have now examined the structure and behaviour of the MoMn complex (Mo–Mn = 3.087(3) Å) and found that it easily dissociates NH3 (this requiring some 40 kJ/mol, according to DFT calculations), to yield the undetectable unsaturated hydride [MoMnCp(μ-H)(μ-PPh2)(CO)5] (computed Mo–Mn = 2.796 Å), the latter readily adding simple donors L such as CNR (R = Xyl, p-C6H4OMe) and P(OMe)3, to give the corresponding electron-precise derivatives [MoMnCp(μ-H)(μ-PPh2)(CO)5(L)]. Thus the ammonia complex eventually behaves as a synthetic equivalent of the unsaturated hydride [MoMnCp(μ-H)(μ-PPh2)(CO)5]. The isocyanide derivatives retained the stereochemistry of the parent complex (Mo–Mn = 3.0770(4) Å when R = Xyl) but a carbonyl rearrangement takes place in the reaction with phosphite to leave the entering ligand trans to the PPh2 group, a position more favoured on steric grounds.
