Detection of Bartonella spp. in a Pipistrellus Pipistrellus Bat from Portugal

In recent years, zoonotic pathogens have become increasingly more relevant in scientific research due to their implications on public health. Understanding their pathogenic potential, the pathways they use to infect and their reservoirs enables better care for both human and animal patients, and possible infection outbreaks can be more easily contained. Bartonella belongs to a vast list of zoonotic pathogens that can infect mammals, including humans, but also companion animals and wildlife, and is capable of causing disease. Bats are a possible source and reservoir of this bacterial genus, and Bartonella spp. has already been identified in these animals in several countries. Using these premises, skin samples of the pinna and wing of 71 bats (Pipistrellus pipistrellus) collected from southern Portugal were tested through PCR for the presence of Bartonella spp., and positive results were found in 1.41% (1/71). The sequence obtained shared genetic proximity with an already known pathogenic Bartonella strain that affects both humans and animals. From the public health perspective, these findings suggest that bats may play a role in the transmission of this pathogen and provides new insights into the presence of this agent in Portugal. The Soprano pipistrelle bat, Pipistrellus pygmaeus, is a common species in large parts of Sweden. Many of its natural habitats are near human habitations. This creates opportunities for ticks infesting these bats to encounter humans and possibly transmit zoonotic pathogens by tick bites. The bats are often infested with Carios vespertilionis, a tick species that, in addition to bats, has been recorded to bite humans on occasion. This study aimed to investigate if C. vespertilionis acts as a reservoir for Anaplasma phagocytophilum, Neoehrlichia mikurensis, Tick-borne encephalitis virus, and species of Babesia and Rickettsia and to improve currently used conventional PCR protocols for molecular species determination of Rickettsia spp. Ninety-two C. vespertilionis ticks were collected from underneath a bat-box harbouring P. pygmaeus. Pathogen-specific PCR assays showed that 58.4% were positive for Rickettsia spp. and negative for the other pathogens analysed. Phylogenetic analyses indicate that the species belong to R. parkeri, R. conorii, R. slovaca, R. sibirica subsp. mongolotimonae, R. rickettsii, and a hitherto uncultured Rickettsia sp. Several of these species are considered pathogenic to humans. Given the ecology and behaviour of C. vespertilionis, it may be a vector of these rickettsiae among bats and occasionally humans. To determine the Rickettsia species with certainty, and to determine if C. vespertilionis may be a reservoir and vector of the Rickettsia spp., further studies are needed. Full article

Toxoplasma gondii infection in healthy animals is often asymptomatic. However, some species with little history of contact with the parasite, such as marsupials and New World primates, present high mortality rates after infection. Despite its potential conservation concern, T. gondii infection in insectivorous bats has received little attention, and its impact on bat populations’ health is unknown. To assess the putative role of insectivorous bats in the cycle of T. gondii, samples of three species of bats (Pipistrellus pipistrellus, P. pygmaeus and P. kuhlii) collected between 2019 and 2021 in NE Spain were tested for the presence of the parasite using a qPCR. All tissues resulted negative (0.0% prevalence with 95% CI: [0.0–2.6]) for the presence of T. gondii. Unlike previous studies on insectivorous bats from Europe, Asia and America, the present study suggests that Pipistrellus spp. bats do not play a significant role in the epidemiology of T. gondii in NE Spain. Further studies are encouraged to elucidate both the epidemiology of T. gondii and its potential impact on the health of microchiropteran species in Europe. Full article

The bat tick Cariosvespertilionis has been reported from Sweden to occasionally feed on humans resulting in disease symptoms. The aim of this study was to investigate C. vespertilionis as a potential vector and reservoir of Borrelia species. In 2015 and 2018 in south-central Sweden, C. vespertilionis ticks were collected from a wooden bat box harboring Soprano pipistrelle bats, Pipistrellus pygmaeus. In addition, one C. vespertilionis tick found inside a house in southern Sweden in 2019 was collected. Ticks were screened for Borrelia spp. using a genus-specific quantitative PCR assay. The Borrelia species of the positive specimens were determined by conventional PCR followed by DNA sequencing and phylogenetic analyses. A total of 24% (22 of 92) of the analyzed C. vespertilionis ticks were Borrelia-positive. Phylogenetic analyses indicate that the bacteria belong to the relapsing fever group of borreliae; some of them appear to be identical with Borrelia sp. CPB1, a spirochete only found twice before—in the United Kingdom and in France. Our results also indicate a temporal and spatial distribution of this Borrelia species. Since C. vespertilionis occasionally bites humans, and since it exhibits a high prevalence of Borrelia bacteria, it is possible that it presents a risk of human disease. Further studies are needed to characterize Borrelia sp. CPB1 to determine if it is human-pathogenic and to determine if C. vespertilionis is a vector and/or reservoir of this agent. Full article

In times of land use changes towards more close-to-nature forestry, the application of bioindicators becomes an interesting tool for effective land-use management schemes. Forest managers are increasingly confronted by alien tree species. Therefore, this case study aimed to investigate the influence of the invasive black cherry (Prunus serotina) on bats (Chiroptera: Verpertilionidae) in pine (Pinus sylvestris) forest ecosystems, in order to identify the potential of bats as bioindicators for a black cherry invasion. In three pre-classified succession stages of the black cherry, the diversity and relative abundance of bats were bioacoustically monitored for a period of 60 nights. From the bat call recordings made during the study period, eight bat species could be identified to species level. Within the succession stages of pine monoculture and light black cherry forest, a comparable bat diversity of eight bat species and three sonotypes with a similar relative abundance were observed. In dense black cherry forest, only four species and one sonotype were detected. Compared to the pine monoculture and light black cherry forest, the overall abundance of the bat community was significantly lower in the dense black cherry forest. Upon evaluation, those bat species associated with the edge and narrow space forager guilds were found to have a high sensitivity to a dense black cherry understory within naturally monocultural pine stands. Their activity patterns indicate that the transition from light to dense black cherry understory can be considered as a potential threshold value for a close-to-nature black cherry understory density in high canopy pine forest stands.
