Background/Objectives: There is evidence concerning herbal medicines and plant-based compounds, including Lamiaceae species, as putative senolytic agents; however, there are only a few reports on Ocimum americanum properties using rat models. The aim of this study was to investigate the neuroprotective effects and potential modes of action of Ocimum americanum L. using ex vivo and in vivo assays to assess the effects of OAEE on hippocampal tissue from young adult and late middle-aged Wistar rats, with a focus on oxidative stress, cholinesterase activity, and neuroinflammatory markers. Methods: Ocimum americanum ethanol extract (OAEE) was incubated with hippocampal slices of young adult and late middle-aged male Wistar rats exposed to H2O2; an acute treatment with OAEE was evaluated in aversive memory performance and neurochemical parameters, such as hippocampal cellular oxidative state, and anticholinesterase activity, and a diet supplementation of OAEE were evaluated on several hippocampal biochemical parameters, such as oxidative state, anticholinesterase activity, and neuroinflammatory parameters in young adult and late middle-aged male rats. Results: OAEE reversed the H2O2-induced impaired cellular viability in hippocampal slices from young adult rats, as well as protected hippocampal slices against H2O2-induced damage in both young adult and late middle-aged Wistar rats, indicating its neuroprotective action. Chronic dietary OAEE supplementation reduced aging-induced increases in reactive species and lipid peroxidation levels in the hippocampus. Indeed, this supplementation reduced the TNF-α content in hippocampus from both ages, and IL-1β levels in young adult rats. Conclusions: The antioxidant actions of OAEE here observed, preventing the lipoperoxidation, as well as its anti-neuroinflammatory effect, might be related to neuroprotective effect. Our findings add evidence to support the idea of the potential use of Ocimum americanum as a nutraceutical or functional food in the aging process. Full article

Despite ongoing control efforts, the mosquito population and diseases vectored by them continue to thrive worldwide, causing major health concerns. There has been growing interest in the use of botanicals as alternatives to insecticides due to their widespread insecticidal properties, biodegradability, and adaptability to ecological conditions. In this study, we investigated the larvicidal activity and cytotoxicity effects of solvent extracts from three aromatic plants—Curcuma longa (turmeric), Ocimum americanum (hoary basil), and Petroselinum crispum (parsley)—against Aedes albopictus. Subsequently, we examined the phytochemical composition of the extracts through gas chromatography–mass spectrometry (GC–MS) analysis. Results revealed that the hexane extracts of O. americanum and P. crispum exhibited the greatest larvicidal activity with the lowest median lethal concentration (LC50) values (<30 µg/mL) at 24 h post-treatment, with the former found to be significantly less toxic towards African monkey kidney (Vero) cells. The GC–MS analysis of the said extract indicated the presence of different classes of metabolites, including phenylpropanoids, very long-chain alkanes, fatty acids and their derivatives, and terpenes, with the most abundant component being methyl eugenol (55.28%), most of which, have been documented for their larvicidal activities. These findings provide valuable insights into the potential use and development of bioinsecticides, particularly from O. americanum. Full article

Basil (Ocimum L.) is widely used as a flavor ingredient, however research on basil flavor is limited. In the current study, nine basil species were selected, including Ocimum basilicum L.var. pilosum (Willd.) Benth., Ocimum sanctum, Ocimum basilicum cinnamon, Ocimum gratissimum var. suave, Ocimum tashiroi, Ocimum basilicum, Ocimum americanum, Ocimum basilicum ct linalool, and Ocimum basilicum var. basilicum, and their fragrance and flavor characteristics were assessed by sensory evaluation. The results indicated that Ocimum basilicum var. basilicum and Ocimum gratissimum var. suave have a strong clove smell and exhibited a piquant taste. Metabolomics and volatilomics analyses measured 100 nonvolatile metabolites and 134 volatiles. Differential analysis showed that eugenol, γ-terpinene, germacrene D and malic acid were among the most varied metabolites in basil species. Combined with sensory evaluation results, correlation analysis revealed that β-pinene and γ-cadinene contributed to the piquant smell, while eugenol and germacrene D contributed to the clove smell, and malic acid and L-(−)-arabitol contributed to the sweet flavor in basil. This study provided comprehensive flavor chemistry profiles of basil species and could be used as a guide for basil flavor improvement. The better understanding of objective sensory attributes and chemical composition of fresh basil could introduce the improved cultivars with preponderant traits, which is also in accordance with the various demands of breeders and growers, food producers, and consumers. Ocimum plants are of great interest to traditional medicine in the history of several civilizations, particularly in terms of chronic human diseases. Essential oils obtained from this genus have also been used as therapeutic agents. In this present work, two plants of the Ocimum species from Djibouti, Ocimum basilicum L. and Ocimum americanum L., were subjected to hydrodistillation to obtain their essential oils. Gas chromatography-mass spectrometry was performed to determine the chemical composition of both essential oils. Linalool (41.2%) and estragole (30.1%) are the major compounds among the 37 compounds that have been identified in the essential oil of Ocimum basilicum L. (EOOB), and carvotanacetol (38.4%) and estragole (27.5%) are the main compounds among the 42 compounds that have been identified in the essential oil of Ocimum americanum L. (EOOA). Morever, the cytotoxic activity of EOs was evaluated against 13 human cancer cell lines (K562, A549, HCT116, PC3, U87-MG, MIA-Paca2, HEK293, NCI-N87, RT4, U2OS, A2780, MRC -5 and JIMT-T1) using a luminescence spectrophotometric method; hence, the oils showed significant cytotoxic activities. The antibacterial activities of the oils were assayed on five Gram-positive bacteria (Staphylococcus aureus, Enterococcus faecalis, Streptococcus agalactiae, Staphylococcus epidermidis and Corynebacterium sp.) and seven Gram-negative bacteria (Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae, Acinetobacter baumannii, Shigella sonnei, Salmonella enterica sv. Typhimurium and Enterobacter cloacae) by agar disc diffusion methods. Both essential oils exhibited moderate activities against Streptococcus agalactiae, and thus each has an activity against Pseudomonas aeruginosa for EOOB and against Enterobacter cloacae for EOOA, while the others are negative. The findings of this work showed the promising anticancer effects of both oils against total cell lines with a potential for use as natural alternatives to synthetic drugs; however, there was only an antibacterial effect against Streptococcus agalactiae. The yield of essential oils in plants is not high and postdistillation wastes rich in phenolic compounds could be used to enhance the profitability of essential oil-bearing plants. The aim of the study was to evaluate polyphenols in a dry extract obtained from the postdistillation waste of the American basil (Ocimum americanum L.) herb, and to conduct the screening of its pharmacological activities. Rosmarinic acid, caffeic acid and rutin were identified in the extract using high-performance thin-layer chromatography. The high-performance liquid chromatography analysis found the presence of a plethora of polyphenols in the extract. Rosmarinic acid, luteolin-7-O-glucoside and rutin were as the main compounds. The total phenolic content in the extract was 106.31 mg GAE/g and free radical scavenging activity against 2,2-diphenyl-1-picrylhydrazyl evaluated as IC50 was 0.298 mg/mL. The tested extract dose-dependently decreased the paw edema in rats, suggesting its potent anti-inflammatory property. The acute toxicity study indicates its safety. Thus, the O. americanum hydrodistilled residue by-product is the promising source of biologically active compounds with significant antioxidant and anti-inflammatory effects. Full article

Although modern medicine is available in many developing countries, such as the Comoros Islands, the primary health-care needs of the local population are based on traditional foods and beverages derived from natural resources and medicinal plants for cultural and historical reasons. Aphloia theiformis (Vahl) Benn. (‘Mfandrabo’), Cinnamomum verum J.Presl (‘Mani yamdrara’), Ocimum gratissimum L. (‘Roulé’), Plectranthus amboinicus (Lour.) Spreng. (‘Ynadombwe’), Cymbopogon nardus (L.) Rendle (‘Sandze monach’) and Ocimum americanum L. (‘Kandza’) are six wild plants that are largely utilised to treat many diseases. The leaves of these plants are used in the traditional Comorian tea (aqueous infusion). This study aimed to identify and quantify the main health-promoting compounds in the traditional formulation of Comorian tea by HPLC profiling together with a preliminary assessment of antioxidant capacity to confirm the traditional use of these plants by the local population. The single plants were also studied. The Comoros tea presented a total polyphenolic content (TPC) of 4511.50 ± 74.41 mgGAE/100 g DW, a value higher than the TPCs of the different plants included in the Comorian tea. Moreover, the Comorian tea showed an antioxidant capacity (AOC) of 578.65 ± 6.48 mmol Fe2+/Kg DW, a value higher if compared to all the AOC values obtained in the single plants. The polyphenolic fraction (771.37 ± 35.76 mg/100 g DW) and organic acids (981.40 ± 38.38 mg/100 g DW) were the most important phytochemical classes in the Comorian tea (40.68% and 51.75% of the total phytocomplex, respectively), followed by the monoterpenes (5.88%) and vitamin C (1.67%), while carotenoids were detected in trace (0.02%). The Comorian tea could be important in meeting the high demand in the Comoros Islands and other developing countries for cost-effective and natural health-promoting foods and/or beverages to be produced by agri-food industries and used by the local population. This study may promote traditional foods in rural communities in the Comoros Islands and contribute to sustainable rural development and a commercial valorisation of these plants for health-promoting and food applications. Dental caries remains the most prevalent and costly oral infectious disease worldwide. Several methods have been employed to prevent this biofilm-dependent disease, including the use of essential oils (EOs). In this systematic review, we discuss the antibacterial activity of EOs and their isolated constituents in view of a potential applicability in novel dental formulations. Seven databases were systematically searched for clinical trials, in situ, in vivo and in vitro studies addressing the topic published up to date. Most of the knowledge in the literature is based on in vitro studies assessing the effects of EOs on caries-related streptococci (mainly Streptococcus mutans) and lactobacilli, and on a limited number of clinical trials. The most promising species with antibacterial potential against cariogenic bacteria are: Achillea ligustica, Baccharis dracunculifolia, Croton cajucara, Cryptomeria japonica, Coriandrum sativum, Eugenia caryophyllata, Lippia sidoides, Ocimum americanum, and Rosmarinus officinalis. In some cases, the major phytochemical compounds determine the biological properties of EOs. Menthol and eugenol were considered outstanding compounds demonstrating an antibacterial potential. Only L. sidoides mouthwash (1%) has shown clinical antimicrobial effects against oral pathogens thus far. This review suggests avenues for further non-clinical and clinical studies with the most promising EOs and their isolated constituents bioprospected worldwide. Full article

Ursolic acid is a very important compound due to its biological potential as an anti-inflammatory, trypanocidal, antirheumatic, antiviral, antioxidant and antitumoral agent. This study presents the HPLC analysis of ursolic acid (UA) content in eight different Ocimum species: O. americanum L., O. basilicum L, O. basilicum var purpurascens Benth, O. basilicum var. minimum L, O. gratissimum L, O. micranthum Willd, O. selloi Benth. and O. tenuiflorum L. grown in Northeastern Brazil. In these Ocimum species, UA was detected in different yields, with O. tenuiflorum showing the highest content (2.02%). This yield is very significant when compared with other sources of UA.
