Assessment of Anticancer Properties of Argemone mexicana L. and Berberine: A Comparative Study

Argemone mexicana L. has been used in traditional Mexican medicine. Among its bioactive constituents, berberine (BER) has garnered attention for its cytotoxic properties against different tumor cell lines. This study investigates the in vitro toxicity against HEP-G2 (human hepatocellular carcinoma) and murine lymphoma (L5178Y-R) cells using the MTT assay of the methanol extract (AmexM), sub-partitions of A. mexicana, and BER. Selectivity indices (SIs) were determined by comparing their cytotoxic effects on VERO (monkey kidney epithelial) and PBMC (human peripheral blood mononuclear) non-tumoral cells. Additionally, the anti-hemolytic effect of these treatments was assessed using the AAPH method. The treatment with the most promising activity against tumor cells and anti-hemolytic efficacy underwent further evaluation for toxicity in Artemia salina and antioxidant activities using DPPH, ABTS, and FRAP assays. BER demonstrated an IC50 = 56.86 µg/mL in HEP-G2 cells and IC50 < 5.0 µg/mL in L5178Y-R cells, with SI values of 15.97 and >5.40 in VERO and PBMC cells, respectively. No significant hemolytic effects were observed, although AmexM and BER exhibited the highest anti-hemolytic activity. BER also demonstrated superior antioxidant efficacy, with lower toxicity in A. salina nauplii compared to the control. Additionally, BER significantly attenuated nitric oxide production. This study highlights the antiproliferative effects of A. mexicana, particularly BER, against HEP-G2 and L5178Y-R tumor cell lines, along with its selectivity towards normal cells. Furthermore, its anti-hemolytic and antioxidant potentials were demonstrated, suggesting that BER is a promising candidate for potent chemotherapeutic agents. Full article

Soybean is a high-value food source, and the invasive weeds Mexican prickly poppy (Argemone ochroleuca) could release allelochemicals that inhibit the growth of this crop. The impact of A. ochroleuca on the germination and growth of soybean is not well documented. Therefore, the aim of this study was to evaluate the TLC profiles of different extracts of A. ochroleuca and assess the effects of extracts on the germination of soybean seeds. Shoots and roots of A. ochroleuca were weighed and 100 g of each was separately extracted with 1000 mL deionized water, hexane or acetone. Ten concentrations of water extracts ranging from 10 to 100 mL per 100 mL of deionized water and three concentrations of acetone and hexane extracts ranging from 2.5 to 7.5 g/L were separately used for seed germination bioassays. Thin-layer chromatography (TLC) analysis was used to compare the chemical profiles in the shoot and root water, and in the hexane and acetone extracts of A. ochroleuca. The highest reduction was recorded from the water extract, at 100%. The TLC profiling of A. ochroleuca addressed different classes of compounds, including alkaloids, phenolic acids and flavanoids. There is, however, a need to identify the most active phytochemicals in the suppression of germination. Full article

Understanding the strategies and mechanisms of invasive species could guide their control and management especially in arid ecosystems. This study compares the vegetative and reproductive functional traits of the invasive Mexican poppy (Argemone ochroleuca), in seven habitat types, in southwestern Saudi Arabia. The results showed that the aboveground phenological attributes such as plant height, leaf area, and leaf dry mass attained the highest values in the wadi channels, whereas these attributes attained the lowest values in the mountain ranges. Maximum specific leaf area, root parameters, and all reproductive traits were recorded in the abandoned fields. In contrast to all other habitats, populations from abandoned fields had a greater investment of resources in belowground structures, while the population growing in the wadi channels and mountain ranges habitat allocated more energy to vegetative parts. The plasticity in vegetative and reproductive resource allocation in A. ochroleuca is an important mechanism in determining its colonizing ability in different habitat types and expanding the distribution range. The present data of the functional traits of A. ochroleuca agree with the resource fluctuation hypothesis, where the plant flourished in the abandoned fields that attained the highest values of organic matter and nutrients. Therefore, the restoration of these disturbed habitats could improve the resistance toward invasion by this noxious weed. Full article
