Coastal lagoons play an essential role in the energy balance and heat exchange to the atmosphere. Furthermore, at mesoscale Monsoon systems and at local scales, sea breeze influences surface processes; however, there is a lack of information on such processes in arid and semiarid regions. We aimed to characterize the atmospheric conditions during sea and land breeze in different seasons and analyze at different temporal scales the variation of atmospheric stability, turbulent fluxes, lifting condensation level, and atmospheric boundary layer height. The study site is a subtropical semiarid coastal lagoon, Estero El Soldado, located in Northwestern Mexico (27°57.248′ N, 110°58.350′ W). Measurements were performed from January 2019 to September 2020 with an Eddy Covariance system (EC) and micrometeorological instruments over the water surface. Results show that there is a strong seasonality that enhances sea–land breeze dominance; sea breeze was 83% more frequent during the Monsoon, and the land breeze was 55% more frequent in the Post-Monsoon. Specific humidity (23.32 ± 3.84 g kg−1, q), potential temperature (307 ± 2.98 K, θp), latent heat (135 W m−2, LE), and turbulent kinetic energy (0.81 m2 s−2, TKE) were significantly higher during the Monsoon season at sea breeze events. Atmospheric boundary layer (ABL) and lifting condensation level (LCL) were higher in the Pre-Monsoon season (3250 ± 71 m and 1142 ± 565 m, respectively). During the Monsoon, surface conditions lead to lower LCL (~800 m) due to the amount of water vapor (q = 23.3 g kg−1). Full article

Despite the critical role latent (LE) and sensible (H) heat play in turbulent processes and heat exchange in the water–air interface, there is a lack of studies of turbulent fluxes over the surface in semiarid regions. We collected continuous measurements of net radiation (Rn), LE, H, and micrometeorological data at a coastal lagoon in the Gulf of California during 2019 with an eddy covariance (EC) system. We analyzed the time series, considering the North American Monsoon System, the pre-monsoon, and post-monsoon season. Results show that Rn (276 ± 118 W m−2) and turbulent fluxes were higher during the monsoon season (July–September) LE (129 ± 18 W m−2), and H (29 ± 9 W m−2). The monthly average of Rn, LE, and H was highest in June (493.9 W m−2), August (142 W m−2), and May (50 W m−2), respectively. Furthermore, during the monsoon season, the (H + LE)/Rn ratio (0.74) suggests that more than half of the Rn reaching the coastal lagoon is used for the turbulent exchange of LE and H. During the pre-monsoon, LE (r2 = 0.36) increases with a higher vapor pressure deficit (VPD), while H (r2 = 0.66) increases with a higher friction velocity (u*) during the monsoon season. MicroRNA Let-7d-3p Contributes to Cardiac Protection via Targeting HMGA2

We tested the hypothesis that Let-7d-3p contributes to cardiac cell protection during hypoxic challenge. Myoblast H9c2 cells and primary neonatal rat ventricular cardiomyocytes (NRVM) were transfected with five selected miRNA mimics. Both cell lines were subjected to 0.2% oxygen hypoxia. The protective effects of these miRNAs were determined by assessment of cell metabolic activity by CCK8 assay and measurement of lactate dehydrogenase (LDH) release as a marker of cell injury. Apoptosis and autophagy flux were assessed by Annexin V/7-AAD double staining and the ratio of LC3 II/I with Baf-A1 treatment, an autophagy flux inhibitor, respectively. Luciferase-reporter assay, RT-qPCR and Western blots were performed to identify the changes of relevant gene targets. Among five miRNA mimic transfections, Let-7d-3p increased CCK8 activity, and decreased LDH release in both H9c2 and NRVM during hypoxia. Apoptosis was significantly reduced in H9c2 cells transfected with Let-7d-3p mimic. Autophagy and autophagy flux were not affected. In silico, mRNAs of HMGA2, YY1, KLF9, KLF12, and MEX3C are predicted targets for Let-7d-3p. Luciferase-reporter assay confirmed that Let-7d-3p bound directly to the 3’-UTR region of HMGA2, MEX3C, and YY1, the down-regulations of these mRNAs were verified in both H9c2 and NRVM. The protein expression of HMGA2, but not others, was downregulated in H9c2 and NRVM. It is known that HMGA2 is a strong apoptosis trigger through the blocking of DNA repair. Thus, we speculate that the anti-apoptotic effects of Let-7d-3p mimic during hypoxia challenge are due to direct targeting of HMGA2. Full article

One challenge in studying the second messenger inositol(1,4,5)-trisphosphate (InsP3) is that it is present in very low amounts and increases only transiently in response to stimuli. To identify events downstream of InsP3, we generated transgenic plants constitutively expressing the high specific activity, human phosphatidylinositol 4-phosphate 5-kinase Iα (HsPIPKIα). PIP5K is the enzyme that synthesizes phosphatidylinositol (4,5)-bisphosphate (PtdIns(4,5)P2); this reaction is flux limiting in InsP3 biosynthesis in plants. Plasma membranes from transgenic Arabidopsis expressing HsPIPKIα had 2–3 fold higher PIP5K specific activity, and basal InsP3 levels in seedlings and leaves were >2-fold higher than wild type. Although there was no significant difference in photosynthetic electron transport, HsPIPKIα plants had significantly higher starch (2–4 fold) and 20% higher anthocyanin compared to controls. Starch content was higher both during the day and at the end of dark period. In addition, transcripts of genes involved in starch metabolism such as SEX1 (glucan water dikinase) and SEX4 (phosphoglucan phosphatase), DBE (debranching enzyme), MEX1 (maltose transporter), APL3 (ADP-glucose pyrophosphorylase) and glucose-6-phosphate transporter (Glc6PT) were up-regulated in the HsPIPKIα plants. Our results reveal that increasing the phosphoinositide (PI) pathway affects chloroplast carbon metabolism and suggest that InsP3 is one component of an inter-organelle signaling network regulating chloroplast metabolism.
