Background: Three herbal extracts (Asparagus racemosus Willd., Tabebuia avellanedae Lorentz, and Glycyrrhiza glabra L.) were mixed with three essential oils (Foeniculum vulgare Mill., Mentha piperita L., and Pimpinella anisum L.) to formulate a product (HEMEO) whose active compounds include saponins and steroids in Asparagus racemosus, known for their anti-inflammatory properties; glycyrrhizin and flavonoids in Glycyrrhiza glabra, which exhibit gastroprotective and antispasmodic effects; menthol in Mentha piperita, contributing with antispasmodic and antimicrobial properties; and anethole and polyphenols in Pimpinella anisum, which modulate intestinal motility and offer antimicrobial activity. Objective: HEMEO was formulated for applications in intestinal motility disorders. Methods: HEMEO was evaluated for spontaneous and induced motility effects in isolated guinea pig ileum, colon, and stomach. Ex vivo experiments were conducted using LabChart software v7.0, and the product’s antibacterial action against Helicobacter pylori and its antioxidant effects were assessed through disc diffusion and FRAP assays. The presence of the volatile compounds in the formulation was confirmed by GC-MS analysis; the TPC of HEMEO, determined using the Folin–Ciocalteu method, was 9.925 ± 0.42 mg GAE/g. Conclusions: HEMEO showed a phenolic content correlated with its antioxidant potential and in addition inhibited H. pylori growth and demonstrated notable antioxidant properties, suggesting its role as a supportive agent in digestive processes and in managing motility disorders. This work describes a straightforward implementation of detecting and tracking pedestrian walking across a public square using computer vision. The methodology consists of the use of the well-known YOLOv3 algorithm over videos recorded during different days of the week. The chosen location was the Piazza Duca d’Aosta in the city of Milan, Italy, in front of the main Centrale railway station, an access point for the subway. Several analyses have been carried out to investigate macroscopic parameters of pedestrian dynamics such as densities, speeds, and main directions followed by pedestrians, as well as testing strengths and weaknesses of computer-vision algorithms for pedestrian detection. The developed system was able to represent spatial densities and speeds of pedestrians along temporal profiles. Considering the whole observation period, the mean value of the Voronoi density was about 0.035 person/m2 with a standard deviation of about 0.014 person/m2. On the other hand, two main speed clusters were identified during morning/evening hours. The largest number of pedestrians with an average speed of about 0.77 m/s was observed along the exit direction of the subway entrances during both morning and evening hours. The second relevant group of pedestrians was observed walking in the opposite direction with an average speed of about 0.65 m/s. The analyses generated initial insights into the future development of a decision-support system to help with the management and control of pedestrian dynamics. Full article

The impossibility of using drugs for the health of farm animals leads to the search for alternative strategies with two purposes: to maintain animal health and safeguard human health. In this perspective, tannins have shown great promises. These phytocomplexes obtained from natural matrices with multiple health properties may be used as a feed supplement in chicken farms. In this work, we studied two tannin-based extracts (from Castanea sativa Mill. wood and from Schinopsis balansae Engl. Quebracho Colorado hardwood) with different chemical compositions on the spontaneous contractility on the isolated intestinal tissues of healthy chicken. The results showed that the chemical composition of the two phytocomplexes influenced the spontaneous intestinal contractility in different ways by regulating the tone and consequent progression of the food bolus. The chemical analysis of the two extracts revealed that Castanea sativa Mill. wood mainly contains hydrolysable tannins, while Schinopsis balansae Engl. hardwood mainly contains condensed tannins. The two phytocomplexes showed different effects towards gastrointestinal smooth muscle contractility, with Castanea sativa Mill. wood providing a better activity profile than Schinopsis balansae Engl. hardwood. In this work, a new methodology to estimate the functional vulnerability of the road network of the city of Catania (Italy) is developed with the purpose to improve the resilience of urban transport during critical events. While the traditional approach for the estimation of vulnerability is based on topological data, the proposed methodology is based on spatial-temporal mobility profiles obtained with floating car data (FCD). The algorithm developed for the estimation of vulnerability combines topological properties of the road network with mobility patterns obtained from FCD to evaluate the consequences of failure events on trajectories and their associated travel times. The core operation of the algorithm is based on the computation of all possible travel paths within their assigned geographical zone every time a road link is disrupted. The procedure may prove useful to evaluate wide failure events and to facilitate emergency plans. Full article

TAR DNA-binding protein 43 (TDP-43) forms intraneuronal cytoplasmic inclusions associated with amyotrophic lateral sclerosis and ubiquitin-positive frontotemporal lobar degeneration. Its N-terminal domain (NTD) can dimerise/oligomerise with the head-to-tail arrangement, which is essential for function but also favours liquid-liquid phase separation and inclusion formation of full-length TDP-43. Using various biophysical approaches, we identified an alternative conformational state of NTD in the presence of Sulfobetaine 3-10 (SB3-10), with higher content of α-helical structure and tryptophan solvent exposure. NMR shows a highly mobile structure, with partially folded regions and β-sheet content decrease, with a concomitant increase of α-helical structure. It is monomeric and reverts to native oligomeric NTD upon SB3-10 dilution. The equilibrium GdnHCl-induced denaturation shows a cooperative folding and a somewhat lower conformational stability. When the aggregation processes were compared with and without pre-incubation with SB3-10, but at the identical final SB3-10 concentration, a slower aggregation was found in the former case, despite the reversible attainment of the native conformation in both cases. This was attributed to protein monomerization and oligomeric seeds disruption by the conditions promoting the alternative conformation. Overall, the results show a high plasticity of TDP-43 NTD and identify strategies to monomerise TDP-43 NTD for methodological and biomedical applications. Full article

Waste represents a cost for companies, in particular for agro-food companies, which can become a resource as a secondary material. In this work, we examine three products of olive-oil waste water, named MOMAST® (Plus30, PW25, and HY100). Based on the chemical composition, obtained with different methods, we hypothesized a possible application as food supplements in irritable bowel syndrome (IBS). We therefore studied MOMASTs on some targets linked to this pathology: antioxidant action and spontaneous and induced intestinal contractility of the ileum and colon. Plus30, which showed a more promising biological of activity also for its oleuropein content, was characterized by an interesting action against some microorganisms. The results highlighted the ability of Plus30 to modulate spontaneous and induced contractility, to exert a good antioxidant action, and to significantly act on various microorganisms. These effects are synergistic in the presence of antibiotics. In conclusion, we can confirm that Plus30 could be a great candidate as a food supplement in patients with IBS. Full article

The main purpose of this review is to analyze published data concerning the antioxidant properties of astaxanthin, a xanthophyll, produced by the microalga Haematococcus pluvialis in response to specific conditions of “environmental stress” and characterized by its typical deep red color. Natural astaxanthin establishes effective protections against oxidative stress, neutralizing free radicals in both the inner and outer layer of cell membranes, especially in mitochondria. The most recent preclinical and clinical studies that have investigated the beneficial properties of this molecule toward the gastrointestinal tract were included. Full article

It is well known that diet and nutrition play a critical role in the etiopathogenesis of many disorders. On the other hand, nutrients or bioactive compounds can specifically target and control various aspects of the mechanism underlying the pathology itself, and, in this context, diseases related to intestinal motility disorders stand out. The Herbal Mix (HM) consisting of Olea europea L. leaf (OEE) and Hibiscus sabdariffa L. (HSE) extracts (13:2) has been proven to be a promising nutraceutical option for many diseases, but its potential in inflammatory-driven gastrointestinal disorders is still unexplored. In this study, HM effects on guinea-pig ileum and colon contractility (induced or spontaneous) and on human iNOS activity, as well as on human colorectal adenocarcinoma Caco-2 cells, were studied. Results showed that the HM can control the ileum and colon contractility without blocking the progression of the food bolus, can selectively inhibit iNOS and possesses a strong pro-apoptotic activity towards Caco-2 cells. In conclusion, the present results suggest that, in some diseases, such as those related to motility disorders, an appropriate nutritional approach can be accompanied by a correct use of nutraceuticals that could help not only in ameliorating the symptoms but also in preventing more severe, cancer-related conditions. Public Transport (PT) planning requires a detailed evaluation of the fleet energy consumption, usually depending on the specific characteristics of the bus service network. These include topography, climate and operational features. In this work, we focus on the additional air conditioning (AC) energy consumption, proposing a method to evaluate the extra energy consumption based not only on climate variables, but also on the PT planned service. Results are presented for a large part of the provincial capitals and regions of Italy, and clearly show that overconsumption for air conditioning are significantly affected by the daily mileage distribution, with large variance even when climatic conditions are similar. The mileage data are extracted from GTFS databases, widely available for PT applications. The developed tool allows us to apply this methodology to any urban and extra-urban area. Reference AC consumption related to climate conditions are derived from a measurement campaign hold in Cagliari (Sardinia, Italy) during September 2018, within the National Research Program on the Electric System. A discussion on how to optimize the use of climatic data is also presented, resulting in the choice to use Heat Index as unique independent variable for air conditioning energy consumption calculation. A methodology to compute the Heat Index from climatic variables for large domains as, for instance, the Italian regions, was also developed. Full article

A new Thymus vulgaris L. solid essential oil (SEO) formulation composed of liquid EO linked to solid excipients has been chemically analysed and evaluated for its intestinal spasmolytic and antispastic effects in ex vivo ileum and colon of guinea pig and compared with liquid EO and excipients. Liquid EO and solid linked EO were analysed by original capillary electrochromatography coupled to diode array detection (CEC-DAD) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) methodologies. The main bioactive constituents are thymol and carvacrol, with minor constituents for a total of 12 selected analysed compounds. Liquid EO was the most effective in decreasing basal contractility in ileum and colon; excipients addiction permitted normal contractility pattern in solid linked EO SEO. In ileum and colon, the Thymus vulgaris L. solid formulation exerted the relaxant activity on K+-depolarized intestinal smooth muscle as well as liquid EO. The solid essential oil exhibits antimicrobial activity against different strains (Staphylococcus aureus, Streptococcus pyogenes, Pseudomonas aeruginosa, Escherichia coli, Salmonella Thyphimurium, Candida albicans) similarly to liquid oil, with activity against pathogen, but not commensal strains (Bifidobacterium Breve, Lactobacillus Fermentum) in intestinal homeostasis. Therefore, Thymus vulgaris L. solid essential oil formulation can be proposed as a possible spasmolytic and antispastic tool in medicine. Full article
