LPCAT4 Knockdown Alters Barrier Integrity and Cellular Bioenergetics in Human Urothelium

Urothelium is a transitional, stratified epithelium that lines the lower urinary tract, providing a tight barrier to urine whilst retaining the capacity to stretch and rapidly resolve damage. The role of glycerophospholipids in urothelial barrier function is largely unknown, despite their importance in membrane structural integrity, protein complex assembly, and the master regulatory role of PPARγ in urothelial differentiation. We performed lipidomic and transcriptomic characterisation of urothelial differentiation, revealing a metabolic switch signature from fatty acid synthesis to lipid remodelling, including 5-fold upregulation of LPCAT4. LPCAT4 knockdown urothelial cultures exhibited an impaired proliferation rate but developed elevated trans-epithelial electrical resistances upon differentiation, associated with a reduced and delayed capacity to restitute barrier function after wounding. Specific reduction in 18:1 PC fatty acyl chains upon knockdown was consistent with LPCAT4 specificity, but was unlikely to elicit broad barrier function changes. However, transcriptomic analysis of LPCAT4 knockdown supported an LPC-induced reduction in DAG availability, predicted to limit PKC activity, and TSPO abundance, predicted to limit endogenous ATP. These phenotypes were confirmed by PKC and TSPO inhibition. Together, these data suggest an integral role for lipid mediators in urothelial barrier function and highlight the strength of combined lipidomic and transcriptomic analyses for characterising tissue homeostasis. This study explores Industry 4.0 in the Australian context, where manufacturing is dominated by smaller firms servicing the domestic market. We interviewed 20 advanced Australian manufacturers from diverse fields, capturing detailed descriptions of the Industry 4.0 implementation process. We compared the themes that emerged from their descriptions with the factors typically assessed in Industry 4.0 frameworks to draw out differences in emphasis. Consistent with these frameworks, Australian manufacturers were focused on using Industry 4.0 technologies to automate the capture, integration and analysis of data. To enable this activity, they were reorganising workforce roles and developing new skill sets. Knowledge sharing and collaborations within and across the organisation were seen to be especially important for small Australian manufacturers (with limited funding for technology investment and research and development) to maintain global competitiveness. However, while most Industry 4.0 frameworks describe supply chain applications, the development of smart products and services, and the need to adopt a strategy-led approach, relatively few participants spoke about these opportunities. Even fewer addressed the need for improved governance, standards and data security in the context of Industry 4.0. We argue that these gaps are best addressed through government policy and investment focusing beyond manufacturers, to support Industry 4.0 uplift across key domestic supply chains. Full article

Action Levels (ALs) are thresholds which are used to determine whether dredged material is suitable for disposal at sea by providing a proxy risk assessment for potential impacts to biological features such as fish and benthos. This project tested proposed scenarios for changes to the UK Action Levels to determine the likely implications for navigational dredge licensing in England and Wales. Approximately 3000 sample data records from 2009 to 2018 were collated with varying numbers of concentrations for contaminant parameters including trace metals, organotins, polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs) and polybrominated diphenyl ethers (PBDEs). Initially, these data were assessed using current ALs to determine the percentages of the samples with levels below AL1 (generally acceptable for disposal), between AL1 and AL2 and those showing levels above AL2 (generally unacceptable for disposal). These results were then used to compare with the results of the proposed new AL scenarios for each contaminant type derived from literature reviews and historic data. The results indicate that there are changes to the ALs which could be made such as updating the current ALs with the revised ALs, as well as the introduction of ALs where there are currently none set. The benefits of changing the ALs include reducing contaminant disposal to the marine environment and increased transparency in decision making. Any proposed scenarios will need to be phased in carefully in full liaison with stakeholders. Full article

Iodine is essential for normal thyroid function, supporting healthy fetal and child development. Iodine requirements increase in pregnancy, but many women in regions without salt iodization have insufficient intakes. We explored associations between iodide intake and urinary iodine concentration (UIC), urinary iodine/creatinine ratio (I/Cr), thyroid stimulating hormone, thyroglobulin, free triiodothyronine, free thyroxine and palpable goiter in a region of mild-to-moderate iodine insufficiency. A total of 246 pregnant women aged 18–40 in Bradford, UK, joined the Health and Iodine in Babies (Hiba) study. They provided detailed information on diet and supplement use, urine and serum samples and were assessed for goiter at around 12, 26 and 36 weeks’ gestation, and 6, 18 and 30 weeks postpartum. Dietary iodide intake from food and drink was estimated using six 24 h recalls. During pregnancy, median (IQR) dietary iodide intake was 101 µg/day (54, 142), with 42% from dairy and 9% from white fish. Including supplements, intake was 143 µg/day (94, 196), with 49% < UK reference nutrient intake (140 µg/day). Women with Pakistani heritage had 129 µg/day (87, 190) median total intake. Total intake during pregnancy was associated with 4% (95% CI: 1%, 7%) higher UIC, 5% (3%, 7%) higher I/Cr, 4% (2%, 6%) lower thyroglobulin and 21% (9%, 32%) lower odds of palpable goiter per 50 µg/day. This cohort consumed less iodide in pregnancy than UK and World Health Organization dietary recommendations. UIC, I/Cr and thyroglobulin were associated with intake. Higher intake was associated with fewer goiters. Because dairy was the dominant source of iodide, women following plant-based or low-dairy diets may be at particular risk of iodine insufficiency. Building on the findings from the national study of mothers in recurrent care proceedings in England, this paper proposes that the concepts of complex trauma and epistemic trust may help explain parents’ difficulties in engaging with child protection services. With the aim of advancing theoretical knowledge, qualitative data drawn from interviews with 72 women who have experienced repeat care proceedings are revisited, with a focus on women’s developmental histories and accounts of engagement with professionals, to probe the issue of service engagement. The article starts with a succinct review of the literature on parental non-engagement in child protection, highlighting strengths and potential limitations of current knowledge. This is followed by an introduction to the theoretical concepts of complex trauma and epistemic trust, outlining how these concepts provide an alternative framing of the reasons why parents may resist, or are reluctant to engage with, professionals. Drawing on women’s first-person accounts, we argue that high levels of maltreatment and adversity in women’s own childhoods shape adult relationships, particularly in relation to vulnerability to harm in adult lives but also mistrust of professional help. Extracts from women’s first-person accounts, chosen for their typicality against the core themes derived from the data, indicate that acts of resistance or rejection of professional help can be seen as adaptive—given women’s childhoods and relationship histories. The authors conclude that parents’ social histories need to be afforded far closer attention in child protection practice, if preventative services are to reach those with histories of developmental trauma. Recent studies of public law care proceedings within the family justice system in England and Wales suggest that up to a quarter of all mothers who appear in such proceedings will reappear within a subsequent—or recurrent—set of such proceedings within seven years. In the last decade, new interdisciplinary research spanning social work, clinical psychology and sociology has defined and investigated the previously hidden challenge and social costs of ‘recurrent care proceedings’ (RCP). This article adds to this new field by analysing the core values, practice and impact of three different local services in the northwest of England working with birth parents to reduce the risk of recurrent proceedings. The article combines data gathered from the three distinct services using a common evaluation framework co-produced by the authors working with service leads, practitioners and users. It explores how all three services are seeking to reduce the risk of recurrent care proceedings in their local areas without requiring women to use long acting reversible contraception (LARC) or other forms of contraception as a condition of accessing the service. It concludes that insights gained from these and cognate services can inform an emergent community of practice in the recurrent care field. Full article

The UK food industry faces significant challenges to remain sustainable. With major challenges, such as Brexit, on the horizon, companies can no longer rely on a low labour cost workforce to maintain low production costs and achieve economic sustainability. Smart Systems (SS) is being seen as an approach towards achieving significant improvements in both economic and environmental sustainability. However, there is little evidence to indicate whether UK food companies are prepared for the implementation of such systems. The purpose of this research is to explore the applicability of Smart Systems in UK food manufacturing companies, and to identify the key priority areas and improvement levers for the implementation of such systems. A triangulated primary research approach is adopted that includes a questionnaire, follow-up interviews, and visits to 32 food manufacturing companies in the UK. The questionnaire and interviews are guided by a unique measuring instrument that the authors developed that focusses upon SS technologies and systems. This paper makes an original contribution in that it is one of the few academic studies to explore the implementation of SS in the industry, and provides a new perspective on the key drivers and inhibitors of its implementation. The findings suggest that the current turbulence in the industry could be bringing food companies closer to the adoption of such systems; hence, it is a good time to define and develop the optimum SS implementation strategy. Ground-penetrating radar (GPR) offers many advantages for assessing archaeological potential in frozen and partially frozen contexts in high latitude and alpine regions. These settings pose several challenges for GPR, including extreme velocity changes at the interface of frozen and active layers, cryogenic patterns resulting in anomalies that can easily be mistaken for cultural features, and the difficulty in accessing sites and deploying equipment in remote settings. In this study we discuss some of these challenges while highlighting the potential for this method by describing recent successful investigations with GPR in the region. We draw on cases from Bering Land Bridge National Preserve, Cape Krusenstern National Monument, Kobuk Valley National Park, and Gates of the Arctic National Park and Preserve. The sites required small aircraft accessibility with light equipment loads and minimal personnel. The substrates we investigate include coastal saturated active layer over permafrost, interior well-drained active layer over permafrost, a frozen thermo-karst lake, and an alpine ice patch. These examples demonstrate that GPR is effective at mapping semi-subterranean house remains in several contexts, including houses with no surface manifestation. GPR is also shown to be effective at mapping anomalies from the skeletal remains of a late Pleistocene mammoth frozen in ice. The potential for using GPR in ice and snow patch archaeology, an area of increasing interest with global environmental change exposing new material each year, is also demonstrated. Full article
