Titanic review on effects of statins on macrophage's populations.

As stated by the authors, this field is filled with contradicting experimental results and is not widely covered in the literature by a comprehensive review on it. The authors fill the gap nicely with this manuscript.

It is exhaustive, organized and well written. 

Few suggestions: for clarity and readability, please increase resolution and size of Fig. 3. Ideally, one entire page should be dedicated to fig. 3A and the following entire page dedicated to fig. 3B.

A careful check of the reference is warranted to ensure quality and consistency (i.e.: ref. 52)

As stated by the authors, this field is filled with contradicting experimental results and is not widely covered in the literature by a comprehensive review on it. The authors fill the gap nicely with this manuscript.

  • We thank the reviewer for considering our manuscript and taking the time to write their positive feedback.

Few suggestions: for clarity and readability, please increase resolution and size of Fig. 3. Ideally, one entire page should be dedicated to fig. 3A and the following entire page dedicated to fig. 3B.

  • We agree and have now divided Fig.3A and Fig.3B into two separate figures (Fig.3 and Fig.4) on two separate pages. The size of each of these figures is now significantly larger and the resolution has been increased. The in-text citations of these figure has also been updated throughout the manuscript to reflect these changes.

A careful check of the reference is warranted to ensure quality and consistency (i.e.: ref. 52)

  • We thank the reviewer for their attention to detail and have reformatted the inconsistent references including ref 52.

The review can be accepted in the present form. It will be beneficial for the readers if the authors can possibly reduce the number of references and just reference the recent paper.

The authors have provided a very extensive report on immunomodulatory effect of statins. The review is well organized and every section is explained in detail, including the tables. The review can be accepted in the present form.

  • We thank the reviewer for considering our manuscript and taking the time to write their positive feedback.

It will be beneficial for the readers if the authors can possibly reduce the number of references and just reference the recent paper.

  • We have removed 11 older references (for example outdated reviews) which we felt in retrospect did not add significant value to our manuscript. We believe this makes our manuscript clearer and hope our changes satisfies the reviewer.

Authors have tried to compile pro and anti-inflammatory effect of statins on macrophage by referring to published articles. Abstract is written more precisely. However, there are some things are missing.

Comments:

  1. In table1: Pharmacokinetic data of stains is not as per reference 15, please verify and correct it.
  2. Evidence has emerged from cell culture experiments, animal studies and clinical trials that statins can affect macrophage inflammatory responses, please cite relevance references.

Authors have tried to compile pro and anti-inflammatory effect of statins on macrophage by referring to published articles. Abstract is written more precisely. However, there are some things are missing.

  • We thank the reviewer for taking this time to consider our manuscript and their comments which we hope we have addressed to their satisfaction.
  • We have re-worded our abstract now to more precisely describe the contents of our review including ‘macrophage polarization and immunomodulatory functions’.

Comments:

  1. In table1: Pharmacokinetic data of stains is not as per reference 15, please verify and correct it.

 

  • We agree that the presentation of our original table could be improved and so have now moved the reference citations from a summary column to the individual properties they are referring to within the table - including reference 15. We feel this makes following-up on the individual references clearer and more intuitive and we hope the reviewer agrees.

 

  1. Evidence has emerged from cell culture experiments, animal studies and clinical trials that statins can affect macrophage inflammatory responses, please cite relevance references.
  • We have re-worded this sentence in the introduction to more clearly convey that these are the topics that our review will cover, with the relevant references cited throughout the body of the main text. We hope this modification addressed the reviewer’s suggestion.
