Nose-to-Brain Targeting via Nanoemulsion: Significance and Evidence

This is a good comprehensive review on the nasal administration of various therapeutic drugs to brain diseases. The followings are my specific comments:
(1) Compare with other sections, “future prospect is too short and some suggestions are more like common sense. For example, of course formulation is important, but how to pursue new formulation? What are the new efforts on that topic, a brief description is needed. The author mentioned in situ nanoemulsion, which is very good. Some extension on how that can be done will be valuable to readers.
(2) I would suggest combining the conclusion and prospective together as both are quite short. Again at least extension on the new directions are needed.
(3) I saw in quite some places that two words are together without space (for example, in table 3, therapeutic agent was shown as “therapeuticagent”, check for many other places, I saw at least ten places like this). The authors may want to carefully check on that types of errors. Besides, some words or phrases are strange. For example, what is “bioavailability” on page 22 line 629. On page 2, line 55, what is “remove lipophilic toxin tend to diffuse cell membrane” looks to me as grammar errors.
(4) The title of the review says: Nose to brain targeting via nanoemulsion”. I did not see any “targeting probe or strategies being reviewed in this manuscript. By the way, this is a good direction if the authors tried to expand the prospective part

From my point of view, the paper here presented constitutes an interesting bibliographic revision about the state-of-the-art of nasal administration of drugs and their enhanced absorption to the central nervous system by the use of nano-emulsions to obtain an effective drug targeting. I consider that the extensive and well-organized revision gives a clear and precise overview of the most reported uses of this technology.

This is a very interesting, voluminous, beautifully illustrated article. The authors have done a great job, analyzing the available volume of modern scientific literature. Perhaps it makes sense for the authors to discuss the effect of nanoparticle size on brain cells. https://pubmed.ncbi.nlm.nih.gov/35806466/
