This is a very interesting manuscript on the mitotane and its interaction/action on the estrogen receptor. The manuscript raises an important clinical problem and approaches it both via structural chemical and in vitro pharmacological cell line experiments. The manuscript is well-written, easy to follow, and the applied methodology, results, discussion and the conclusions are clear. The results have clear clinical relevance, as they seem to explain clinical findings, and can form the basis of further translational studies that could eventually lead to better compounds in the combat against adrenocortical cancer.
