This review is of interest in the global knowledge of Alzheimer's disease, the main problem with it is that there is to much information and as it is structured it is difficult to follow.

There are many concepts and different approaches that should explained in a more schematic way. Even the figure need some more explanation in order to understand it fully.

Perhaps the authors could use some index in the more complex sections in order to separated and explain in a separate way the different aspects.
