The authors investigated the mechanisms of Aloe Vera treatment on rat thyroid cell line. The work is important, but the text has some deficiencies. Please, find hereby, a point-by-point comment list to the manuscript.

 There are many unreferenced statements and the writing is not adequate throughout the manuscript.

 1-Abstract. The first sentence (“Homeostasis is important for maintaining a healthy body”) must be withdrawn.

 2-Abstract. The authors wrote in the abstract and throughout the text Aloe Vera, A. vera and Aloe Vera in italics in the keywords. What is the correct way? The authors must standardize the term used.

3- Abstract. Line 22. The full name of AKT is missing.

4-Introduction. Page 2, 2nd paragraph, line 45-46. The sentences” Homeostasis is considered important in oriental medicine. We evaluated whether herbal medicines contributed to endocrine tissue homeostasis” must be withdrawn.

5-Introduction. Page 2, 3rd paragraph. The entire paragraph must be rewritten. The language is truncated.

6-Materials and Methods. Cell Culture and treatment. Authors must justify the dose of Aloe Vera used. Were no cytotoxicity assays used? Have the cells been tested for the presence of mycoplasma.

7-Materials and Methods. Cell proliferation. The production of formazan with the consequent formation of WST is mainly used for toxicity tests and determination of inhibitory doses and effective doses of compounds tested in vitro. Why did the authors use this method for cell proliferation?

8-Materials and Methods. Real-time PCR. Why the authors only evaluated the transcriptional levels of NIS, TG and TPO? Why was mRNA expression not analyzed at the protein level?

9-Results. In all figures, the graphics must have a title.

10-Results. How many experiments with the cells were performed to obtain the presented results? This must be indicated in the figure legends.

11- Many results were presented as a comparison between cells treated with Aloe Vera with the control in the respective treatment media (5H or 6H). It is this correct. However, there are comparisons between the treatment with Aloe Vera in one media (5H or 6H) with the control in another media (5H or 6H; please see Figures 2, 3, 4 and 5). Why did the authors make these comparisons?

12- Discussion.  The discussion should be rewritten. There are many spelling errors. This compromises the understanding of the manuscript.

13-Discussion. Figure 6. All abbreviations must be defined in the figure legend. The proposed mechanism should also be explained in the figure legend.

Dear authors,

The manuscript entitled "Effects of Aloe vera on the regulation of thyroxine release in FRTL-5 thyroid cells” describes  was to evaluate the ability of A. vera to maintain homeostasis, similar to TSH. We examined the role of A. vera in regulating the cellular export of thyroid hormones and its cellular mechanism in FRTL-5 thyroid cells. It presents scientific relevance for the Food Science and Herbal Medicines area.

However, you need to change some details/information in the abstract, Introduction, Material and Methods, Results and discussion.

- Please check the word count for your abstract.

- I suggest specify the temperature of the  A. vera water extract. Is there any reference protocol in the literature?

-Please specify the name of the abbreviations from table 5

Conclusion: I suggest to indicate disadvantages/limitations of the method and the study! Perhaps, to highlight the text in the 'Limits of the study' section (extend the conclusions).

References: Spelling of references must be checked to meet the journal style.

The authors incorporated in the text some corrections and suggestions proposed. However, the text still contains statements without references, and the English language still needs improvement. In figure legends, would not it be n=3 instead of n≥3? Graph titles have not been entered. Would this be an option of the authors?
