This manuscript describes the role of pyruvate carboxylase (PYC) in regulation of D-aspartate oxidase (DDO) gene expression. PYC was found to modulate DDO gene expression through metabolism of pyruvate to oxaloacetate, with pyruvate acting as a negative regulator of DDO expression and oxaloacetate exerting positive regulation.

This study was thorough and the conclusions are sound. The manuscript is well-written.

Two (very) minor issues:

  1. In Figure 2, the p-value should be indicated by ** for consistency with other figures.
  2. In Figure 6, it is unnecessary to go beyond the standard **** for p-value<0.0005.

POINT 1:  I reccomend the following title "Regulation of D-Aspartate oxidase gene expression by pyruvate metabolism in the yeast Cryptococcus humicola"

According to the reviewer’s suggestion, we have changed the previous title with the proposed title “Regulation of d-Aspartate Oxidase Gene Expression by Pyruvate Metabolism in the Yeast Cryptococcus humicola.”

POINT 2: Check block letters in "D-Aspartate oxidase" (Lines 2, 8, 22, 26 )

We thank the reviewer for the suggestion. We have checked the block letters in the “d-Aspartate oxidase.” We have used the word "d-Aspartate oxidase" in the title and at the beginning of a sentence and "d-aspartate oxidase" at the middle of a sentence and in the keywords, according to the journal’s submission guidelines.

Another correction:

In Figure 7, we have changed the English error "Nuclear" to "Nucleus."
