In this manuscript, the authors presented a literature review and a population genetics secondary data analysis on the epidemiology and genetics of hyperuricemia and gout across major racial groups. The topic of this article is of great interest to other researchers in the field. The results are believed to be able to inspire more excellent research work in the future. The manuscript was very well written with clear logic, enabling the readers to easily catch the key points. Overall, this article is of high quality and importance. The reviewer would like to recommend the publication of this manuscript after the following revisions are addressed.
- The authors are suggested to provide a figure to summarize the outline of the present work.
- It would be great that the authors could add some background information about the relationship between the current work and personalized medicine in the introduction and give a discussion about the impact of current work on personalized medicine in the discussion section. These modifications would make the topic of the current work fit better with the scope of the Journal of Personalized Medicine.
- The format and layout of the manuscript should follow the style of the journal. For example, the abstract section needs to be reorganized.

Dear sir, thank you to select me to review manuscript: Butler F et al. The Epidemiology and Genetics of Hyperuricemia and Gout Across Major Racial Groups: A literature Review and Population Genetics Secondary Data Analysis. Authors analysed in original article genetic aspects of hyperuricemia and gout and also epidemiology of hyperuricemia and gout in Africans in Southwest U.S. (ASW), Han-Chinese (CHS), Japanese (JPT),
and Mexican (MXL). This study has 2 parts. Authors used NHANES data (2007-16) about about hyperuricemia and gout for epidemiology and data from the 1000 Genomes Project database for genetic analysis. The SNP frequencies was 7/11 in ASW, 9/11 in MXL, 9/11 JPT, and 11/11 in CHS. HU or gout risk allele indices were 5, 6, 9, and 11 in ASW, MXL, CHS, and JPT. CHS and JPT populations had the highest HU or gout risk allele frequencies, followed by MXL and ASW. Asian population has 2.7 fold higher risk of gout compared to Caucasians. Design of the study is good, data are clearly presented, discussion is adequate, references are well selected.
