RESPONSE: The previously published review articles and book chapters were focused on the modifications of xanthan gum (XG) to improve its physicochemical properties and have highlighted its applications as a drug delivery carrier particularly using conventional formulations. However, our review is focused on describing the application of XG in designing disease-specific drug delivery systems. Furthermore, it includes the delivery of genetic materials, peptides, and proteins using XG-based carriers.

  1. English is not scientifically sound. In some cases, sentences formation is correct. Also, avoid the use of third person like "we"

RESPONSE: The revised manuscript incorporates the suggested revisions and improved language.

  1. Author must provide a new section about challenges and their appropriate solutions as well

RESPONSE: Challenges in the use of XG and their probable solutions have been incorporated into the conclusion and future prospective section.  

  1. Increase the resolution of Figure 3 and 4. Also, draw the structure using ChemDraw

RESPONSE: The images (Figures 3 and 4) with improved resolution are now provided. The chemical structures are drawn using ChemDraw.

  1. The rationale of the study is missing. Authors should provide one paragraph about this at end of the introduction

Major:

X. campestris is a plant pathogen that represents an important economic loss for crop production. The authors should add a paragraph in the introduction what are disadvantages of X. campestris.

Use of Xanthan gum for probiotics – which strains of probiotic bacteria? Be more specific.

Use of Xanthan gum for antiviral drugs – which viruses? Be more specific.

Line 36: Explain what is predictable outcomes in the human body.

Section 5.1: Add if XG is antibacterial or is just a deliverer.

Line 466: Check if the drug can have pH 1.2.

 

Minor:

The authors should check and revise the manuscript so that the names of bacterial strains are written correctly – in Italics. Check the entire manuscript.

RESPONSE: Xanthan gum is used as a prebiotic with certain probiotics. The specific probiotic bacteria (Bifidobacterium bifidum) have been mentioned in the revised manuscript.

RESPONSE: The specific viruses have been mentioned. Please refer to lines 311, 342, and Table 2 (Sr. No. 3 & 10), in the revised manuscript.

RESPONSE: Predictable outcomes in the body meant the biocompatibility and complete biodegradability of the nanoparticles made up of natural polymers in comparison to those prepared using synthetic polymers. The explanation for the same has been incorporated in the revised manuscript.

RESPONSE: Xanthan gum is used as a carrier for the delivery of antimicrobial drugs. The sentence is revised to clarify this point.

1. is Figure 3 a reused figure? If yes, the reference source should be indicated, and a picture with better resolution should be replaced.

2. The structure of Fig. 2 should be reorganized to accurately show the conformational change of xanthan gum.

3.In 5.7, furosemide is not an anti-inflammatory drug and should not be included in this scope. It should be corrected.

4.It is suggested to delete the title 5.5. Other classifications in this document are based on the type of disease. 5.5 is "gastrointestinal drug delivery", and it is suggested to list this part in 5.7.

5.The format of the references cited in this paper is not uniform and need to be revised.

RESPONSE: Yes, Figure 3 is a reused figure after receiving the reprint permission from the publisher. The reference number is cited in the figure legend. For a better resolution of the image, we obtained the original copy of the figure from the authors which is now used.

  1. The structure of Fig. 2 should be reorganized to accurately show the conformational change of xanthan gum

RESPONSE: The structure of the xanthan gum is reorganized with appropriate conformation in the revised manuscript.

RESPONSE: Yes, furosemide is a diuretic drug and therefore, it has been removed from the anti-inflammatory section.
