Treatment of infections caused by Pseudomonas aeruginosa presents a challenge due to its ability to adapt and acquire drug resistance rapidly. Therefore, a key challenge is identifying and investigating new compounds with antibacterial and anti-virulence activity. Tannins, a group of plant polyphenolic compounds, can interact with bacterial cells and their virulence factors. The purpose of this study was to assess the antibacterial potential of using 3,6-bis-O-di-O-galloyl-1,2,4-tri-O-galloyl-β-D-glucose (C55H40O34) isolated from Rhus typhina against P. aeruginosa ATCC BAA-1744. The investigation involved viability analyses using the INT assay, fluorescence analyses of the tannins’ interaction with the cell membrane and membrane proteins of P. aeruginosa, and analysis of changes in the Zeta potential. The results obtained allowed us to conclude that C55H40O34 exhibits antimicrobial activity by inducing changes in the biophysical properties of P. aeruginosa’s cell membrane. The thermodynamic parameters indicated that C55H40O34 binds to bacterial membrane proteins through hydrophobic interactions. These interactions with proteins may impact their structure and disrupt their functions, such as disturbing or inhibiting the efflux pumps, which are part of P. aeruginosa’s resistance mechanisms. Therefore, C55H40O34 may be a new, natural agent and could potentially be used against P. aeruginosa. Full article

Sumac, Rhus coriaria L., is employed as a natural preservative in the food sector, due to its rich content of antioxidant compounds, including hydrolysable tannins, phenolic acids, anthocyanins, and flavonoids. In this work, the phytochemical characterization of sumac fruits from five Sicilian accessions was performed to evaluate their potential as a food preservative for nutraceutical exploitation. Spectrophotometric tests and HPLC-MS/MS analyses were conducted to assess and compare the antioxidant power of the water extracts produced with the five sumac accessions. Principal component analysis was also carried out to better visualize the obtained results. Flavonoids and phenolic acids, namely isoquercitrin (20,342.82 mg/kg dry extract) and gallic acid (197,489.19 mg/kg dry extract), were more abundant in fruits from the population of San Biagio Platani, while the one from Giarratana was characterized by a higher content of anthocyanins such as cyanidin-3-glucoside (20,889.81 mg/kg dry extract). These two populations can be recognized as the most suitable settings for the implementation of sumac cultivation and the development of sumac-based products, especially for food and nutraceutical purposes. Tannins are natural plant origin polyphenols that are promising compounds for pharmacological applications due to their strong and different biological activities, including antibacterial activity. Our previous studies demonstrated that sumac tannin, i.e., 3,6-bis-O-di-O-galloyl-1,2,4-tri-O-galloyl-β-D-glucose (isolated from Rhus typhina L.), possesses strong antibacterial activity against different bacterial strains. One of the crucial factors of the pharmacological activity of tannins is their ability to interact with biomembranes, which may result in the penetration of these compounds into cells or the realization of their activity on the surface. The aim of the current work was to study the interactions of sumac tannin with liposomes as a simple model of the cellular membrane, which is widely used in studies focused on the explanation of the physicochemical nature of molecule–membrane interactions. Additionally, these lipid nanovesicles are very often investigated as nanocarriers for different types of biologically active molecules, such as antibiotics. In the frame of our study, using differential scanning calorimetry, zeta-potential, and fluorescence analysis, we have shown that 3,6-bis-O-di-O-galloyl-1,2,4-tri-O-galloyl-β-D-glucose interacts strongly with liposomes and can be encapsulated inside them. A formulated sumac–liposome hybrid nanocomplex demonstrated much stronger antibacterial activity in comparison with pure tannin. Overall, by using the high affinity of sumac tannin to liposomes, new, functional nanobiomaterials with strong antibacterial activity against Gram-positive strains, such as S. aureus, S. epidermitis, and B. cereus, can be formulated. In recent years, utilization of Rhus coriaria L. (sumac) is upgrading not only in their culinary use and human nutrition, but also in the pharmaceutical industry, food industry and veterinary practices. This is driven by accumulating evidence that support the ethnobotanical use of this plant; in particular, advanced knowledge of the content of nutritional, medicinal and techno-functional bioactive ingredients. Herein, we discuss polyphenolic compounds as the main bioactive ingredients in Rhus coriaria L., which contribute mainly to the significance and utility of this spice. Most of the antioxidant potential and therapeutic roles of sumac are increasingly attributed to its constituent tannins, flavonoids, and phenolic acids. Hydroxyphenyl pyranoanthocyanins and other anthocynins are responsible for the highly desired red pigments accounting for the strong pigmentation capacity and colorant ability of sumac. Certain polyphenols and the essential oil components are responsible for the peculiar flavor and antimicrobial activity of sumac. Tannin-rich sumac extracts and isolates are known to enhance the food quality and the oxidative stability of animal products such as meat and milk. In conclusion, polyphenol-rich sumac extracts and its bioactive ingredients could be exploited towards developing novel food products which do not only address the current consumers’ interests regarding organoleptic and nutritional value of food, but also meet the growing need for ‘clean label’ as well as value addition with respect to antioxidant capacity, disease prevention, and health promotion in humans. Rhus coriaria L. (Anacardiaceae), commonly known as sumac, has been used since ancient times for many different applications, and nowadays is used mostly as a spice obtained from its in the Mediterranean and the Middle ground fruits and employed for flavoring and garnishing food, predominantly Eastern regions. Traditionally, sumac has been also used in popular medicine for the treatment of many ailments including hemorrhoids, wound healing, diarrhea, ulcers, and eye inflammation. Sumac drupes are indeed rich in various classes of phytochemicals including organic acids, flavonoids, tannins, and others, which are responsible of their powerful antioxidant capacity, from which treatment of many common diseases such as cardiovascular disease, diabetes, and cancer could benefit. In this work we evaluated the influence of fruit ripeness, conservation, and processing. To this aim, a phytochemical characterization of six different samples of Rhus coriaria L. was carried out. Specifically, headspace solid-phase micro extraction gas chromatography coupled to mass spectrometry and comprehensive two-dimensional liquid chromatography coupled to photodiode array and mass spectrometry detection, were employed. A total of 263 volatile compounds, including terpene hydrocarbons, acids, and aldehydes, as well as 83 polyphenolic compounds, mainly gallic acid derivatives, were positively identified. All samples showed a significant antioxidant activity by means of oxygen radical absorbance capacity, in line with their polyphenolic content and composition. Such findings set a solid ground to support the utilization of this plant as an attractive target for novel nutraceutical approaches and for drug discovery. Full article

Rhus coriaria L. (Anacardiaceae), commonly known as sumac, is a commonly used spice, condiment, and flavoring agent, especially in the Mediterranean region. Owing to its bountiful beneficial values, sumac has been used in traditional medicine for the management and treatment of many ailments including hemorrhoids, wound healing, diarrhea, ulcer, and eye inflammation. This plant is rich in various classes of phytochemicals including flavonoids, tannins, polyphenolic compounds, organic acids, and many others. By virtue of its bioactive, Rhus coriaria possesses powerful antioxidant capacities that have ameliorative and therapeutic benefits for many common diseases including cardiovascular disease, diabetes, and cancer. This review describes the phytochemical properties of R. coriaria and then focuses on the potent antioxidant capacities of sumac. We then dissect the cellular and molecular mechanisms of sumac’s action in modulating many pathophysiological instigators. We show how accumulating evidence supports the antibacterial, antinociceptive, antidiabetic, cardioprotective, neuroprotective, and anticancer effects of this plant, especially that toxicity studies show that sumac is very safe to consume by humans and has little toxicity. Taken together, the findings we summarize here support the utilization of this plant as an attractive target for drug discovery. Full article

The extraction of condensed tannins from Aleppo pine bark and sumac roots (Brown Rhus tripartitum) was examined in near industrial conditions, using a water medium in the presence of 2% NaHCO3 and 0.5% NaHSO3 at two different temperatures (70 °C and at 100 °C). The tannins extracts were recovered in high yields (~25% of Aleppo pine and ~30% for sumac) with high phenolic contents (>75%). The tannins were characterized by 13C-NMR and MALDI TOF and showed characteristics of procyanidin/prodelphinidin units. The tannins extracted at 100 °C were composed of smaller flavonoid oligomers (DP < 8) compared to those extracted at a lower temperature (DP > 10). Adhesive resin formulations were prepared using Aleppo or sumac tannins and four different cross linkers (hexamine, glutaraldehyde, furfural, and glyoxal). The resins were studied by TMA in bending and tannins-based formaldehyde-free wood particleboards were produced. The panels displayed internal bond strengths > 0.35 MPa with the four hardeners and all of them passed relevant international standard specifications for interior grade panels. The best results were observed with the tannins extracted at 70 °C with furfural as hardener (IB = 0.81 MPa for Aleppo pine and IB = 0.76 MPa for sumac). Full article

We have demonstrated that polyphenol-rich sorghum bran diets alter fecal microbiota; however, little is known regarding their effect on colon inflammation. Our aim was to characterize the effect of sorghum bran diets on intestinal homeostasis during dextran sodium sulfate (DSS)-induced colitis. Male Sprague-Dawley rats (N = 20/diet) were provided diets containing 6% fiber from cellulose, or Black (3-deoxyanthocyanins), Sumac (condensed tannins) or Hi Tannin Black (both) sorghum bran. Colitis was induced (N = 10/diet) with three separate 48-h exposures to 3% DSS, and feces were collected. On Day 82, animals were euthanized and the colon resected. Only discrete mucosal lesions, with no diarrhea or bloody stools, were observed in DSS rats. Only bran diets upregulated proliferation and Tff3, Tgfβ and short chain fatty acids (SCFA) transporter expression after a DSS challenge. DSS did not significantly affect fecal SCFA concentrations. Bran diets alone upregulated repair mechanisms and SCFA transporter expression, which suggests these polyphenol-rich sorghum brans may suppress some consequences of colitis. Full article
